    Directional swimming in the box jellyfish Tripedalia cystophora (cubozoa, cnidaria) is controlled by the shape of the velarium, which is a thin muscular sheet that forms the opening of the bell. It was unclear how different patterns of visual stimulation control directional swimming and that is the focus of this study. Jellyfish were tethered inside a small experimental tank, where the four vertical walls formed light panels. All four panels were lit at the start of an experiment. The shape of the opening in the velarium was recorded in response to switching off different combinations of panels. We found that under the experimental conditions the opening in the velarium assumed three distinct shapes during a swim contraction. The opening was (1) centred or it was off-centred and pocketed out either towards (2) a rhopalium or (3) a pedalium. The shape of the opening in the velarium followed the direction of the stimulus as long as the stimulus contained directional information. When the stimulus contained no directional information, the percentage of centred pulses increased and the shape of the off-centred pulses had a random orientation. Removing one rhopalium did not change the directional response of the animals, however, the number of centred pulses increased. When three rhopalia were removed, the percentage of centred pulses increased even further and the animals lost their ability to respond to directional information

    Box jellyfish respond to visual stimuli by changing the dynamics and frequency of bell contractions. In this study, we determined how the contrast and the dimming time of a simple visual stimulus affected bell contraction dynamics in the box jellyfish Tripedalia cystophora. Animals were tethered in an experimental chamber where the vertical walls formed the light stimuli. Two neighbouring walls were darkened and the contraction of the bell was monitored by high-speed video. We found that (1) bell contraction frequency increased with increasing contrast and decreasing dimming time. Furthermore, (2) when increasing the contrast and decreasing the dimming time pulses with an off-centred opening had a better defined direction and (3) the number of centred pulses decreased. Only weak effects were found on the relative diameter of the contracted bell and no correlation was found for the duration of bell contraction. Our observations show that visual stimuli modulate swim speed in T. cystophora by changing the swim pulse frequency. Furthermore, the direction of swimming is better defined when the animal perceives a high-contrast, or fast dimming, stimulus

    Cross-validation of generic risk assessment tools for animal disease incursion based on a case study for African swine fever

    In recent years, several generic risk assessment (RA) tools have been developed that can be applied to assess the incursion risk of multiple infectious animal diseases allowing for a rapid response to a variety of newly emerging or re-emerging diseases. Although these tools were originally developed for different purposes, they can be used to answer similar or even identical risk questions. To explore the opportunities for cross-validation, seven generic RA tools were used to assess the incursion risk of African swine fever (ASF) to the Netherlands and Finland for the 2017 situation and for two hypothetical scenarios in which ASF cases were reported in wild boar and/or domestic pigs in Germany. The generic tools ranged from qualitative risk assessment tools to stochastic spatial risk models but were all parameterized using the same global databases for disease occurrence and trade in live animals and animal products. A comparison of absolute results was not possible, because output parameters represented different endpoints, varied from qualitative probability levels to quantitative numbers, and were expressed in different units. Therefore, relative risks across countries and scenarios were calculated for each tool, for the three pathways most in common (trade in live animals, trade in animal products, and wild boar movements) and compared. For the 2017 situation, all tools evaluated the risk to the Netherlands to be higher than Finland for the live animal trade pathway, the risk to Finland the same or higher as the Netherlands for the wild boar pathway, while the tools were inconclusive on the animal products pathway. All tools agreed that the hypothetical presence of ASF in Germany increased the risk to the Netherlands, but not to Finland. The ultimate aim of generic RA tools is to provide risk-based evidence to support risk managers in making informed decisions to mitigate the incursion risk of infectious animal diseases. The case study illustrated that conclusions on the ASF risk were similar across the generic RA tools, despite differences observed in calculated risks.     Generating transgenic reporter lines for studying nervous system development in the cnidarian nematostella vectensis

    Neurons often display complex morphologies with long and fine processes that can be difficult to visualize, in particular in living animals. Transgenic reporter lines in which fluorescent proteins are expressed in defined populations of neurons are important tools that can overcome these difficulties. By using membrane-attached fluorescent proteins, such reporter transgenes can identify the complete outline of subsets of neurons or they can highlight the subcellular localization of fusion proteins, for example at pre- or postsynaptic sites. The relative stability of fluorescent proteins furthermore allows the tracing of the progeny of cells over time and can therefore provide information about potential roles of the gene whose regulatory elements are controlling the expression of the fluorescent protein. Here we describe the generation of transgenic reporter lines in the sea anemone Nematostella vectensis, a cnidarian model organism for studying the evolution of developmental processes. We also provide an overview of existing transgenic Nematostella lines that have been used to study conserved and derived aspects of nervous system development.acceptedVersio

    Propulsion in cubomedusae : mechanisms and utility

Cubomedusae, which often possess large prolate bells and are thought to swim via jet propulsion, appear to violate the theoretical constraints which determine the medusan morphospace. To examine propulsion by cubomedusae, we quantified size related changes in wake dynamics, bell shape, swimming and turning kinematics of two species of cubomedusae, Chironex fleckeri and Chiropsella bronzie. During growth, these cubomedusae transitioned from using jet propulsion at smaller sizes to a rowing-jetting hybrid mode of propulsion at larger sizes. Simple modifications in the flexibility and kinematics of their velarium appeared to be sufficient to alter their propulsive mode. Turning occurs during both bell contraction and expansion and is achieved by generating asymmetric vortex structures during both stages of the swimming cycle.     Curvature facilitates prey fixation in predatory insect claws

    Insects show a large variety in prey capture strategies, with a correspondingly large diversity in predatory adaptations. We studied a specific type of predatory claws, these can for example be found in praying mantis species. The claw is closeable over its entire length and the prey is fixed between the femur (upper arm) and the tibia (lower arm) of the insect leg. The morphology of these predatory claws is diverse. Some species have straight claws covered with spines, while other species have smooth, curved claws. We have studied the mechanics of this femur-tibia type of predatory insect claws, by making a physical model, eventually trying to explain why in some insect species the claws are curved instead of straight. The main results are (1) when comparing curved claws to straight claws, curvature leads to a strong reduction of forces driving the prey away from the pivoting point, thereby reducing the need for friction generating structures. (2) In the curved claw model a position exists where the resulting force on the prey is exactly zero. This is because the normal forces on the femur and tibia are opposed, and in line. At this position the prey is perfectly clamped and not driven out of the claw. This feature does not exist in straight claws. (3) In the curved claw, the prey cannot be placed at a position further than a certain maximum distance from the pivoting point. Near this maximum position, the resulting force on the prey reaches high values because moment arms are near zero. (4) Between the zero position and the maximum position the resulting force is directed toward the pivoting point, which stabilizes prey fixation
