    Bone quality changes as measured by Raman and FTIR spectroscopy in primiparous cows with humeral fracture from New Zealand.

    The occurrence of spontaneous humeral fractures in primiparous dairy cows from New Zealand prompted the study of bone material from affected cows to further characterize this condition and to outline a likely pathogenesis. Previous studies indicate that these cows developed osteoporosis due to periods of suboptimal bone formation followed by increased bone resorption during the period of lactation complicated by copper deficiency. We hypothesized that there are significant differences in the chemical composition/bone quality in bones from cows with spontaneous humeral fracture compared to cows without humeral fractures. In this study, Raman and Fourier transform infrared spectroscopy band ratios were, for the first time, measured, calculated, and compared in bone samples from 67 primiparous dairy cows that suffered a spontaneous fracture of the humerus and 14 age-matched post-calving cows without humeral fractures. Affected bone showed a significantly reduced mineral/matrix ratio, increased bone remodeling, newer bone tissue with lower mineralization and, lower carbonate substitution, and reduced crystallinity. As such, is likely that these have detrimentally impacted bone quality and strength in affected cows.Published onlin

    The use of natural products in the leather industry: Depilation without damage

    Content: Sheepskin, a by-product of the meat industry, is then often processed to leather, primarily for the clothing industry. Where the tanneries are distant from the abattoirs and freezing works, the raw skins have to be transported long distances to be processed. In warm weather, there is the potential for putrefaction of the skins which then have to be disposed of at a cost. Depilation, the first step of leather processing, is designed to remove the wool from the skin without damaging it. Conventional depilation involves the use of strong alkali and sulfides, that are harmful to both the environment and the personnel carrying out the process due to the potential production of hydrogen sulfide, a toxic, flammable gas. To solve this problem, scientists have been looking to depilate skins using enzymes as these are environmentally friendly. Various enzymes, such as collagenase, keratinase, protease and lipase have been shown to be able to remove hair from skin, but unfortunately usually damage it. Furthermore, at present, they are not cost effective at an industrial level. We have found a simple solution, a derivative of a dairy by-product, that prevents putrefaction, preserving the skin for days at room temperature. In addition, it allows easy removal of the wool from the skin. Scanning electron microscopy showed there was no obvious damage to the surface of the depilated skin and that the wool is cleanly removed from the hair follicle. To assess any less visible damage that may have occurred as a result of soaking the skin, biochemical analyses were carried out to measure the changes to the amino acid composition, collagen crosslinks and proteoglycan concentrations of the depilated skins. Sheepskin depilated with this method was processed to leather and its physical properties such as tear, and tensile strength analysed. Culture dependent methods were used to isolate the microorganisms present in the solution after depilation, showed that only four main species were consistently found in the depilation fluid and on the skins. Metagenomic analysis, confirmed these findings and was used to follow the changes in the microbiome during the course of depilation. This presentation will describe the progress that has been made to understand the science behind these observations and compare the properties of skins depilated using this method with those depilated using the traditional beamhouse process.     Infected pancreatic necrosis: outcomes and clinical predictors of mortality. A post hoc analysis of the MANCTRA-1 international study

    : The identification of high-risk patients in the early stages of infected pancreatic necrosis (IPN) is critical, because it could help the clinicians to adopt more effective management strategies. We conducted a post hoc analysis of the MANCTRA-1 international study to assess the association between clinical risk factors and mortality among adult patients with IPN. Univariable and multivariable logistic regression models were used to identify prognostic factors of mortality. We identified 247 consecutive patients with IPN hospitalised between January 2019 and December 2020. History of uncontrolled arterial hypertension (p = 0.032; 95% CI 1.135-15.882; aOR 4.245), qSOFA (p = 0.005; 95% CI 1.359-5.879; aOR 2.828), renal failure (p = 0.022; 95% CI 1.138-5.442; aOR 2.489), and haemodynamic failure (p = 0.018; 95% CI 1.184-5.978; aOR 2.661), were identified as independent predictors of mortality in IPN patients. Cholangitis (p = 0.003; 95% CI 1.598-9.930; aOR 3.983), abdominal compartment syndrome (p = 0.032; 95% CI 1.090-6.967; aOR 2.735), and gastrointestinal/intra-abdominal bleeding (p = 0.009; 95% CI 1.286-5.712; aOR 2.710) were independently associated with the risk of mortality. Upfront open surgical necrosectomy was strongly associated with the risk of mortality (p < 0.001; 95% CI 1.912-7.442; aOR 3.772), whereas endoscopic drainage of pancreatic necrosis (p = 0.018; 95% CI 0.138-0.834; aOR 0.339) and enteral nutrition (p = 0.003; 95% CI 0.143-0.716; aOR 0.320) were found as protective factors. Organ failure, acute cholangitis, and upfront open surgical necrosectomy were the most significant predictors of mortality. Our study confirmed that, even in a subgroup of particularly ill patients such as those with IPN, upfront open surgery should be avoided as much as possible.     On the Application of Automated Machine Vision for Leather Defect Inspection and Grading: A Survey

    Reliably and effectively detecting and classifying leather surface defects is of great importance to tanneries and industries that use leather as a major raw material such as leather footwear and handbag manufacturers. This paper presents a detailed and methodical review of the leather surface defects, their effects on leather quality grading and automated visual inspection methods for leather defect inspection. A detailed review of inspection methods based on leather defect detection using image analysis methods is presented, which are usually classified as heuristic or basic machine learning based methods. Due to the recent success of deep learning methods in various related fields, various architectures of deep learning are discussed that are tailored to image classification, detection, and segmentation. In general, visual inspection applications, where recent CNN architectures are classified, compared, and a detailed review is subsequently presented on the role of deep learning methods in leather defect detection. Finally, research guidelines are presented to fellow researchers regarding data augmentation, leather quality quantification, and simultaneous defect inspection methods, which need to be investigated in the future to make progress in this crucial area of research

    Novel Assessment of Collagen and Its Crosslink Content in the Humerus from Primiparous Dairy Cows with Spontaneous Humeral Fractures Due to Osteoporosis from New Zealand.

    Numerous cases of spontaneous humeral fracture in primiparous dairy cows from New Zealand have prompted the study of the condition to establish probable causes or risk factors associated with the condition. Previous studies identified inadequate protein-calorie malnutrition as an important contributory factor. Earlier case studies also reported that ~50% of cows have low liver and/or serum copper concentration at the time of humeral fracture. Because copper is so closely associated with the formation of collagen cross-links, the aim of this study was to compare collagen and collagen crosslink content in the humerus from primiparous cows with and without humeral fractures and to determine the role of copper in the occurrence of these fractures. Humeri were collected from cows with and without humeral fractures, ground, and the collagen and collagen cross-link content measured using high-performance liquid chromatography. Collagen content was significantly higher in the humeri of cows without humeral fractures, while total collagen crosslink content was significantly higher in the humerus of cows with humeral fractures. These results indicate other factor/s (e.g., protein-calorie undernutrition) might be more important than the copper status in the occurrence of humeral fractures in dairy cows in New Zealand.Published onlin
