    Analysis of a spontaneous mutant and selected clones of cv. Italia (Vitis vinifera) by AFLP markers

    A spontaneously formed mutant and selected clones (from clonal selection) of the table grape cv. Italia (Vitis vinifera) were evaluated with regard to the the possible use of molecular markers for grapevine clone differentiation. The identified off-type grapevine, which presents a mutated branch and a normal one on the same plant, removes any doubt as to its origin and allows a better evaluation of the suitability of molecular markers for the differentiation of grape clones. AFLPs were used as molecular markers because a large number of loci can be screened in a single assay, which is useful for any study on genotype relationships when a large number of bands (variables) is required. Different primer combinations (49) produced 3880 scorable AFLP bands but none showed any polymorphism among clones. Nevertheless it is suggested to use both AFLP and morphological markers for the differentiation of grapevine clones. The AFLPs would confirm the high level of DNA similarity among the suspected clones while morphological characters would allow to verify, through appropriate field experimental designs, the reliability of the phenotypic differences detected among grape clones.

    Diversité spécifique de la macrofaune benthique dans la côte ouest de la baie de Tunis et la côte de l'île de Djerba (sud-ouest de la Méditerranée)

    In this study, 21 stations were sampled, 11 in the Djerba island coast and 10 in the western coast of Tunis bay. Then, samples were sifted on a square mesh of 1 mm a side, and the animals collected were identified and counted. Lacking an efficient single index suitable for application in all situations, several traditional nonparametric indices of biodiversity were selected, calculated and discussed. Results show that the community composition is totally different in the two studied sites, and, within each site, several species are common, butthey exchange the leader ranks from one station to another. For biodiversity measurement and since results show varying values depending on indices at several stations, an arithmetic mean index was calculated at each station on the basis of the most similar indices. It shows that, overall, the biodiversity is significantly higher in Tunis bay coast than in Djerba coast. These results are more reliable considering the real environmental /anthropogenic constraints imposed on both sites. Indeed, Djerba coast is subject, these last decades, to strong industrial, urban and fishing activities, causing significant regression of benthic habitats, which can led to a reduction of the biodiversity.Dans cette étude, 21 stations ont été échantillonnées, 11 dans la côte de l'île de Djerba et 10 dans la côte ouest de la baie de Tunis. Ensuite, les échantillons ont été tamisés sur une maille carrée de 1 mm de côté, et les animaux capturés ont été identifiés et comptés. Faute d'un indice unique efficace et adapté à toutes les situations, plusieurs indices traditionnels de biodiversité ont été choisis, calculés et discutés. Les résultats montrent que la composition de la communauté est totalement différente dans les deux sites étudiés, et, à chaque site, plusieurs espèces sont communes, mais ils se substituent aux places des leaders d'une station à l’autre. Pour la mesure de la biodiversité et étant donné la grande différence dans les valeurs enregistrées par les indices retenus, une moyenne arithmétique a été calculée à chaque station sur la base des indices les plus cohérents. Elle montre que, globalement, la biodiversité est nettement plus élevée dans la baie de Tunis par rapport à la côte de Djerba. Ces résultats semblent être plus fiables tenant en compte les contraintes réelles environnementales et anthropiques imposées aux deux sites. En effet, la côte de Djerba est l'objet, ces dernières décennies, à de fortes activités urbaines et de pêche industrielle, entraînant une régression significative des habitats benthiques, ce qui peut conduire à la réduction de la biodiversité

    Electro-coagulation coupled Electro-floatation process: Feasible choice in Doxycycline removal from Pharmaceutical effluents

    Article CC-BYInternational audienceElectrochemical treatment involving a coupled coagulation and floatation was performed in the removal of Doxycycline Hyclate (DCH) from aqueous solutions. All the experiments were carried out in an electrochemical reactor of 1.5 L which contained aluminium electrodes as cathode and anode. The removal of doxycycline hyclate (DCH) species by EC/EF was determined as a function of electrolysis time, pH, current intensity, flow rate and DCH concentration. From the observed results, it was corroborated that the DCH removal through the EC/EF process was excellent. The effective contribution from initial pH (7.03) and current intensity (5.39 mA cm-2) was very much remarkable and well apparent from flocs of good buoyancy. The removal of DCH was inversely proportional to spacing between electrodes (SBE) and circulating flow rate in the presence of the supporting NaCl electrolyte of 1 g L-1. It was also highly promoted by the addition of NaCl in comparison to NaNO3 and KCl to the electrolytic system. The compliance of four kinetic models was verified with DCH removal system. The free energy values from DKR model suggested the nature of bonding by chemical forces. Characterization by FTIR, SEM and XRD interpreted the assignments of various functional groups, surface morphology and crystalline incorporated amorphous nature, respectively of electro – generated flocs.     Isolation, Identification, and Characterization of Phosphate-Solubilizing Bacteria from Tunisian Soils

    Soil microorganisms play an important role in maintaining natural ecological balance through active participation in carbon, nitrogen, sulfur, and phosphorous cycles. Phosphate-solubilizing bacteria (PSB) are of high importance in the rhizosphere, enhancing the solubilization of inorganic phosphorus complexes into soluble forms available for plant nutrition. The investigation of this species of bacteria is of major interest in agriculture, as they can be used as biofertilizers for crops. In the present study, 28 isolates of PSB were obtained after the phosphate enrichment of soil samples from five Tunisian regions. Five PSB species were identified by 16S rRNA gene sequencing including Pseudomonas fluorescens, P. putida, and P. taiwanensis, Stenotrophomonas maltophilia, and Pantoea agglomerans. Solid and liquid Pikovskaya’s (PVK) and National Botanical Research Institute’s (NBRIP) media containing insoluble tricalcium phosphate were used for the evaluation of the phosphate solubilization ability of the bacterial isolates by two methods: visual evaluation of the solubilization zone around colonies (halo) and determination of solubilized phosphates in liquid medium by the colorimetric method of the vanado-molybdate yellow. Based on the results of the halo method, the isolate of each species that showed the higher phosphate solubilization index was selected for evaluation of phosphate solubilization by the colorimetric method. In the liquid media, the bacterial isolates showed phosphate solubilization ranging from 535.70 to 618.57 µg mL−1 in the NBRIP medium, and 374.20 to 544.28 µg mL−1 in the PVK medium, with the highest values produced by P. fluorescens. The best phosphate solubilization ability and higher reduction in broth pH, which indicates higher organic acid production, were achieved in NBRIP broth for most of the PSB. Strong correlations were observed between the average capability of PSB to solubilize phosphates and both the pH and total phosphorous content in the soil. The production of the hormone indole acetic acid (IAA), which can promote plant growth, was observed for all five PSB species.     Revision of the systematics of the Polystomoidinae (Platyhelminthes, Monogenea, Polystomatidae) with redefinition of Polystomoides Ward, 1917 and Uteropolystomoides Tinsley, 2017

    Polystomatids are platyhelminth parasites that infect mainly amphibians and freshwater turtles. For more than seven decades, chelonian polystomes were classified into three genera according to the number of hamuli, i.e. absent for Neopolystoma, one pair for Polystomoidella and two pairs for Polystomoides. Following re-examination of morphological characters, seven new genera were erected the past six years, namely Apaloneotrema, Aussietrema, Fornixtrema, Manotrema, Pleurodirotrema, Uropolystomoides and Uteropolystomoides. However, the polyphyly of Neopolystoma and Polystomoides on the one hand, and the nested position of Uteropolystomoides within a clade encompassing all Neopolystoma and Polystomoides spp. on the other, still raised questions about the validity of these genera. We therefore re-examined several types, paratypes and voucher specimens, and investigated the molecular phylogeny of polystomes sampled from the oral cavity of North American turtles to re-evaluate their systematic status. We show that all Polystomoides Ward, 1917, sensu Du Preez et al., 2022, Neopolystoma Price, 1939, sensu Du Preez et al., 2022 and Uteropolystomoides Tinsley, 2017 species, display vaginae that are peripheral and extend well beyond the intestine. We thus reassign all species of the clade to Polystomoides and propose nine new combinations; however, although Uteropolystomoides is nested within this clade, based on its unique morphological features, we propose to keep it as a valid taxon. Polystomoides as redefined herein groups all polystome species infecting either the oral cavity or the urinary bladder of cryptodires, with peripheral vaginae and with or without two pairs of small hamuli. Uteropolystomoides nelsoni (Du Preez & Van Rooyen 2015), originally described from Pseudemys nelsoni Carr is now regarded as Uteropolystomoides multifalx (Stunkard, 1924) n. comb. infecting three distinct Pseudemys species of North America

    Botanical and Genetic Identification Followed by Investigation of Chemical Composition and Biological Activities on the Scabiosa atropurpurea L. Stem from Tunisian Flora

    Scarce information about the phenolic composition of Scabiosa atropurpurea L. is available, and no carotenoid compounds have been reported thus far. In this study the phenolic and carotenoid composition of this plant was both investigated and associated bioactivities were evaluated. Aiming to obtain extracts and volatile fractions of known medicinal plants to valorize them in the pharmaceutical or food industries, two techniques of extraction and five solvents were used to determine the biologically active compounds. Gas chromatography coupled to flame ionization and mass spectrometry and liquid chromatography coupled to photodiode array and atmospheric pressure chemical ionization/electrospray ionization mass spectrometry highlighted the presence of 15 volatiles, 19 phenolic, and 24 natural pigments in Scabiosa atropurpurea L. stem samples; among them, the most abundant were 1,8-cineole, chlorogenic acid, cynaroside, and lutein. Bioactivity was assessed by a set of in vitro tests checking for antioxidant, antibacterial, antifungal, and allelopathic (against Brassica oleracea L. and Lens culinaris Medik) effects. Scabiosa atropurpurea L. stem extracts presented a considerable antioxidant, antibacterial, and allelopathic potential, with less antifungal effectiveness. These results indicate that the volatile fractions and extracts from S. atropurpurea L. stem could be considered as a good source of bioactive agents, with possible applications in food-related, agriculture, and pharmaceutical fields.     A Neutron Elastic Diffuse Scattering Study of PMN

    We have performed elastic diffuse neutron scattering studies on the relaxor Pb(Mg1/3_{1/3}Nb2/3_{2/3})O3_3 (PMN). The measured intensity distribution near a (100) Bragg peak in the (hk0) scattering plane assumes the shape of a butterfly with extended intensity in the (110) and (11ˉ\bar{1}0) directions. The temperature dependence of the diffuse scattering shows that both the size of the polar nanoregions (PNR) and the integrated diffuse intensity increase with cooling even for temperatures below the Curie temperature TC∼213T_C \sim 213 K.Comment: Submitted to PR
