    Source to tap investigation of natural organic matter in non-disinfected drinking water distribution systems

    Despite being reduced by treatment, natural organic matter (NOM) is ubiquitous in drinking water distribution systems (DWDSs) from sources to consumers' taps where it can potentially have negative impacts on drinking water quality. While a few studies have investigated its behaviour in disinfected and NOM-rich DWDSs, its dynamics in non-disinfected systems, characterized by low NOM content, have not yet been explored. In this study, we monitored the NOM variations occurring between groundwater sources and consumers' taps of a non-disinfected DWDS, including three drinking water treatment plants, using both fluorescence and absorbance, selected due to their increasing adoption by water utilities. PARAFAC analysis of fluorescence data, combined with absorbance indices, highlighted how NOM characteristics in groundwater vary due to the combination of multiple factors (e.g., well depth, pumping rate), especially in the case of shallower aquifers. The treatment processes display different effects on NOM when monitored by fluorescence and absorbance, due to the differences among fluorophores and between fluorescent and chromophoric molecules. Variations of the NOM characteristics between the treatment plant outlets and sampling locations within the network were detected only in few locations, suggesting the importance of the processes occurring in specific sections of the network and the last meter before consumption. These findings highlight the overall stability of water quality within non- disinfected NOM-poor DWDSs, but they stress the importance of (i) properly selecting the analytical method to be used for monitoring and (ii) localized water quality variations mainly related to pipe materials, suggesting several implications for DWDS monitoring and management

    Growth of Wild Gilthead Seabream (Sparus aurata L.) Juveniles for Organic Aquaculture

    The majority of organic marine fish farms currently begin the production cycle with non-organic juveniles from conventional hatcheries, permitted by the European Regulation on organic fish origin (EC 710/2009) until the end of 2016. Wild juvenile gilthead seabream (Sparus aurata) from coastal lagoons and hatcheries were experimentally reared under organic conditions, in order (1) to investigate differences in fillet lipid content and fatty acids composition, and (2) to propose a possible future source of juveniles destined for organic aquaculture. Wild juveniles were readily distinguishable by their fatty acid signature, showing significantly higher ratio levels of n-3 polyunsaturated fatty acids and n-3/n-6. Fillet lipid composition of organically fed wild S. aurata juveniles was preferable to that from domesticated juveniles. These results seem promising for organic aquaculture, where fish feed is more environmentally sustainable but is of lower nutritional qualit

    Skin pigmentation in gilthead seabream (Sparus aurata l.) fed conventional and novel protein sources in diets deprived of fish meal

    The pattern of yellowish pigmentation of the skin was assessed in gilthead seabream (Sparus aurata) fed for 12 weeks iso-proteic (45%) and iso-lipidic (20%) diets deprived of fish meal and containing either a blend of vegetable protein-rich ingredients or where graded levels of the vegetable protein blend were replaced by insect (Hermetia illucens\u201410%, 20% or 40%) pupae meal, poultry by-product meal (20%, 30% or 40%), red swamp crayfish meal (10%) and marine microalgae (Tisochrysis lutea and Tetraselmis suecica\u201410%) dried biomass. Digital images of fish fed diets differing in protein sources were analyzed by means of an automatic and non-invasive image analysis tool, in order to determine the number of yellow pixels and their dispersion on the frontal and lateral sides of the fish. The relationship between the total carotenoid concentration in the diet and the number of yellow pixels was investigated. Test diets differently affected gilthead seabream skin pigmentation both in the forefront and the operculum, due to their carotenoid content. The highest yellow pixels\u2019 number was observed with the diet containing microalgae. Fish fed poultry by-product meal were characterized by the lowest yellow pixels\u2019 number, diets containing insect meal had an intermediate coloring capacity. The vegetable control, the microalgae mix diet and the crayfish diet had significantly higher values of yellow pixels at both inspected skin sites

    Regenerative potential of DPSCs and revascularization. direct, paracrine or autocrine effect?

    A new source of mesenchymal stem cells has recently been discovered, the so-called dental pulp derived stem cells (DPSCs) which therefore could represent potentially tools for regenerative medicine. DPSC originate from the neural crest and are physiologically involved in dentin homeostasis; moreover, they contribute to bone remodeling and differentiation into several tissues including cartilage, bone, adipose and nervous tissues. DPSCs have also been shown to influence the angiogenesis process, for example through the release of secretory factors or by differentiating into vascular and/or perivascular cells. Angiogenesis, that has a pivotal role in tissue regeneration and repair, is defined as the formation of new vessels from preexisting vessels and is mediated by mutual and reciprocal interactions between endothelial cells and perivascular cells. It is also known that co-cultures of perivascular and endothelial cells (ECs) can form a vascular network in vitro and also in vivo. Since DPSCs seem to have characteristics similar to pericytes, understanding the possible mechanism of interaction between DPSCs and ECs during neo-angiogenesis is dramatically important for the development of advanced clinical application in the field of regeneration.     Treatment of Comorbid Obesity and Major Depressive Disorder: A Prospective Pilot Study for their Combined Treatment

    Background. Obese individuals who suffer from major depressive disorder are routinely screened out of weight loss trials. Treatments targeting obesity and depression concurrently have not been tested. Purpose. To test the short-term efficacy of a treatment that combined behavioral weight management and cognitive behavioral therapy (CBT) for obese adults with depression. Methods. Twelve obese females diagnosed with major depressive disorder received weekly group behavioral weight management, combined with CBT for depression, for 16 weeks. Weight, symptoms of depression, and cardiovascular disease (CVD) risk factors were measured at baseline and week 16. Results. Participants lost 11.4% of initial weight and achieved significant improvements in symptoms of depression and CVD risk factors. Conclusions. Obese individuals suffering from major depressive disorder can lose weight and achieve improvements in symptoms of depression and CVD risk factors with 16 weeks of combined treatment. A larger randomized controlled trial is needed to establish the efficacy of this treatment

    Hypoxia-Mimetic CoCl2 Agent Enhances Pro-Angiogenic Activities in Ovine Amniotic Epithelial Cells-Derived Conditioned Medium

    Amniotic epithelial stem cells (AECs) are largely studied for their pro-regenerative properties. However, it remains undetermined if low oxygen (O2) levels that AECs experience in vivo can be of value in maintaining their biological properties after isolation. To this aim, the present study has been designed to evaluate the effects of a hypoxia-mimetic agent, cobalt chloride (CoCl2), on AECs’ stemness and angiogenic activities. First, a CoCl2 dose-effect was performed to select the concentration able to induce hypoxia, through HIF-1α stabilization, without promoting any cytotoxicity effect assessed through the analysis of cell vitality, proliferation, and apoptotic-related events. Then, the identified CoCl2 dose was evaluated on the expression and angiogenic properties of AECs’ stemness markers (OCT-4, NANOG, SOX-2) by analysing VEGF expression, angiogenic chemokines’ profiles, and AEC-derived conditioned media activity through an in vitro angiogenic xeno-assay. Results demonstrated that AECs are sensitive to the cytotoxicity effects of CoCl2. The unique concentration leading to HIF-1α stabilization and nuclear translocation was 10 µM, preserving cell viability and proliferation up to 48 h. CoCl2 exposure did not modulate stemness markers in AECs while progressively decreasing VEGF expression. On the contrary, CoCl2 treatment promoted a significant short-term release of angiogenic chemokines in culture media (CM). The enrichment in bio-active factors was confirmed by the ability of CoCl2-derived CM to induce HUVEC growth and the cells’ organization in tubule-like structures. These findings demonstrate that an ap-propriate dose of CoCl2 can be adopted as a hypoxia-mimetic agent in AECs. The short-term, chemical-induced hypoxic condition can be targeted to enhance AECs’ pro-angiogenic properties by providing a novel approach for stem cell-free therapy protocols

    Prioritising research areas for antibiotic stewardship programmes in hospitals: a behavioural perspective consensus paper

    SCOPE: Antibiotic stewardship programmes (ASPs) are necessary in hospitals to improve the judicious use of antibiotics. While ASPs require complex change of key behaviours on individual, team, organisation and policy levels, evidence from the behavioural sciences is underutilised in antibiotic stewardship studies across the world, including high-income countries (HICs). A consensus procedure was performed to propose research priority areas for optimising effective implementation of ASPs in hospital settings, using a behavioural perspective. METHODS: A workgroup for behavioural approaches to ASPs was convened in response to the fourth call for leading expert network proposals by the Joint Programming Initiative on Antimicrobial Resistance (JPIAMR). Eighteen clinical and academic specialists in antibiotic stewardship, implementation science and behaviour change from four high-income countries with publicly-funded health care systems (that is Canada, Germany, Norway and the UK), met face-to-face to agree on broad research priority areas using a structured consensus method. QUESTION ADDRESSED AND RECOMMENDATIONS: The consensus process on the 10 identified research priority areas resulted in recommendations that need urgent scientific interest and funding to optimise effective implementation of antibiotic stewardship programmes for hospital inpatients in HICs with publicly-funded health care systems. We suggest and detail, behavioural science evidence-guided research efforts in the following areas: 1) Comprehensively identifying barriers and facilitators to implementing antibiotic stewardship programmes and clinical recommendations intended to optimise antibiotic prescribing; 2) Identifying actors ('who') and actions ('what needs to be done') of antibiotic stewardship programmes and clinical teams; 3) Synthesising available evidence to support future research and planning for antibiotic stewardship programmes; 4) Specifying the activities in current antibiotic stewardship programmes with the purpose of defining a 'control group' for comparison with new initiatives; 5) Defining a balanced set of outcomes and measures to evaluate the effects of interventions focused on reducing unnecessary exposure to antibiotics; 6) Conducting robust evaluations of antibiotic stewardship programmes with built-in process evaluations and fidelity assessments; 7) Defining and designing antibiotic stewardship programmes; 8) Establishing the evidence base for impact of antibiotic stewardship programmes on resistance; 9) Investigating the role and impact of government and policy contexts on antibiotic stewardship programmes; and 10) Understanding what matters to patients in antibiotic stewardship programmes in hospitals. Assessment, revisions and updates of our priority-setting exercise should be considered, at intervals of 2 years.     Food Components and Dietary Habits: Keys for a Healthy Gut Microbiota Composition.

    The gut microbiota is a changing ecosystem, containing trillions of bacteria, continuously shaped by many factors, such as dietary habits, seasonality, lifestyle, stress, antibiotics use, or diseases. A healthy host-microorganisms balance must be respected in order to optimally maintain the intestinal barrier and immune system functions and, consequently, prevent disease development. In the past several decades, the adoption of modern dietary habits has become a growing health concern, as it is strongly associated with obesity and related metabolic diseases, promoting inflammation and both structural and behavioral changes in gut microbiota. In this context, novel dietary strategies are emerging to prevent diseases and maintain health. However, the consequences of these different diets on gut microbiota modulation are still largely unknown, and could potentially lead to alterations of gut microbiota, intestinal barrier, and the immune system. The present review aimed to focus on the impact of single food components (macronutrients and micronutrients), salt, food additives, and different dietary habits (i.e., vegan and vegetarian, gluten-free, ketogenic, high sugar, low FODMAP, Western-type, and Mediterranean diets) on gut microbiota composition in order to define the optimal diet for a healthy modulation of gut microbiota

    FORESHELL project is funded by the FLAG Costa Blu trough 2014-20 EMFF program of the Abruzzo Region. It is carrying out a pilot initiative to develop sanitary/weather-environmental predictive technological tools to enhance the efficiency and sustainability of the mussel farm in the Giulianova Maritime District. The hydrographic basins of the rivers close to the farm, Vibrata and Salinello, are constantly monitored trough the hydrological model (CHyM), to forecast occurrences of discharge peaks at the river mouth. Moreover, a specific sampling programme is established before and after severe weather events to determine the concentration of E. coli in freshwater at the river mouths and in mussels/seawater at the farming area. Until September 2021, there were 4 meteorological events that did not cause a peak discharge at the river mouth. Results did not show a significant increase of E. coli in the mussels. At the same time, the environmental parameters such as sea water temperature, salinity, chlorophyll a, sea currents and wave motion are acquired by the satellites and in situ probe. The web application for data visualisation is under construction, as well as the early warning signalling to the farmer. Furthermore, the growth of mussels is constantly monitored with biometric controls. In conclusion, the implementation of all phases of the FORESHELL project are progressing according to the timeline in order to develop innovative tools useful for the management of mussel farming area
