    Decaying grid turbulence in a rotating stratified fluid

    Rotating grid turbulence experiments have been carried out in a stably stratified fluid for relatively large Reynolds numbers (mesh Reynolds numbers up to 18000). Under the combined effects of rotation and stratification the flow degenerates into quasihorizontal motions. This regime is investigated using a scanning imaging velocimetry technique which provides time-resolved velocity fields in a volume. The most obvious effect of rotation is the inhibition of the kinetic energy decay, in agreement with the quasi-geostrophic model which predicts the absence of a direct energy cascade, as found in two-dimensional turbulence. In the regime of small Froude and Rossby numbers, the dynamics is found to be non-dissipative and associated with a symmetric and highly intermittent vertical vorticity field, that displays k(h)(-3) energy spectra. For higher Rossby numbers, fundamental differences with the quasi-geostrophic model are found. A significant decay of kinetic energy, which does not depend on the stratification, is observed. Moreover, in this regime, although both cyclones and anticyclones are initially produced, the intense vortices are only cyclones. For late times the flow consists of an assembly of coherent interacting Structures.     This paper highlights examples of the various ways students in beginner Russian, intermediate Spanish and advanced French engage with the local and regional community in educational and noneducational settings. It explains the history of these engagements and provides instructor impressions as well as impressions from student participants and community partners concerning learning outcomes and the benefits to both students and the community. In all three examples, the majority of students were taking the course to fulfill a core requirement.     International audienceA ferritic oxide dispersion strengthened steel is under study for fuel cladding applications in future nuclear systems. Tensile tests and creep tests are carried out at various temperatures to determine its mechanical properties along the extrusion direction. For these two types of loading, the material exhibits a high mechanical resistance. Its ductility appears to be strongly influenced by the strain rate and the temperature. Deformation mechanisms linked to diffusion phenomena are suspected and intergranular damage is observed on fractured specimens

    Extraction of coherent structures in a rotating turbulent flow experiment

    The discrete wavelet packet transform (DWPT) and discrete wavelet transform (DWT) are used to extract and study the dynamics of coherent structures in a turbulent rotating fluid. Three-dimensional (3D) turbulence is generated by strong pumping through tubes at the bottom of a rotating tank (48.4 cm high, 39.4 cm diameter). This flow evolves toward two-dimensional (2D) turbulence with increasing height in the tank. Particle Image Velocimetry (PIV) measurements on the quasi-2D flow reveal many long-lived coherent vortices with a wide range of sizes. The vorticity fields exhibit vortex birth, merger, scattering, and destruction. We separate the flow into a low-entropy ``coherent'' and a high-entropy ``incoherent'' component by thresholding the coefficients of the DWPT and DWT of the vorticity fields. Similar thresholdings using the Fourier transform and JPEG compression together with the Okubo-Weiss criterion are also tested for comparison. We find that the DWPT and DWT yield similar results and are much more efficient at representing the total flow than a Fourier-based method. Only about 3% of the large-amplitude coefficients of the DWPT and DWT are necessary to represent the coherent component and preserve the vorticity probability density function, transport properties, and spatial and temporal correlations. The remaining small amplitude coefficients represent the incoherent component, which has near Gaussian vorticity PDF, contains no coherent structures, rapidly loses correlation in time, and does not contribute significantly to the transport properties of the flow.     Early activation of wheat polyamine biosynthesis during Fusarium head blight implicates putrescine as an inducer of trichothecene mycotoxin production

    <p>Abstract</p> <p>Background</p> <p>The fungal pathogen <it>Fusarium graminearum </it>causes Fusarium Head Blight (FHB) disease on wheat which can lead to trichothecene mycotoxin (<it>e.g</it>. deoxynivalenol, DON) contamination of grain, harmful to mammalian health. DON is produced at low levels under standard culture conditions when compared to plant infection but specific polyamines (<it>e.g</it>. putrescine and agmatine) and amino acids (<it>e.g</it>. arginine and ornithine) are potent inducers of DON by <it>F. graminearum </it>in axenic culture. Currently, host factors that promote mycotoxin synthesis during FHB are unknown, but plant derived polyamines could contribute to DON induction in infected heads. However, the temporal and spatial accumulation of polyamines and amino acids in relation to that of DON has not been studied.</p> <p>Results</p> <p>Following inoculation of susceptible wheat heads by <it>F. graminearum</it>, DON accumulation was detected at two days after inoculation. The accumulation of putrescine was detected as early as one day following inoculation while arginine and cadaverine were also produced at three and four days post-inoculation. Transcripts of ornithine decarboxylase (ODC) and arginine decarboxylase (ADC), two key biosynthetic enzymes for putrescine biosynthesis, were also strongly induced in heads at two days after inoculation. These results indicated that elicitation of the polyamine biosynthetic pathway is an early response to FHB. Transcripts for genes encoding enzymes acting upstream in the polyamine biosynthetic pathway as well as those of ODC and ADC, and putrescine levels were also induced in the rachis, a flower organ supporting DON production and an important route for pathogen colonisation during FHB. A survey of 24 wheat genotypes with varying responses to FHB showed putrescine induction is a general response to inoculation and no correlation was observed between the accumulation of putrescine and infection or DON accumulation.</p> <p>Conclusions</p> <p>The activation of the polyamine biosynthetic pathway and putrescine in infected heads prior to detectable DON accumulation is consistent with a model where the pathogen exploits the generic host stress response of polyamine synthesis as a cue for production of trichothecene mycotoxins during FHB disease. However, it is likely that this mechanism is complicated by other factors contributing to resistance and susceptibility in diverse wheat genetic backgrounds.</p

    Validation of a new automatic smoking machine to study the effects of cigarette smoke in newborn lambs

    The aim of this study was to describe the characteristics and validate the use of a new, custom-built automatic smoking machine (ASM), primarily designed to study the effects of an environmental tobacco smoke surrogate (ETS surrogate) exposure in animals of various sizes, including large animals. The equipment includes a programmable ASM coupled to a vented whole body chamber, where animals can be exposed to both mainstream and sidestream smoke. The user-friendly interface allows for full programming of puff volume (1-60 mL), time interval between two puffs (1-60 s) and between two cigarettes (1-60 min). Eight newborn lambs were exposed to either 10 (4 lambs, C10 group) or 20 (4 lambs, C20 group) cigarettes, 8 h per day for 15 days. Four additional control, lambs were exposed to air (C0 group). Weight gain was identical in all three groups of lambs. Urinary cotinine/creatinine ratio increased with the number of cigarettes smoked (C0: 11 ± 7 ng/mg; C10: 961 ± 539 ng/mg; C20: 1821 ± 312 ng/mg), with levels in the C10 and C20 groups in keeping with values published in infants exposed to ETS. Overall, results show that our new ASM is especially well suited for ETS surrogate exposure in non-restrained, non-anaesthetized large animals such as sheep

    Effects of postnatal smoke exposure on laryngeal chemoreflexes in newborn lambs

    Laryngeal chemoreflexes (LCR), which are elicited by the contact of liquids such as gastric refluxate with laryngeal mucosa, may trigger some cases of sudden infant death syndrome. Indeed, while LCR in mature mammals consist in lower airway protective responses, previous animal data have shown that LCR in immature newborns can include laryngospasm, apnea, bradycardia and desaturation. The present study was aimed at testing the hypothesis that postnatal exposure to cigarette smoke is responsible for enhancing cardiorespiratory inhibition observed with LCR. Eight lambs were exposed to cigarette smoke (20 cig/day) during 16 days and compared to seven control lambs. Urinary cotinine/creatinine ratio was measured at a level relevant to previously published levels in infants. On D15-D16, 0.5 ml of HCl (pH 2), milk, distilled water or saline was injected onto the larynx via a chronic supra-glottal catheter during sleep. Results showed that exposure to cigarette smoke significantly enhanced apneas and bradycardias during LCR while significantly decreasing swallowing and arousal. These results were observed independently of the state of alertness and of the experimental solution tested. In conclusion, 15 day-postnatal exposure to cigarette smoke increases cardiorespiratory inhibition and decreases lower airway protective mechanisms in non-sedated, full-term lambs
