The contribution of rural community businesses to integrated rural development: “Local services for local people”
Policy responses to the problems facing rural areas across Europe have involved the replacement of “productivist” measures that subsidise agriculture to strategies promoting “integrated rural development”, emphasising the interconnections between various facets of the rural economy. Thus farm modernisation and product processing and marketing are linked with the promotion of a more diversified economic base centred on tourism and recreation and the maintenance of services for local residents. An essential element of this model is its reliance on collaborative actions involving a range of community or civil society actors. This paper examines the extent to which the operation of community-owned businesses in rural parts of the Yorkshire and Humber region in the UK corresponds to these ideals of integrated rural development. Evidence is presented on their geographical footprint with respect to both direct economic impacts and linkages with social and institutional networks. This allows an assessment to be made of the contribution that such enterprises make to rural economic development as a whole. The conclusion is that they do have the potential to assist integrated rural development, but only as a small part of a much wider series of economic, social and environmental actions.integrated rural development, rural community businesses, economic impacts, geographical footprint, volunteering
A Cholecystokinin B Receptor-Specific Aptamer Does Not Activate Receptor Signaling
Targeted nanoparticles which deliver effective doses of chemotherapeutic drugs directly to pancreatic tumors could improve treatment efficacy without the toxicities associated with systemic drug administration. One protein on tumor cells that can be targeted by nanoparticles is a G-protein coupled cell surface receptor, the cholecystokinin B receptor (CCKBR). Previously, we had shown that attaching the CCKBR ligand gastrin to the surface of nanoparticles can enhance their up-take by tumors. The drawback of using gastrin is that it can also activate the receptor, causing tumor cell growth. This study shows that a DNA aptamer that binds to the CCKBR and enhances nanoparticle up-take by tumors does not activate this receptor.
PANC-1 cells, a cultured human pancreatic cancer cell line, were treated for 24 h with CCKBR aptamer 1153. Cell lysates were run on Bis-Tris gels, transferred to membranes, blocked in 5% BSA and incubated overnight with primary antibodies, including antibodies directly against phosphorylated-Akt (Ser473), total Akt, and beta-actin, a protein loading control. Although the CCKBR aptamer 1153 is internalized by pancreatic cancer cells in a receptor-mediated fashion, it does not stimulate cell proliferation. Because of this, we anticipate that it will not activate CCKBR signaling. If aptamer 1153 does not activate downstream receptor signaling, our future work will test whether the aptamer could be used to specifically direct drug-containing nanoparticles to tumors, making chemotherapy treatments for pancreatic cancer patients more effective with fewer off-target effects and toxicity
Illegal dumping and crime prevention: A case study of Ash Road, Liverpool Council
Illegal waste disposal is an increasingly significant and costly problem. This paper considers a specific hot-spot for illegal dumping in Sydney, Australia from criminological perspectives. We contribute to the developing criminological literature that considers environmental harms as a crime. This draws upon the symbolic aspect of criminal law, contributing to the notion of environmental harms as wrongs worthy of sanction, and facilitates analysis through the prism of criminological literature. Communication difficulties reported by patients diagnosed with idiopathic pulmonary fibrosis and their carers : a European focus group study
This paper explores commonalities in the experience and unmet needs of idiopathic pulmonary fibrosis (IPF) patients and carers in Europe throughout the care pathway, focusing specifically on the unmet communication needs of patients and carers. Four patient organisations/groups in Europe held focus groups (Italy (seven patients and four carers); Belgium (six patients); Ireland (23 patients and 10 carers); and England, UK (five patients and three carers)). A focus group schedule was provided and translated into the language of each focus group by the European Lung Foundation (ELF). Content analysis was conducted by the ELF and verified by the authors of the paper. Three main themes emerged: professional-patient, professional-professional and patient-patient communication. Within these themes, eight priority areas were highlighted by two or more of the focus groups. In addition, 17 suggested action points were identified. Patients and carers in Europe have unmet communication needs, which could be met by specialist physicians and specialist centres providing more effective information and signposting to support services, including support groups and patient organisations
Human iPSC-Derived Cerebellar Neurons from a Patient with Ataxia-Telangiectasia Reveal Disrupted Gene Regulatory Networks
Ataxia-telangiectasia (A-T) is a rare genetic disorder caused by loss of function of the ataxia-telangiectasia-mutated kinase and is characterized by a predisposition to cancer, pulmonary disease, immune deficiency and progressive degeneration of the cerebellum. As animal models do not faithfully recapitulate the neurological aspects, it remains unclear whether cerebellar degeneration is a neurodevelopmental or neurodegenerative phenotype. To address the necessity for a human model, we first assessed a previously published protocol for the ability to generate cerebellar neuronal cells, finding it gave rise to a population of precursors highly enriched for markers of the early hindbrain such as EN1 and GBX2, and later more mature cerebellar markers including PTF1α, MATH1, HOXB4, ZIC3, PAX6, and TUJ1. RNA sequencing was used to classify differentiated cerebellar neurons generated from integration-free A-T and control induced pluripotent stem cells. Comparison of RNA sequencing data with datasets from the Allen Brain Atlas reveals in vitro-derived cerebellar neurons are transcriptionally similar to discrete regions of the human cerebellum, and most closely resemble the cerebellum at 22 weeks post-conception. We show that patient-derived cerebellar neurons exhibit disrupted gene regulatory networks associated with synaptic vesicle dynamics and oxidative stress, offering the first molecular insights into early cerebellar pathogenesis of ataxia-telangiectasia
Gadolinium-Enhanced Magnetic Susceptibility Contrast Is Reduced in the Corpus Callosum of a Mouse Model of Tauopathy
Alterations to white matter have been implicated in a number of neurodegenerative diseases. In this work we investigate the sensitivity of MRI magnetic susceptibility mapping to these changes using a mouse model of Tauopathy. A non-rigid registration of contrast enhanced, high resolution ex vivo mouse brain images is used to transform susceptibility maps into a common space and a voxel-wise group comparison is performed. Significant differences were observed that may indicate disruption to the tissue of the corpus callosum
