Treatment of hemorrhagic shock with hypertonic saline solution modulates the inflammatory response to live bacteria in lungs
Shock and resuscitation render patients more susceptible to acute lung injury due to an exacerbated immune response to subsequent inflammatory stimuli. To study the role of innate immunity in this situation, we investigated acute lung injury in an experimental model of ischemia-reperfusion (I-R) followed by an early challenge with live bacteria. Conscious rats (N = 8 in each group) were submitted to controlled hemorrhage and resuscitated with isotonic saline (SS, 0.9% NaCl) or hypertonic saline (HS, 7.5% NaCl) solution, followed by intratracheal or intraperitoneal inoculation of Escherichia coli. After infection, toll-like receptor (TLR) 2 and 4 mRNA expression was monitored by RT-PCR in infected tissues. Plasma levels of tumor necrosis factor α and interleukins 6 and 10 were determined by ELISA. All animals showed similar hemodynamic variables, with mean arterial pressure decreasing to nearly 40 mmHg after bleeding. HS or SS used as resuscitation fluid yielded equal hemodynamic results. Intratracheal E. coli inoculation per se induced a marked neutrophil infiltration in septa and inside the alveoli, while intraperitoneal inoculation-associated neutrophils and edema were restricted to the interseptal space. Previous I-R enhanced lung neutrophil infiltration upon bacterial challenge when SS was used as reperfusion fluid, whereas neutrophil influx was unchanged in HS-treated animals. No difference in TLR expression or cytokine secretion was detected between groups receiving HS or SS. Reversals in nature and the nature of reversals
The asymmetric shape of reversals of the Earth's magnetic field indicates a
possible connection with relaxation oscillations as they were early discussed
by van der Pol. A simple mean-field dynamo model with a spherically symmetric
coefficient is analysed with view on this similarity, and a comparison
of the time series and the phase space trajectories with those of paleomagnetic
measurements is carried out. For highly supercritical dynamos a very good
agreement with the data is achieved. Deviations of numerical reversal sequences
from Poisson statistics are analysed and compared with paleomagnetic data. The
role of the inner core is discussed in a spectral theoretical context and
arguments and numerical evidence is compiled that the growth of the inner core
might be important for the long term changes of the reversal rate and the

Endogenous cholinergic inputs and local circuit mechanisms govern the phasic mesolimbic dopamine response to nicotine
Nicotine exerts its reinforcing action by stimulating nicotinic acetylcholine receptors (nAChRs) and boosting dopamine (DA) output from the ventral tegmental area (VTA). Recent data have led to a debate about the principal pathway of nicotine action: direct stimulation of the DAergic cells through nAChR activation, or disinhibition mediated through desensitization of nAChRs on GABAergic interneurons. We use a computational model of the VTA circuitry and nAChR function to shed light on this issue. Our model illustrates that the α4β2-containing nAChRs either on DA or GABA cells can mediate the acute effects of nicotine. We account for in vitro as well as in vivo data, and predict the conditions necessary for either direct stimulation or disinhibition to be at the origin of DA activity increases. We propose key experiments to disentangle the contribution of both mechanisms. We show that the rate of endogenous acetylcholine input crucially determines the evoked DA response for both mechanisms. Together our results delineate the mechanisms by which the VTA mediates the acute rewarding properties of nicotine and suggest an acetylcholine dependence hypothesis for nicotine reinforcement.Peer reviewe
Distribution of the daily Sunspot Number variation for the last 14 solar cycles
The difference between consecutive daily Sunspot Numbers was analysed. Its
distribution was approximated on a large time scale with an exponential law. In
order to verify this approximation a Maximum Entropy distribution was generated
by a modified version of the Simulated Annealing algorithm. The exponential
approximation holds for the generated distribution too. The exponential law is
characteristic for time scales covering whole cycles and it is mostly a
characteristic of the Sunspot Number fluctuations and not of its average

Co-founding ant queens prevent disease by performing prophylactic undertaking behaviour
Abstract Background Social insects form densely crowded societies in environments with high pathogen loads, but have evolved collective defences that mitigate the impact of disease. However, colony-founding queens lack this protection and suffer high rates of mortality. The impact of pathogens may be exacerbated in species where queens found colonies together, as healthy individuals may contract pathogens from infectious co-founders. Therefore, we tested whether ant queens avoid founding colonies with pathogen-exposed conspecifics and how they might limit disease transmission from infectious individuals. Results Using Lasius niger queens and a naturally infecting fungal pathogen Metarhizium brunneum, we observed that queens were equally likely to found colonies with another pathogen-exposed or sham-treated queen. However, when one queen died, the surviving individual performed biting, burial and removal of the corpse. These undertaking behaviours were performed prophylactically, i.e. targeted equally towards non-infected and infected corpses, as well as carried out before infected corpses became infectious. Biting and burial reduced the risk of the queens contracting and dying from disease from an infectious corpse of a dead co-foundress. Conclusions We show that co-founding ant queens express undertaking behaviours that, in mature colonies, are performed exclusively by workers. Such infection avoidance behaviours act before the queens can contract the disease and will therefore improve the overall chance of colony founding success in ant queens
Prolonged oral cannabinoid administration prevents neuroinflammation, lowers β-amyloid levels and improves cognitive performance in Tg APP 2576 mice
Background: Alzheimer’s disease (AD) brain shows an ongoing inflammatory condition and non-steroidal antiinflammatories
diminish the risk of suffering the neurologic disease. Cannabinoids are neuroprotective and antiinflammatory
agents with therapeutic potential.
Methods: We have studied the effects of prolonged oral administration of transgenic amyloid precursor protein
(APP) mice with two pharmacologically different cannabinoids (WIN 55,212-2 and JWH-133, 0.2 mg/kg/day in the
drinking water during 4 months) on inflammatory and cognitive parameters, and on 18F-fluoro-deoxyglucose
(18FDG) uptake by positron emission tomography (PET).
Results: Novel object recognition was significantly reduced in 11 month old Tg APP mice and 4 month
administration of JWH was able to normalize this cognitive deficit, although WIN was ineffective. Wild type mice
cognitive performance was unaltered by cannabinoid administration. Tg APP mice showed decreased 18FDG
uptake in hippocampus and cortical regions, which was counteracted by oral JWH treatment. Hippocampal GFAP
immunoreactivity and cortical protein expression was unaffected by genotype or treatment. In contrast, the density
of Iba1 positive microglia was increased in Tg APP mice, and normalized following JWH chronic treatment. Both
cannabinoids were effective at reducing the enhancement of COX-2 protein levels and TNF-a mRNA expression
found in the AD model. Increased cortical b-amyloid (Ab) levels were significantly reduced in the mouse model by
both cannabinoids. Noteworthy both cannabinoids enhanced Ab transport across choroid plexus cells in vitro.

Proportion and characteristics of secondary progressive multiple sclerosis in five European registries using objective classifiers
Background: To assign a course of secondary progressive multiple sclerosis (MS) (SPMS) may be difficult and the proportion of persons with SPMS varies between reports. An objective method for disease course classification may give a better estimation of the relative proportions of relapsing-remitting MS (RRMS) and SPMS and may identify situations where SPMS is under reported.Materials and methods: Data were obtained for 61,900 MS patients from MS registries in the Czech Republic, Denmark, Germany, Sweden, and the United Kingdom (UK), including date of birth, sex, SP conversion year, visits with an Expanded Disability Status Scale (EDSS) score, MS onset and diagnosis date, relapses, and disease-modifying treatment (DMT) use. We included RRMS or SPMS patients with at least one visit between January 2017 and December 2019 if ≥ 18 years of age. We applied three objective methods: A set of SPMS clinical trial inclusion criteria ("EXPAND criteria") modified for a real-world evidence setting, a modified version of the MSBase algorithm, and a decision tree-based algorithm recently published.Results: The clinically assigned proportion of SPMS varied from 8.7% (Czechia) to 34.3% (UK). Objective classifiers estimated the proportion of SPMS from 15.1% (Germany by the EXPAND criteria) to 58.0% (UK by the decision tree method). Due to different requirements of number of EDSS scores, classifiers varied in the proportion they were able to classify; from 18% (UK by the MSBase algorithm) to 100% (the decision tree algorithm for all registries). Objectively classified SPMS patients were older, converted to SPMS later, had higher EDSS at index date and higher EDSS at conversion. More objectively classified SPMS were on DMTs compared to the clinically assigned.Conclusion: SPMS appears to be systematically underdiagnosed in MS registries. Reclassified patients were more commonly on DMTs.</p
