    Into the Wild: Parallel Transcriptomics of the Tsetse-Wigglesworthia Mutualism within Kenyan Populations

    Tsetse flies (Diptera: Glossinidae) have medical significance as the obligate vectors of African trypanosomes. In addition, tsetse harbor a simple gut microbiota. A predominant gut microbiota member, the Gammaproteobacterium Wigglesworthia spp., has coevolved with tsetse for a significant portion of Glossina radiation proving critical to tsetse fitness. Although multiple roles have been described for Wigglesworthia within colony flies, little research has been dedicated towards functional characterization within wild tsetse. Here, dual RNA-Seq was performed to characterize the tsetse-Wigglesworthia symbiosis within flies captured in Nguruman, Kenya. A significant correlation in Gene Ontology (GO) distribution between tsetse and Wigglesworthia was observed, with homogeneous enrichment in metabolic and transport categories, likely supporting a hallmark of the symbiosis-bidirectional metabolic exchange. Within field flies, highly transcribed Wigglesworthia loci included those involved in B vitamin synthesis and in substrate translocation, including amino acid transporters and multidrug efflux pumps, providing a molecular means for interaction. The universal expression of several Wigglesworthiaand G. pallidipes orthologs, putatively involved in nutrient provisioning and resource allocation, was confirmed in sister tsetse species. These transcriptional profiles varied through host age and mating status likely addressing varying symbiont demands and also confirming their global importance within Glossina. This study, not only supports symbiont nutrient provisioning roles, but also serves as a foundation for insight into novel roles and molecular mechanisms associated with vector–microbiota interactions. The role of symbiont B vitamin provisioning towards impacting host epigenetics is discussed. Knowledge of vector–microbiota interactions may lead to the discovery of novel targets in pest control

    Impact of sulfur starvation in autotrophic and heterotrophic cultures of the Extremophilic Microalga Galdieria Phlegrea (Cyanidiophyceae)

    In plants and algae, sulfate assimilation and cysteine synthesis are regulated by sulfur (S) accessibility from the environment. This study reports the effects of S deprivation in autotrophic and heterotrophic cultures of Galdieria phlegrea (Cyanidiophyceae), a unicellular red alga isolated in the Solfatara crater located in Campi Flegrei (Naples, Italy), where H2S is the prevalent form of gaseous S in the fumarolic fluids and S is widespread in the soils near the fumaroles. This is the first report on the effects of S deprivation on a sulfurous microalga that is also able to grow heterotrophically in the dark. The removal of S from the culture medium of illuminated cells caused a decrease in the soluble protein content and a significant decrease in the intracellular levels of glutathione. Cells from heterotrophic cultures of G. phlegrea exhibited high levels of internal proteins and high glutathione content, which did not diminish during S starvation, but rather glutathione significantly increased. The activity of O-acetylserine(thiol)lyase (OASTL), the enzyme synthesizing cysteine, was enhanced under S deprivation in a time-dependent manner in autotrophic but not in heterotrophic cells. Analysis of the transcript abundance of the OASTL gene supports the OASTL activity increase in autotrophic cultures under S deprivation

    Extending the temporal context of ethnobotanical databases: the case study of the Campania region (southern Italy)

    <p>Abstract</p> <p>Background</p> <p>Ethnobotanical studies generally describe the traditional knowledge of a territory according to a "hic et nunc" principle. The need of approaching this field also embedding historical data has been frequently acknowledged. With their long history of civilization some regions of the Mediterranean basin seem to be particularly suited for an historical approach to be adopted. Campania, a region of southern Italy, has been selected for a database implementation containing present and past information on plant uses.</p> <p>Methods</p> <p>A relational database has been built on the basis of information gathered from different historical sources, including diaries, travel accounts, and treatises on medicinal plants, written by explorers, botanists, physicians, who travelled in Campania during the last three centuries. Moreover, ethnobotanical uses described in historical herbal collections and in Ancient and Medieval texts from the Mediterranean Region have been included in the database.</p> <p>Results</p> <p>1672 different uses, ranging from medicinal, to alimentary, ceremonial, veterinary, have been recorded for 474 species listed in the data base. Information is not uniformly spread over the Campanian territory; Sannio being the most studied geographical area and Cilento the least one. About 50 plants have been continuously used in the last three centuries in the cure of the same affections. A comparison with the uses reported for the same species in Ancient treatises shows that the origin of present ethnomedicine from old learned medical doctrines needs a case-by-case confirmation.</p> <p>Conclusion</p> <p>The database is flexible enough to represent a useful tool for researchers who need to store and compare present and previous ethnobotanical uses from Mediterranean Countries.</p

    Original research: Trauma exposure and posttraumatic stress disorder among employees of New York City companies affected by the september 11, 2001 attacks on the World Trade Center

    OBJECTIVE: Several studies have provided prevalence estimates of posttraumatic stress disorder (PTSD) related to the September 11, 2001 (9/11) attacks in broadly affected populations, although without sufficiently addressing qualifying exposures required for assessing PTSD and estimating its prevalence. A premise that people throughout the New York City area were exposed to the attacks on the World Trade Center (WTC) towers and are thus at risk for developing PTSD has important implications for both prevalence estimates and service provision. This premise has not, however, been tested with respect to DSM-IV-TR criteria for PTSD. This study examined associations between geographic distance from the 9/11 attacks on the WTC and reported 9/11 trauma exposures, and the role of specific trauma exposures in the development of PTSD. METHODS: Approximately 3 years after the attacks, 379 surviving employees (102 with direct exposures, including 65 in the towers, and 277 with varied exposures) recruited from 8 affected organizations were interviewed using the Diagnostic Interview Schedule/Disaster Supplement and reassessed at 6 years. The estimated closest geographic distance from the WTC towers during the attacks and specific disaster exposures were compared with the development of 9/11–related PTSD as defined by the Diagnostic and Statistical Manual, Fourth Edition, Text Revision. RESULTS: The direct exposure zone was largely concentrated within a radius of 0.1 mi and completely contained within 0.75 mi of the towers. PTSD symptom criteria at any time after the disaster were met by 35% of people directly exposed to danger, 20% of those exposed only through witnessed experiences, and 35% of those exposed only through a close associate’s direct exposure. Outside these exposure groups, few possible sources of exposure were evident among the few who were symptomatic, most of whom had preexisting psychiatric illness. CONCLUSIONS: Exposures deserve careful consideration among widely affected populations after large terrorist attacks when conducting clinical assessments, estimating the magnitude of population PTSD burdens, and projecting needs for specific mental health interventions

    A new hexapeptide from the leader peptide of rMnSOD enters cells through the oestrogen receptor to deliver therapeutic molecules

    A 24-amino acid leader peptide of a new human recombinant manganese superoxide dismutase can enter cells and carry molecules. Here, we demonstrated that six of the 24 amino acids penetrate cells through a particular gate represented by a specific amino acid sequence of the oestrogen receptor (ER). We analysed the internalization of the synthetic hexapeptide and the cytotoxic activity of the hexapeptide conjugated to cisplatin on a cell line panel. In most cell lines, the hexapeptide delivered an amount of cisplatin that was 2 to 8 times greater than that released by cisplatin when the drug was used alone. This increased delivery increases the therapeutic index of cisplatin and reduces side effects caused by a high dosage or long-term treatment times. We may consider this hexapeptide a new molecular carrier to deliver molecules with therapeutic activity into ER+ cells for diagnostic purposes and clinical or immune therapy

    Remittance micro-worlds and migrant infrastructure: circulations, disruptions, and the movement of money

    Remittances are increasingly central to development discourses in Africa. The development sector seeks to leverage transnational migration and rapid innovations in financial technologies (fintech), to make remittance systems cheaper for end-users and less risky for states and companies. Critical scholarship, however, questions the techno-fix tendency, calling for grounded research on the intersections between remittances, technologies, and everyday life in African cities and beyond. Building on this work, we deploy the concepts of “micro-worlds” and “migrant infrastructure” to make sense of the complex networks of actors, practices, regulations, and materialities that shape remittance worlds. To ground the work, we narrate two vignettes of remittance service providers who operate in Cape Town, South Africa, serving the Congolese diaspora community. We showcase the important role of logistics companies in the “informal” provision of remittance services and the rise of fintech companies operating in the remittance space. These vignettes give substance to the messy and relational dynamics of remittance micro-worlds. This relationality allows us to see how remittances are circulations, not unidirectional flows; how they are not split between formal and informal, but in fact intersect in blurry ways; how digital technologies are central to the story of migrant infrastructures; and how migrants themselves are compositional of these networks. In doing so, we tell a more relational story about how remittance systems are constituted and configured

    Not just for romance: applications of speed dating in social work education

    In this article we address how a contemporary adaptation of the \u27speed dating\u27 model was used for educational purposes with two cohorts of social work students. We outline the dimensions of \u27speed dating\u27 as a contemporary social phenomenon, then address how this model relates specifically to groupwork process, and can be used to facilitate social work student learning. The curriculum for two classroom group activities using the \u27speed dating\u27 model are outlined, the first to develop university level study skills, the second for debriefing field placement learning experiences. Finally we examine why the \u27speed dating\u27 metaphor was successful in provoking a playful yet constructively creative space for students to engage in groupwork process.<br /

    Mental Health of Long Term Survivors of Childhood and Young Adult Cancer

    Background/Objectives Leukemic cells produce higher amounts of ROS than healthy cells and express low levels of antioxidant enzymes such as catalase (CAT) and superoxide dismutase (SOD). Recently, it was demonstrated that rMnSOD, a recombinant new isoform of MnSOD, recently isolated from human liposarcoma cell line, is able to trigger apoptosis of human T-ALL cells (99%) without toxic effects on healthy cells. The present study was aimed to verify if rMnSOD is able to induce apoptosis and to stop in G0/G1 stage of the cell cycle in pediatric patients with B-cell acute lymphoblastic leukaemia (ALL) and if it displays synergic action with daunorubicin. Design and Methods Cells were collected from three patients diagnosed for B-cell ALL at the Pediatric Oncology of University “Luigi Vanvitelli”. Cell viability, apoptosis and cell cycle of lymphoblasts and SUP-B15 cell line (ATCC) were analyzed through “ Muse Kit” after rMnSOD treatment. ROS analysis was detected with NAC. Apoptotic fragmentations of SUP-B15 were demonstrated by confocal imaging. Results We observed that low concentrations of rMnSOD are able to trigger apoptosis of SUP-B15 cell line and B-ALL cells from pediatric patients. In detail, we observed an apoptosis rate from 30% to 46% in B-ALL pediatric patient cells and of 45% in SUP-B15 cell line. Instead, cell cycle analysis showed a cell fraction decrease in G0/G1 phase. Moreover, the synergic activity of rMnSOD and Daunorubicin induces apoptosis in 92% of cells, by comparing the treatment with the single substances, rMnSOD (64,7%) and Daunorubicin (23,9%). Confocal microscopy analysis revealed internalization of rMnSOD in SUP-B15 cells and evident apoptotic alterations, such as nuclear fragmentations and apoptotic-bodies. In conclusion, rMnSOD exerts toxic activity only against cancer cells, by enhancing drug effect, thus allowing to utilize lower concentration of standard chemotherapy to increase the apoptosis level
