    Chip Off the Old Block : Generation, Development, and Ancestral Concepts of Heredity

    Heredity is such a fundamental concept that it is hard to imagine a world where the connection between parents and offspring is not understood. Three hundred years ago thinking of the phenomenon of heredity bore on a cluster of distinct philosophical questions inherited from antiquity concerning the nature and origin of substances or beings that lacked biological meaning. We are reminded of this philosophical heritage by the fact that in the 18th century the study of reproduction, embryology and development was referred to as "the science of generation". It is now clear that reproduction, the biological process by which parents produce offspring, is a fundamental feature of all life on Earth. Heredity, the transmission of traits from parents to offspring via sexual or asexual reproduction, allows differences between individuals to accumulate and evolve through natural selection. Genetics is the study of heredity, and in particular, variation of fundamental units responsible for heredity. Ideas underlying this theory evolved in considerably different and unrelated ways across a number of knowledge domains, including philosophy, medicine, natural history, and breeding. The fusion of these different domains into a single comprehensive theory in 19th century biology was a historically and culturally interdependent process, thus examining genetic prehistory should unravel these entanglements. The major goal of our review is tracing the various threads of thought that gradually converged into our contemporary understanding of heredity.Peer reviewe

    Internal transcribed spacer (ITS) evolution in populations of the hyperparasitic European mistletoe pathogen fungus, Sphaeropsis visci (Botryosphaeriaceae) : The utility of ITS2 secondary structures

    We investigated patterns of nucleotide polymorphism in the internal transcribed spacer (ITS) region for Sphaeropsis visci, a hyperparasitic fungus that causes the leaf spot disease of the hemiparasite European mistletoe (Viscum album). Samples of S. visci were obtained from Hungary covering all major infected forest areas. For obtaining PCR products we used a fast and efficient direct PCR approach based on a high fidelity DNA polymerase. A total of 140 ITS sequences were subjected to an array of complementary sequence analyses, which included analyses of secondary structure stability, nucleotide polymorphism patterns, GC content, and presence of conserved motifs. Analysed sequences exhibited features of functional rRNAs. Overall, polymorphism was observed within less conserved motifs, such as loops and bulges, or, alternatively, as non-canonical G–U pairs within conserved regions of double stranded helices. The secondary structure of ITS2 provides new opportunities for obtaining further valuable information, which could be used in phylogenetic analyses, or at population level as demonstrated in our study. This is due to additional information provided by secondary structures and their models. The combined score matrix was used with the methods implemented in the programme 4SALE.     The little-known history of cleanliness and the forgotten pioneers of handwashing

    Handwashing is a simple method for preventing the spread of pathogens. It is now common practice, but this was not always the case. Advocating for it often costed a doctor his career in the 1840s. Hospitals in the early 1800s had little idea of the significance of hygiene; thus, they were often mocked as disease-producing incubators or as "houses of death." Many of the ill and dying were kept on wards with no ventilation or access to clean water; hospitals were found to offer only the most basic care. The mortality rate for patients admitted to hospital was three to five times greater than that for individuals cared for at home. Doctors did not routinely wash their hands until the mid-1800s, and they would proceed straight from dissecting a corpse to delivering a baby, providing the basis for the spread of puerperal fever. Despite advances in modern medicine, healthcare providers still face the issue of infection outbreaks caused by patient care. While the body of scientific data supporting hand hygiene as the key strategy to prevent the spread of pathogens is substantial, we highlight that achieving this crucial, long-awaited breakthrough was a hard task through history.Peer reviewe

    MolMarker: A Simple Tool for DNA Fingerprinting Studies and Polymorphic Information Content Calculation

    Molecular markers and mapping are used to analyze an organism’s genes. They allow the selection of target genetic areas based on marker genotype (and not trait phenotype), facilitate the study of genetic variability and diversity, create linkage maps, and follow individuals or lines carrying certain genes. They may be used to select parental genotypes, remove linkage drag in back-crossing, and choose difficult-to-measure characteristics. Due to a lack of genetic variety in crops, the gene pools of wild crop relatives for future agricultural production have been examined. The invention of RFLP (Restriction Fragment Length Polymorphism) for linkage mapping allowed for the creation of other traditional approaches such as RAPD (Random Amplified Polymorphic DNA) and AFLP (Amplified Fragment Length Polymorphism). Accordingly, the need to describe the polymorphic information content (PIC) of the ideal marker has been raised. Marker selection reliability depends on the marker’s relationship to the genomic area of interest. Although informativeness must be estimated for genetic study design, there are no readily available tools. Earlier, PICcalc was developed to calculate heterozygosity (H) and PIC to simplify molecular investigations. These two values were corrected for dominant and co-dominant markers (binary and allelic data) to determine polymorphism quality.     Magnesium transporter Gene Family: Genome-Wide Identification and Characterization in Theobroma cacao, Corchorus capsularis, and Gossypium hirsutum of Family Malvaceae

    Magnesium (Mg) is an element involved in various key cellular processes in plants. Mg transporter (MGT) genes play an important role in magnesium distribution and ionic balance maintenance. Here, MGT family members were identified and characterized in three species of the plant family Malvaceae, Theobroma cacao, Corchorus capsularis, and Gossypium hirsutum, to improve our understanding of their structure, regulatory systems, functions, and possible interactions. We identified 18, 41, and 16 putative non-redundant MGT genes from the genome of T. cacao, G. hirsutum, and C. capsularis, respectively, which clustered into three groups the maximum likelihood tree. Several segmental/tandem duplication events were determined between MGT genes. MGTs appear to have evolved slowly under a purifying selection. Analysis of gene promoter regions showed that MGTs have a high potential to respond to biotic/abiotic stresses and hormones. The expression patterns of MGT genes revealed a possible role in response to P. megakarya fungi in T. cacao, whereas MGT genes showed differential expression in various tissues and response to several abiotic stresses, including cold, salt, drought, and heat stress in G. hirsutum. The co-expression network of MGTs indicated that genes involved in auxin-responsive lipid metabolism, cell wall organization, and photoprotection can interact with MGTs

    A revision of the Old World Black Nightshades (Morelloid clade of Solanum L., Solanaceae)

    The Morelloid clade, also known as the black nightshades or ?Maurella? (Morella), is one of the 10 major clades within Solanum L. The pantropical clade consists of 75 currently recognised non-spiny herbaceous and suffrutescent species with simple or branched hairs with or without glandular tips, with a centre of distribution in the tropical Andes. A secondary centre of diversity is found in Africa, where a set of mainly polyploid taxa occur. A yet smaller set of species is found in Australasia and Europe, including Solanum nigrum L., the type of the genus Solanum. Due to the large number of published synonyms, combined with complex morphological variation, our understanding of species limits and diversity in the Morelloid clade has remained poor despite detailed morphological studies carried out in conjunction with breeding experiments. Here we provide the first taxonomic overview since the 19th century of the entire group in the Old World, including Africa, Asia, Australia, Europe and islands of the Pacific. Complete synonymy, morphological descriptions, distribution maps and common names and uses are provided for all 19 species occurring outside the Americas (i.e. Africa, Asia, Australia, Europe and islands of the Pacific). We treat 12 species native to the Old World, as well as 7 taxa that are putatively introduced and/or invasive in the region. The current knowledge of the origin of the polyploid species is summarised. A key to all of the species occurring in the Old World is provided, together with line drawings and colour figures to aid identification both in herbaria and in the field.
