Chromato-panning: an efficient new mode of identifying suitable ligands from phage display libraries
<p>Abstract</p> <p>Background</p> <p>Phage Display technology is a well established technique for high throughput screening of affinity ligands. Here we describe a new compact chromato-panning procedure for selection of suitable binders from a phage peptide display library.</p> <p>Results</p> <p>Both phages and <it>E. coli </it>cells pass non-hindered through the interconnected pores of macroporous gel, so called <it>cryogel</it>. After coupling a ligand to a monolithic cryogel column, the phage library was applied on the column and non-bound phages were washed out. The selection of strong phage-binders was achieved already after the first panning cycle due to the efficient separation of phage-binders from phage-non-binders in chromatographic mode rather than in batch mode as in traditional biopanning procedures. <it>E. coli </it>cells were applied on the column for infection with the specifically bound phages.</p> <p>Conclusion</p> <p>Chromato-panning allows combining several steps of the panning procedure resulting in 4–8 fold decrease of total time needed for phage selection.</p
Assessment of Therapeutic Efficacy of 5% Sodium Hydrocarbonate Solution in Case of Nodular Dermatitis of Cattle
This article provides information on studies and evaluations of therapeutic efficacy of 5 % sodium bicarbonate solution in case of nodular dermatitis of cattle. Due to the market expansion of veterinary drugs, the periodization of unusual and new medicines as well as control methods of pathogens of farm livestock, having therapeutic and prophylactic effect, is under permanent control of specialists in the field of veterinary medicine. Recently the importance of finding modern means and methods of dealing with animal diseases aimed at the restoration of the body homeostasis has increased significantly in order to ensure the physiologically normal functioning of many enzymes, hormones and the whole body. There is a need for their rational use based on the study of changes in clinical and biochemical parameters occurring in the body under their influence and other factors contributing to the development of normal physiological status of the body (homeostasis). Based on the above, the present studies were based not only on etiological and pathogenetic, but also sanogenetic ideas about diseases with the aim of choosing the means that form the body’s normal physiological status (body homeostasis restoration), taking into account physiological characteristics of animals, and thereby providing the best medical and prophylactic efficacy [4]. Periodization of new drugs and methods to combat animal diseases that contribute to the restoration of body homeostasis is very important. This importantance has become even more significant since the increased sanitary and hygienic requirements for the use of veterinary drugs in animal husbandry, primarily for the treatment of dairy animals, which will serve as a guarantee for obtaining livestock products safe from the point of view of veterinary and sanitary norms and excellent sanitary quality
Blood Chemistry Value Assesment in Hypodermatosis of Cattle in the Chechen Republic
This article is devoted to the problem of cattle hypodermatosis caused by Hypoderma bovis De Geer and Hypoderma lineatum De Villers hypoderma flies. The results of studies of blood chemistry value of cattle diseased with hypodermatosis in the Chechen Republic are revealed in the article. The examination of ten blood samples taken from cattle diseased with animals hypodermatosis showed an increase in the activity of creatinine parameters above normal by 60 % or more, a significant decrease in carotene indicators (within 50 % or more), a relative decrease in crude protein, a decrease in glucose levels, an increase in alkaline phosphatase and alanine-aminotransferase, reduction of the studied reserve alkalinity index vol. % CO2 from 2 to 5 units (12 %) is lower, in six samples an increase in the activity of immunoglobulin IgA. All this is connected with the complex of pathomorphological changes expressed in the mechanical impact – traumatization, intoxication and hypersusceptibility of the organism of the infested animal with the development of inflammatory and dystrophic manifestations associated with injuries of muscle cells and tissues, intoxication, and an acid-alkali balance of the body shift in the acidic side (acidosis), pathological liver abnormality or diseases, with the defeat of organs and body systems, edema in different parts of the body, defect of immunoreaction and homeostasis in the body. Information obtained from biochemical screen provide an opportunity to conduct a deeper assessment of homeostasis and the body’s ability to self-regulate, promptly and comprehensively diagnosing diseases, study the dynamics of the development of pathological processes and conditions in the body, determine the clinical course and justifiably prescribe a complex of effective household and medical-preventive activities aimed at reducing the economic damage to livestock [8, 10]
Hosts of lung helminth <i>Crenosoma vulpis</i> (Rudolphi, 1819)
The purpose of the research is to study the species composition of Crenosoma spp. dwelling in the body of wild canines (Vulpes vulpes, Canis lupus, and Canis aureu).Materials and methods. The research material was 36 wild canines belonging to three species (19 Vulpes vulpes, six Canis lupus, and 11 Canis aureu). The animals were studied using the method of partial helminthological dissections per K. I. Skryabin. The dissections determined the infection intensity (sp./animal) and prevalence (%).Results and discussion. We studied pulmonary helminth infections of wild animals in the Republic of Ingushetia. The study results showed a high infection rate in wild carnivores. The average prevalence in the animals was 47.2%. The foxes were the most heavily infected (73.6%); they were found to have two Crenosoma spp., Crenosoma vulpis and C. petrowi. The prevalence was 16.6% in wolf, and 18.2% in jackal
Persian Population of Terek Region in Second Half of 19th — Early 20th Centuries
The migration movement of the population of Northern Iran to the Terek region in the era of its post-reform modernization in the second half of the 19th — early 20th centuries is considered. The reasons, nature, time and features of the migration history of the Iranians are considered. The novelty of the study lies in the identification of specific starting points of migration on the territory of Iran, routes of advancement on Russian territory and places of settlement in the Terek region. Particular attention is paid to the study of the social composition and economic adaptation of migrants. On the basis of archival sources, corrections were made to the prevailing ideas about social strata, about the labor nature of migration, and about the ethnic composition of migrants. Significant differences in the situation of migrants in the Transcaucasia and the North Caucasus are revealed. The dependence of these differences on the characteristics of the host society — the Terek region during the period of post-reform modernization of the economy and social development -is determined. The specifics of the arrangement of Iranian migrants in national regions of the region have been determined. It is concluded that ethnic entrepreneurship was the basis of the adaptation process and the life of migrants who made a significant contribution to the development of the region’s economy
Pemphigus Erythematosus of the Scalp Developed Around the Postoperative Scar
We present a clinical case reflecting the difficulties of differential diagnosis of pemphigus erythematosus (PE) in a 40-yearold male presented with erythematous topic lesions, pustules, and crusts on the skin of the scalp and face, which had existed for the previous 20 years of his life. In our case, clinical manifestations of dermatosis were atypical due to the presence of pustules and crusts impregnated with seropurulent exudates, resistant to antibiotic therapy. The patient had areas of atrophy on the scalp that could be a sign of discoid lupus erythematosus. The negative results of tests for LE cells and double-strand DNA antibodies, as well as ineffective treatment with plaquenil, antibiotics and itraconazole, helped to verify the diagnosis of PE
