    Gobernanza global: visiones teóricas y pluralidad de actores

    A presente pesquisa tem o objetivo de fazer uma análise teórica sobre o movimento da governança global, através do estudo dos principais atores, articulações e mecanismos alternativos. A metodologia utilizada no trabalho foi revisão bibliográfica de abordagem qualitativa. Teve-se como critério a utilização de literatura clássica, artigos, estudos contemporâneos sobre a temática. A delimitação temporal da pesquisa se deu a partir da década de 1990, devido á ascensão dos processos de globalização após a guerra-fria, da nova ordem multipolar e do recrudescimento do neoliberalismo. Ocorreram assim mudanças nos paradigmas internacionais das relações políticas e econômicas entre atores heterogêneos e plurais –Estados nacionais, empresas multinacionais, sociedade civil global, organizações internacionais, blocos regionais-. Isso ampliou os cenários e espaços de disputa pelo poder. Essas dinâmicas comportam uma pluralidade de atores públicos e privados que disputam espaço e relacionam-se ao mesmo tempo. É nesse cenário de disputa, controle e condução pelo poder que as dinâmicas da governança global são estabelecidas, sem necessariamente um governo mundial constituído e revestido de uma única autoridade. O caráter contraditório e por vezes não explícito da governança global tem apresentado desafios teóricos para qualquer tentativa de entender seu funcionamento e sua evolução.The objective of this research is to carry out a theoretical analysis of the global governance movement through the study of the main actors, articulations and alternative mechanisms. The methodology used in the study was a bibliographical review of a qualitative approach. The criteria included the use of classical literature, articles and contemporary studies on the subject. The temporal delimitation of the research began in the 1990s, due to the rise of globalization processes after the Cold War, with the new multipolar order and the resurgence of neo-liberalism. So many changes occurred in international paradigms of political and economic relations between heterogeneous actors and plurals - nation states, multinational corporations, global civil society, international organizations, regional blocks - that the scenarios and spaces of power struggle expanded. These dynamics involve a plurality of public and private actors competing for space and relating at the same time. It is in this scenario that dispute for and control of the power dynamics of global governance are established, without necessarily a single world government anointed with authority. The contradictory, and sometimes not explicit, character of global governance has presented theoretical challenges for any attempts to understand its operation and its evolution.La presente investigación tiene como objetivo hacer un análisis teórico sobre el movimiento de la gobernanza global a través del estudio de los principales actores, articulaciones y mecanismos alternativos.     Phytoplankton Size Structure in Association with Mesoscale Eddies off Central-Southern Chile: The Satellite Application of a Phytoplankton Size-Class Model

    Understanding the influence of mesoscale and submesoscale features on the structure of phytoplankton is a key aspect in the assessment of their influence on marine biogeochemical cycling and cross-shore exchanges of plankton in Eastern Boundary Current Systems (EBCS). In this study, the spatio-temporal evolution of phytoplankton size classes (PSC) in surface waters associated with mesoscale eddies in the EBCS off central-southern Chile was analyzed. Chlorophyll-a (Chl-a) size-fractionated filtration (SFF) data from in situ samplings in coastal and coastal transition waters were used to tune a three-component (micro-, nano-, and pico-phytoplankton) model, which was then applied to total Chl-a satellite data (ESA OC-CCI product) in order to retrieve the Chl-a concentration of each PSC. A sea surface, height-based eddy-tracking algorithm was used to identify and track one cyclonic (sC) and three anticyclonic (ssAC1, ssAC2, sAC) mesoscale eddies between January 2014 and October 2015. Satellite estimates of PSC and in situ SFF Chl-a data were highly correlated (0.64 < r < 0.87), although uncertainty values for the microplankton fraction were moderate to high (50 to 100% depending on the metric used). The largest changes in size structure took place during the early life of eddies (~2 months), and no major differences in PSC between eddy center and periphery were found. The contribution of the microplankton fraction was ~50% (~30%) in sC and ssAC1 (ssAC2 and sAC) eddies when they were located close to the coast, while nanoplankton was dominant (~60–70%) and picoplankton almost constant (<20%) throughout the lifetime of eddies. These results suggest that the three-component model, which has been mostly applied in oceanic waters, is also applicable to highly productive coastal upwelling systems. Additionally, the PSC changes within mesoscale eddies obtained by this satellite approach are in agreement with results on phytoplankton size distribution in mesoscale and submesoscale features in this region, and are most likely triggered by variations in nutrient concentrations and/or ratios during the eddies’ lifetimes

    A model for scour around bridge piers caused by flood waves

    The local scour process during flood waves is modeled introducing the idea of an effective work by the flow on the sediment bed around the pier. Dimensional considerations show different possible formulations for the dimensionless effective flow work W* that, in each case, is shown to be a generalization of the flow intensity concept, commonly used in existing scour formulas. A novel experimental installation able to reproduce any hydrograph with high precision in the laboratory flume is used to carry out steady and unsteady flow experiments in order to calibrate and validate the mathematical model. Results confirm the uniqueness of the relationship between the dimensionless, effective flow work W* and the relative scour depth Z*, highlighting the high predictive capacity of scour depth caused by any hydrograph. The proposed model provides good performances allowing a straightforward prediction of maximum scour depth and its evolution

    Probiotic bacteria regulate intestinal epithelial permeability in experimental ileitis by a TNF-dependent mechanism

    Background: We previously showed that the probiotic mixture, VSL#3, prevents the onset of ileitis in SAMP/YitFc (SAMP) mice, and this effect was associated with stimulation of epithelial-derived TNF. The aim of this study was to determine the mechanism(s) of VSL#3-mediated protection on epithelial barrier function and to further investigate the "paradoxical" effects of TNF in preventing SAMP ileitis. Methods: Permeability was evaluated in SAMP mice prior to the onset of inflammation and during established disease by measuring transepithelial electrical resistance (TEER) on ex vivo-cultured ilea following exposure to VSL#3 conditioned media (CM), TNF or VSL#3-CM + anti-TNF. Tight junction (TJ) proteins were assessed by qRT-PCR, Western blot, and confocal microscopy, and TNFRI/TNFRII expression measured in freshly isolated intestinal epithelial cells (IEC) from SAMP and control AKR mice. Results: Culture with either VSL#3-CM or TNF resulted in decreased ileal paracellular permeability in pre-inflamed SAMP, but not SAMP with established disease, while addition of anti-TNF abrogated these effects. Modulation of the TJ proteins, claudin-2 and occludin, occurred with a significant decrease in claudin-2 and increase in occludin following stimulation with VSL#3-CM or TNF. TNF protein levels increased in supernatants of SAMP ilea incubated with VSL#3-CM compared to vehicle, while IEC-derived TNFR mRNA expression decreased in young, and was elevated in inflamed, SAMP versus AKR mice. Conclusions: Our data demonstrate that the previously established efficacy of VSL#3 in preventing SAMP ileitis is due to direct innate and homeostatic effects of TNF on the gut epithelium, modulation of the TJ proteins, claudin-2 and occludin, and overall improvement of intestinal permeability. © 2012 Corridoni et al

    Towards High-resolution Imaging from Underwater Vehicles

    Large area mapping at high resolution underwater continues to be constrained by sensor-level environmental constraints and the mismatch between available navigation and sensor accuracy. In this paper, advances are presented that exploit aspects of the sensing modality, and consistency and redundancy within local sensor measurements to build high-resolution optical and acoustic maps that are a consistent representation of the environment. This work is presented in the context of real-world data acquired using autonomous underwater vehicles (AUVs) and remotely operated vehicles (ROVs) working in diverse applications including shallow water coral reef surveys with the Seabed AUV, a forensic survey of the RMS Titanic in the North Atlantic at a depth of 4100 m using the Hercules ROV, and a survey of the TAG hydrothermal vent area in the mid-Atlantic at a depth of 3600 m using the Jason II ROV. Specifically, the focus is on the related problems of structure from motion from underwater optical imagery assuming pose instrumented calibrated cameras. General wide baseline solutions are presented for these problems based on the extension of techniques from the simultaneous localization and mapping (SLAM), photogrammetric and the computer vision communities. It is also examined how such techniques can be extended for the very different sensing modality and scale associated with multi-beam bathymetric mapping.     Congenital heart diseases: post-operative appearance on multi-detector CT—a pictorial essay

    Echocardiography is considered as an initial imaging modality of choice in patients with congenital heart disease (CHD), and magnetic resonance (MR) imaging is preferred for detailed functional information. Multi-detector computed tomography (CT) plays an important role in clinical practice in assessing post-operative morphological and functional information of patients with complex CHD when echocardiography and MR imaging are not contributory. Radiologists should understand and become familiar with the complex morphology and physiology of CHD, as well as with various palliative and corrective surgical procedures performed in these patients, to obtain CT angiograms with diagnostic quality and promptly recognise imaging features of normal post-operative anatomy and complications of these complex surgeries

    Coxiella burnetii Phagocytosis Is Regulated by GTPases of the Rho Family and the RhoA Effectors mDia1 and ROCK

    The GTPases belonging to the Rho family control the actin cytoskeleton rearrangements needed for particle internalization during phagocytosis. ROCK and mDia1 are downstream effectors of RhoA, a GTPase involved in that process. Coxiella burnetii, the etiologic agent of Q fever, is internalized by the host´s cells in an actin-dependent manner. Nevertheless, the molecular mechanism involved in this process has been poorly characterized. This work analyzes the role of different GTPases of the Rho family and some downstream effectors in the internalization of C. burnetii by phagocytic and non-phagocytic cells. The internalization of C. burnetii into HeLa and RAW cells was significantly inhibited when the cells were treated with Clostridium difficile Toxin B which irreversibly inactivates members of the Rho family. In addition, the internalization was reduced in HeLa cells that overexpressed the dominant negative mutants of RhoA, Rac1 or Cdc42 or that were knocked down for the Rho GTPases. The pharmacological inhibition or the knocking down of ROCK diminished bacterium internalization. Moreover, C. burnetii was less efficiently internalized in HeLa cells overexpressing mDia1-N1, a dominant negative mutant of mDia1, while the overexpression of the constitutively active mutant mDia1-ΔN3 increased bacteria uptake. Interestingly, when HeLa and RAW cells were infected, RhoA, Rac1 and mDia1 were recruited to membrane cell fractions. Our results suggest that the GTPases of the Rho family play an important role in C. burnetii phagocytosis in both HeLa and RAW cells.     Somatostatin subtype-2 receptor-targeted metal-based anticancer complexes

    Conjugates of a dicarba analogue of octreotide, a potent somatostatin agonist whose receptors are overexpressed on tumor cells, with [PtCl 2(dap)] (dap = 1-(carboxylic acid)-1,2-diaminoethane) (3), [(η 6-bip)Os(4-CO 2-pico)Cl] (bip = biphenyl, pico = picolinate) (4), [(η 6-p-cym)RuCl(dap)] + (p-cym = p-cymene) (5), and [(η 6-p-cym)RuCl(imidazole-CO 2H)(PPh 3)] + (6), were synthesized by using a solid-phase approach. Conjugates 3-5 readily underwent hydrolysis and DNA binding, whereas conjugate 6 was inert to ligand substitution. NMR spectroscopy and molecular dynamics calculations showed that conjugate formation does not perturb the overall peptide structure. Only 6 exhibited antiproliferative activity in human tumor cells (IC 50 = 63 ± 2 μ in MCF-7 cells and IC 50 = 26 ± 3 μ in DU-145 cells) with active participation of somatostatin receptors in cellular uptake. Similar cytotoxic activity was found in a normal cell line (IC 50 = 45 ± 2.6 μ in CHO cells), which can be attributed to a similar level of expression of somatostatin subtype-2 receptor. These studies provide new insights into the effect of receptor-binding peptide conjugation on the activity of metal-based anticancer drugs, and demonstrate the potential of such hybrid compounds to target tumor cells specifically. © 2012 American Chemical Society
