    A Link Between the Semi-Major Axis of Extrasolar Gas Giant Planets and Stellar Metallicity

    The fact that most extrasolar planets found to date are orbiting metal-rich stars lends credence to the core accretion mechanism of gas giant planet formation over its competitor, the disc instability mechanism. However, the core accretion mechanism is not refined to the point of explaining orbital parameters such as their unexpected semi-major axes and eccentricities. We propose a model, which correlates the metallicity of the host star with the original semi-major axis of its most massive planet, prior to migration, considering that the core accretion scenario governs giant gas planet formation. The model predicts that the optimum regions for planetary formation shift inward as stellar metallicity decreases, providing an explanation for the observed absence of long period planets in metal-poor stars. We compare our predictions with the available data on extrasolar planets for stars with masses similar to the mass of the Sun. A fitting procedure produces an estimate of what we define as the Zero Age Planetary Orbit (ZAPO) curve as a function of the metallicity of the star. The model also hints that the lack of planets circling metal-poor stars may be partly caused by an enhanced destruction probability during the migration process, since the planets lie initially closer to the central stars.Comment: Nature of the replacement: According to recent simulations, the temperature profile, T, is more adequately reproduced by beta = 1 rather than beta = 2.     Bioarchaeological and palaeogenomic portrait of two Pompeians that died during the eruption of Vesuvius in 79 AD

    The archaeological site of Pompeii is one of the 54 UNESCO World Heritage sites in Italy, thanks to its uniqueness: the town was completely destroyed and buried by a Vesuvius’ eruption in 79 AD. In this work, we present a multidisciplinary approach with bioarchaeological and palaeogenomic analyses of two Pompeian human remains from the Casa del Fabbro. We have been able to characterize the genetic profle of the frst Pompeian’ genome, which has strong afnities with the surrounding central Italian population from the Roman Imperial Age. Our fndings suggest that, despite the extensive connection between Rome and other Mediterranean populations, a noticeable degree of genetic homogeneity exists in the Italian peninsula at that time. Moreover, palaeopathological analyses identifed the presence of spinal tuberculosis and we further investigated the presence of ancient DNA from Mycobacterium tuberculosis. In conclusion, our study demonstrates the power of a combined approach to investigate ancient humans and confrms the possibility to retrieve ancient DNA from Pompeii human remains. Our initial fndings provide a foundation to promote an intensive and extensive paleogenetic analysis in order to reconstruct the genetic history of population from Pompeii, a unique archaeological site

    The role of micronutrients in high-yielding dairy ruminants: Choline and vitamin E

    This review addresses the potential role of antioxidants and methyl-group sources in optimising the metabolic health of dairy ruminants. The productivity of high-yielding dairy cows has increased over the past 40 years and the milk yield has doubled. Such increases in milk production have been observed not only in dairy cows but also to some extent in other dairy ruminants such as ewes, goats and buffaloes (Bubalus bubalis). As a consequence, in all specialized dairy ruminants it is essential to optimize the macro and micro-nutrient supply, especially during the most critical period in the animals' production cycle i.e. from parturition until the peak of lactation. In this critical phase, an array of factors can enhance the balance between the intake and demand for nutrients, although the availability and supply of the selected micronutrients is also important. The supplementation of dietary antioxidants or boosting the endogenous methyl group status, via vitamin E, selenium and choline are proposed as possible strategies in maintaining stable metabolic health and optimising milk production

    Tailoring the CRISPR system to transactivate coagulation gene promoters in normal and mutated contexts

    Engineered transcription factors (TF)have expanded our ability to modulate gene expression and hold great promise as bio-therapeutics. The first-generation TF, based on Zinc Fingers or Transcription-Activator-like Effectors (TALE), required complex and time-consuming assembly protocols, and were indeed replaced in recent years by the CRISPR activation (CRISPRa)technology. Here, with coagulation F7/F8 gene promoters as models, we exploited a CRISPRa system based on deactivated (d)Cas9, fused with a transcriptional activator (VPR), which is driven to its target by a single guide (sg)RNA. Reporter gene assays in hepatoma cells identified a sgRNA (sgRNA F7.5 )triggering a ~35-fold increase in the activity of F7 promoter, either wild-type, or defective due to the c.-61T>G mutation. The effect was higher (~15-fold)than that of an engineered TALE-TF (TF4)targeting the same promoter region. Noticeably, when challenged on the endogenous F7 gene, the dCas9-VPR/sgRNA F7.5 combination was more efficient (~6.5-fold)in promoting factor VII (FVII)protein secretion/activity than TF4 (~3.8-fold). The approach was translated to the promoter of F8, whose reduced expression causes hemophilia A. Reporter gene assays in hepatic and endothelial cells identified sgRNAs that, respectively, appreciably increased F8 promoter activity (sgRNA F8.1 , ~8-fold and 3-fold; sgRNA F8.2 , ~19-fold and 2-fold)with synergistic effects (~38-fold and 2.7-fold). Since modest increases in F7/F8 expression would ameliorate patients' phenotype, the CRISPRa-mediated transactivation extent might approach the low therapeutic threshold. Through this pioneer study we demonstrated that the CRISPRa system is easily tailorable to increase expression, or rescue disease-causing mutations, of different promoters, with potential intriguing implications for human disease models

    Efficient Location Training Protocols for Localization in Heterogeneous Sensor and Actor Networks

    International audienceAbstract--In this work we consider a large-scale geographic area populated by tiny sensors and some more powerful devices called actors, authorized to organize the sensors in their vicinity into short-lived, actor-centric sensor networks. The tiny sensors run on miniature non-rechargeable batteries, are anonymous and are unaware of their location. The sensors differ in their ability to dynamically alter their sleep times. Indeed, the periodic sensors have sleep periods of predefined lengths, established at fabrication time; by contrast, the free sensors can dynamically alter their sleep periods, under program control. The main contribution of this work is to propose an energy-efficient location training protocol for heterogeneous actor-centric sensor networks where the sensors acquire coarse-grain location awareness with respect to the actor in their vicinity. Our analytical analysis, confirmed by experimental evaluation, show that the proposed protocol outperforms the best previously-known location training protocols in terms of the number of sleep/awake transitions, overall sensor awake time, and energy consumption

    Rumen-protected choline supplementation in periparturient dairy goats: effects on liver and mammary gland

    The current study investigated the effects of supplementing rumen-protected choline (RPC) on metabolic profile, selected liver constituents and transcript levels of selected enzymes, transcription factors and nuclear receptors involved in mammary lipid metabolism in dairy goats. Eight healthy lactating goats were studied: four received no choline supplementation (CTR group) and four received 4 g RPC chloride/day (RPC group). The treatment was administered individually starting 4 weeks before expected kidding and continuing for 4 weeks after parturition. In the first month of lactation, milk yield and composition were measured weekly. On days 7, 14, 21 and 27 of lactation, blood samples were collected and analysed for glucose, β-hydroxybutyrate, non-esterified fatty acids and cholesterol. On day 28 of lactation, samples of liver and mammary gland tissue were obtained. Liver tissue was analysed for total lipid and DNA content; mammary tissue was analysed for transcripts of lipoprotein lipase (LPL), fatty acid synthase (FAS), sterol regulatory binding proteins 1 and 2, peroxisome proliferator-activated receptor γ and liver X receptor α. Milk yield was very similar in the two groups, but RPC goats had lower (P<0·05) plasma β-hydroxybutyrate. The total lipid content of liver was unaffected (P=0·890), but the total lipid/DNA ratio was lower (both P<0·05) in RPC than CTR animals. Choline had no effect on the expression of the mammary gland transcripts involved in lipid metabolism. The current plasma and liver data indicate that choline has a positive effect on liver lipid metabolism, whereas it appears to have little effect on transcript levels in mammary gland of various proteins involved in lipid metabolism. Nevertheless, the current results were obtained from a limited number of animals, and choline requirement and function in lactating dairy ruminants deserve further investigatio

    Efficient Binary scheme for Training Heterogeneous Sensor Actor Networks

    International audienceSensor networks are expected to evolve into long-lived, autonomous networked systems whose main mission is to provide in-situ users – called actors – with real-time information in support of specific goals supportive of their mission. The network is populated with a heterogeneous set of tiny sensors. The free sensors alternate between sleep and awake periods, under program control in response to computational and communication needs. The periodic sensors alternate between sleep periods and awake periods of predefined lengths, established at the fabrication time. The architectural model of an actor-centric network used in this work comprises in addition to the tiny sensors a set of mobile actors that organize and manage the sensors in their vicinity. We take the view that the sensors deployed are anonymous and unaware of their geographic location. Importantly, the sensors are not, a priori, organized into a network. It is, indeed, the interaction between the actors and the sensor population that organizes the sensors in a disk around each actor into a short-lived, mission-specific, network that exists for the purpose of serving the actor and that will be disbanded when the interaction terminates. The task of setting up this form of actor-centric network involves a training stage where the sensors acquire dynamic coordinates relative to the actor in their vicinity. The main contribution of this work is to propose an energy- efficient training protocol for actor-centric heterogeneous sensor networks. Our protocol outperforms all know training protocols in the number of sleep/awake transitions per sensor needed by the training process.     Legal Status and the Criminal Activity of Immigrants

    We exploit exogenous variation in legal status following the January 2007 European Union enlargement to estimate its effect on immigrant crime. We difference out unobserved time-varying factors by (i) comparing recidivism rates of immigrants from the “new” and “candidate” member countries; and (ii) using arrest data on for eign detainees released upon a mass clemency that occurred in Italy in August 2006. The timing of the two events allows us to setup a difference-in-differences strategy. Legal status leads to a 50 percent reduction in recidivism, and explains one-half to two-thirds of the observed differences in crime rates between legal and illegal immigrants

    A Drone-based Application for Scouting Halyomorpha halys Bugs in Orchards with Multifunctional Nets

    In this work, we consider the problem of using a drone to collect information within orchards in order to scout insect pests, i.e., the stink bug Halyomorpha halys. An orchard can be modeled as an aisle-graph, which is a regular and constrained data structure formed by consecutive aisles where trees are arranged in a straight line. For monitoring the presence of bugs, a drone flies close to the trees and takes videos and/or pictures that will be analyzed offline. As the drone's energy is limited, only a subset of locations in the orchard can be visited with a fully charged battery. Those places that are most likely to be infested should be selected to promptly detect the pest. We implemented the proposed approach on a DJI drone and evaluated its performance in the real-world environment
