    Cell surface carbohydrates existing as glycoproteins and glycolipids represent the first information about the cell with the outside world and are closely involved in various biological processes such as cell communication, and the molecular recognition and cell targeting. However, the mechanisms of most of the processes at the molecular level are still unclear. It is therefore very important to develop specific carbohydrate-binding molecules for a fast, efficient, sensitive and accurate analysis of complex carbohydrates structures and functions, known as glycomics. In addition, certain carbohydrate-binding molecules can be used in medical applications such as biomarkers for diseases diagnosis and targeted drug delivery applications. In this thesis study, I conducted synthesis, characterization and glyco-capturing capacity investigation of bovine serum albumin (BSA)-boronic acid (BA) conjugates as lectin mimetics. The BSA-BA conjugates were synthesized by amidation of carboxylic acid groups in BSA with aminophenyl boronic acid in the presence of EDC, and were characterized by Alizarin Red S (ARS) assay and SDS-PAGE gel. The BSA-BA conjugates were immobilized onto maleimide-functionalized silica beads and their sugar capturing capacity and specificity were confirmed by Alizarin Red displacement assay. As a result, the interactions between the sugars and SB-BSA-BA was in the order of Fructose \u3eGalactose \u3eMannose \u3eFucose \u3eLactose \u3eSialic Acid \u3eGlcNAc \u3eGlucose at pH 7.4 and Fructose \u3eFucose \u3eLactose \u3eGalactose \u3eMannose \u3eGlcNAc \u3eSialic Acid \u3eGlucose at pH 8.3. Further, SPR analysis of BSA-BA-based glyco-capturing test was conducted by immobilizing BSA-BA onto SPR gold chip. As a result, the interactions between the sugars and BSA-BA on the gold chip were in the order of Lactose \u3eMannose \u3eGlucose \u3eFucose \u3eSialic acid \u3eFructose \u3eGlcNAc \u3eGalactose at pH7.4 and Glucose \u3eGalactose \u3eMannose \u3eGlcNAc \u3eFructose \u3eFucose \u3eSialic acid \u3eLactose at pH8.3.     The quantity of test that is added to the web in the digital form continues to grow and the quest for tools that can process this huge amount of data to retrieve the data of our interest is an ongoing process. Moreover, observing these large volumes of data over a period of time is a tedious task for any human being. Text mining is very helpful in performing these kinds of tasks. Text mining is a process of observing patterns in the text data using sophisticated statistical measures both quantitatively and qualitatively. Using these text mining techniques and the power of the internet and its technologies, we have developed a tool that retrieves documents concerning topics of interest, which utilizes novel and sensitive classification tools.;This thesis presents an intelligent web crawler, named Intel-Crawl. This tool identifies web pages of interest without the user\u27s guidance or monitoring. Documents of interest are logged (by URL or file name). This package uses automatically generated semantic signatures to identify documents with content of interest. The tool also produces a vector that is a quantification of a document\u27s content based on the semantic signatures. This provides a rich and sensitive characterization of the document\u27s content. Documents are classified according to content and presented to the user for further analysis and investigation.;Intel-Crawl may be applied to any area of interest. It is likely to be very useful in areas such as law enforcement, intelligence gathering, and monitoring changes in web site contents over time. It is well-suited for scrutinizing the web activity of large collection of web pages pertaining to similar content. The utility of Intel-Crawl is demonstrated in various situations using different parameters and classification techniques

    Wavelet-Based Volume Rendering

    Various biomedical technologies like CT, MRI and PET scanners provide detailed cross-sectional views of the human anatomy. The image information obtained from these scanning devices is typically represented as large data sets whose sizes vary from several hundred megabytes to about one hundred gigabytes. As these data sets cannot be stored on one\u27s local hard drive, SDSC provides a large data repository to store such data sets. These data sets need to be accessed by researchers around the world to collaborate in their research. But the size of these data sets make them difficult to be transmitted over the current network. This thesis presents a 3-D Haar wavelet algorithm which enables these data sets to be transformed into smaller hierarchical representations. These transformed data sets are transmitted over the network and reconstructed to a 3-D volume on the client\u27s side through progressive refinement of the images and 3-D texture mapping techniques

    Development and validity of the “Red flag developmental screening checklist”

    The aim of this pilot study was to develop an indigenous, simple developmental screening tool based on red flags, for administration to Indian children at routine immunization visits and to determine its validity. A 44 items-checklist was designed by listing milestones based on the red flags for the age- groups 3.5-4.5months, 9-12months, 18-20months and 24-26months, in 6 domains- GrossMotor, Fine-Motor, Language, Social, Vision, Hearing. This checklist was administered and difficulties experienced by parents in comprehension were recorded. Then the child underwent evaluation by a developmental expert that included administration of Developmental Profile II and neurodevelopmental assessment. The validity of checklist for detecting developmental delay was calculated. The RFDSC was administered to 102 subjects (29 aged 3.5-4.5months, 32 aged 9- 12months, 26 aged 18-20months, 15 aged 24-26months). 43% were screening test positive (RFDSC fail). The average time taken for its administration was 2.37 minutes (SD=0.62). 32% mothers faced problems in comprehension of certain questions. The RFDSC was found to be a fairly valid screening tool (sensitivity 63%; specificity 87%). The RFDSC was maximally valid for the age-group 3.5- 4.5months with a sensitivity of 88% and a specificity of 90%. However, the sensitivity of the checklist for the age-groups 18-20months and 24-26months was less than 50% with a specificity of 100%. The checklist developed is parent-report based, quick to administer, easily comprehensible, having easy scoring pattern and interpretation. The checklist can be a valid screening tool for the evaluation of development in children coming for immunization in the age-groups 3.5- 4.5months and 9-12monţhs. The ultimate goal is incorporating the administration of this checklist as a part of National Immunization Program so as to facilitate early identification and intervention in developmentally delayed children
