    Synchronized somatic embryo development in embryogenic suspensions of grapevine Muscadinia rotundifolia (Michx.) Small

    High-frequency, synchronous embryogenic systems in liquid culture facilitate plant regeneration and can be used as an essential model for performing functional genomics studies and understanding molecular aspect of the ontogenesis of higher plants. In the present study, synchronous somatic embryogenic cultures were developed for Muscadinia rotundifolia cv. Darlene and Vitis vinifera cv. Velika. High cell density and presence of 2,4-dichlorophenoxyacetic acid (2,4-D) proved to be essential for the establishment of the suspension cultures. Low cell density and continuous availability of auxin (2,4-D) was crucial for maintenance of suspension cultures. High cell density and withdrawal of 2,4-D is sufficient to advance somatic embryo development toward embryo differentiation and plantlets regeneration. Cells and cell clusters fractionation by density gradient centrifugation in Ficoll solution demonstrated to be a suitable method for separation of subpopulations with various potential for embryo development. The high frequency of synchronous development and differentiation of somatic embryos was attained essentially for the heaviest (at 16-18 % and >18 % Ficoll layer) cell population

    How to Build Collective Capabilities: The 3C-Model for Grassroots-led Development

    Capabilities need to be built from the bottom-up. Social innovations at the grassroots seek to present new solutions to existing social problems. However, since the poor suffer from limitations on their individual capabilities and agency, they engage in acts of collective agency to generate new collective capabilities that each individual alone would not be able to achieve. The question is: how can these acts of collective agency be initiated, supported and sustained in practice? What roles can development actors (such as the state, donors and NGOs) play in supporting these acts of collective agency? Drawing on the literature on social innovation, the capability approach, participation and empowerment, the paper argues that three crucial C-processes are integral conditions for promoting successful, scalable and sustainable social innovations at the grassroots, namely: (1) Conscientization; (2) Conciliation and (3) Collaboration. By linking the individual, collective and institutional levels of analysis, the paper demonstrates the importance of individual behavioural changes, collective agency and local institutional reforms for the success, sustainability and scalability of social innovations at the grassroots. The paper acknowledges conflict, capture and cooptation as potential limitations and recognizes the role of contextual factors in initiating, implementing and sustaining social innovations at the grassroots

    Assistive technologies to address capabilities of people with dementia: from research to practice

    Assistive technologies (AT) became pervasive and virtually present in all our life domains. They can be either an enabler or an obstacle leading to social exclusion. The Fondation Médéric Alzheimer gathered international experts of dementia care, with backgrounds in biomedical, human and social sciences, to analyse how AT can address the capabilities of people with dementia, on the basis of their needs. Discussion covered the unmet needs of people with dementia, the domains of daily life activities where AT can provide help to people with dementia, the enabling and empowering impact of technology to improve their safety and wellbeing, barriers and limits of use, technology assessment, ethical and legal issues. The capability approach (possible freedom) appears particularly relevant in person-centered dementia care and technology development. The focus is not on the solution, rather on what the person can do with it: seeing dementia as disability, with technology as an enabler to promote capabilities of the person, provides a useful framework for both research and practice. This article summarizes how these concepts took momentum in professional practice and public policies in the past fifteen years (2000-2015), discusses current issues in the design, development and economic model of AT for people with dementia, and covers how these technologies are being used and assessed

    Are social innovation paradigms incommensurable?

    This paper calls attention to the problematic use of the concept of social innovation which remains undefined despite its proliferation throughout academic and policy discourses. Extant research has thus far failed to capture the socio-political contentions which surround social innovation. This paper therefore draws upon the work of Thomas Kuhn and conducts a paradigmatic analysis of the field of social innovation which identifies two emerging schools: one technocratic, the other democratic. The paper identifies some of the key thinkers in each paradigm and explains how the struggle between these two paradigms reveals itself to be part of a broader conflict between neoliberalism and it opponents and concludes by arguing that future research focused upon local contextualised struggles will reveal which paradigm is in the ascendancy

    Hyper-IgG4 disease: report and characterisation of a new disease

    BACKGROUND: We highlight a chronic inflammatory disease we call 'hyper-IgG4 disease', which has many synonyms depending on the organ involved, the country of origin and the year of the report. It is characterized histologically by a lymphoplasmacytic inflammation with IgG4-positive cells and exuberant fibrosis, which leaves dense fibrosis on resolution. A typical example is idiopathic retroperitoneal fibrosis, but the initial report in 2001 was of sclerosing pancreatitis. METHODS: We report an index case with fever and severe systemic disease. We have also reviewed the histology of 11 further patients with idiopathic retroperitoneal fibrosis for evidence of IgG4-expressing plasma cells, and examined a wide range of other inflammatory conditions and fibrotic diseases as organ-specific controls. We have reviewed the published literature for disease associations with idiopathic, systemic fibrosing conditions and the synonyms: pseudotumour, myofibroblastic tumour, plasma cell granuloma, systemic fibrosis, xanthofibrogranulomatosis, and multifocal fibrosclerosis. RESULTS: Histology from all 12 patients showed, to varying degrees, fibrosis, intense inflammatory cell infiltration with lymphocytes, plasma cells, scattered neutrophils, and sometimes eosinophilic aggregates, with venulitis and obliterative arteritis. The majority of lymphocytes were T cells that expressed CD8 and CD4, with scattered B-cell-rich small lymphoid follicles. In all cases, there was a significant increase in IgG4-positive plasma cells compared with controls. In two cases, biopsies before and after steroid treatment were available, and only scattered plasma cells were seen after treatment, none of them expressing IgG4. Review of the literature shows that although pathology commonly appears confined to one organ, patients can have systemic symptoms and fever. In the active period, there is an acute phase response with a high serum concentration of IgG, and during this phase, there is a rapid clinical response to glucocorticoid steroid treatment. CONCLUSION: We believe that hyper-IgG4 disease is an important condition to recognise, as the diagnosis can be readily verified and the outcome with treatment is very good

    Does Reducing Implicit Prejudice Increase Out-Group Identification? The Downstream Consequences of Evaluative Training on Associations Between the Self and Racial Categories

    The present experiments were designed to investigate whether an intervention that targeted racial attitudes influenced not only prejudice but also self–Black associations. Because past research has demonstrated that people strive to build connections with favorable social categories, we predicted that positive evaluative training would increase identification with Blacks. Results from three studies provide evidence that practice in associating positive concepts with Blacks reduced implicit prejudice which in turn increased implicit self–Black associations. Notably, prejudice, in this case, had an intervening variable effect. Study 3 also investigated the impact of an alternative intervention that directly targeted self-associations rather than racial attitudes. Unlike evaluative training, associating the self with Blacks directly reduced both implicit prejudice and increased self–Black associations. These findings extend theorizing on the causal relationship between prejudice and out-group identification and provide important process information on how particular interventions reduce intergroup biases
