    One-step grafting of polymers to graphene oxide

    The direct grafting of poly(N-isopropylacrylamide) to the basal plane of graphene oxide has been achieved in a single step: cleavage of the terminal thiocarbonylthio group on RAFT grown poly(N-isopropylacrylamide) reveals a reactive thiol that attacks the epoxides present across the surface of graphene oxide.     A massive nebula around the Luminous Blue Variable star RMC143 revealed by ALMA

    The luminous blue variable (LBV) RMC143 is located in the outskirts of the 30~Doradus complex, a region rich with interstellar material and hot luminous stars. We report the 3σ3\sigma sub-millimetre detection of its circumstellar nebula with ALMA. The observed morphology in the sub-millimetre is different than previously observed with HST and ATCA in the optical and centimetre wavelength regimes. The spectral energy distribution (SED) of RMC143 suggests that two emission mechanisms contribute to the sub-mm emission: optically thin bremsstrahlung and dust. Both the extinction map and the SED are consistent with a dusty massive nebula with a dust mass of 0.055±0.018 M0.055\pm0.018~M_{\odot} (assuming κ850=1.7cm2g1\kappa_{850}=1.7\rm\,cm^{2}\,g^{-1}). To date, RMC143 has the most dusty LBV nebula observed in the Magellanic Clouds. We have also re-examined the LBV classification of RMC143 based on VLT/X-shooter spectra obtained in 2015/16 and a review of the publication record. The radiative transfer code CMFGEN is used to derive its fundamental stellar parameters. We find an effective temperature of 8500\sim 8500~K, luminosity of log(L/L)=5.32(L/L_{\odot}) = 5.32, and a relatively high mass-loss rate of 1.0×105 M1.0 \times 10^{-5}~M_{\odot}~yr1^{-1}. The luminosity is much lower than previously thought, which implies that the current stellar mass of 8 M\sim8~M_{\odot} is comparable to its nebular mass of 5.5 M\sim 5.5~M_{\odot} (from an assumed gas-to-dust ratio of 100), suggesting that the star has lost a large fraction of its initial mass in past LBV eruptions or binary interactions. While the star may have been hotter in the past, it is currently not hot enough to ionize its circumstellar nebula.     Frequency, intensity and duration of physical tasks performed by Australian rural firefighters during bushfire suppression

    The current study combined, for the first time, video footage of individual firefighters wearing heart rate monitors and personal GPS units to quantify the frequency, duration and intensity of tasks performed by Australian rural fire crews when. suppressing bushfires. Across the four fireground \u27shifts\u27, the firefighters performed 34 distinct fireground tasks. Per shift, the task frequency ranged from once (raking fireline in teams, carrying a quick fill pump) to 103 times {lateral repositioning of a 38-mm charged firehose) .. The tasks lasted between 4 &plusmn; 2 s (bowling out 3-8-mm firehose) and 461 &plusmn; 387 s (raking fireline in teams). The task intensity~ as measured by average heart rate ranged between 97 &plusmn; 16 beats&middot;min-1 (55.7 &plusmn; 8.7. %HRmax) and 157 &plusmn; 15 beats&middot;min-1 (86.2 &plusmn; 10.8 %HRmax)&middot; The tasks were performed a speeds that ranged from 0.12 &plusmn; 0.08 m&middot;s-1 (manual hose retraction of 38-mm charged firehose) to 0.79 &plusmn; 0.40 m&middot;s-1 (carrying a 38-mm coiled hose). Tasks found to be simultaneously frequent, long and intense (or two of these three) are likely to form the basis for job-specific testing of Australian rural firefighters suppressing bushfires.<br /

    The Team Sports Risk Exposure Framework (TS-REF) was developed in July 2020 by experts in sports medicine, virology, sports science and public health to facilitate the safe return of sport during the COVID-19 pandemic. The TS-REF was developed at the time when the outdoor transmission risk of SARS-CoV-2 during sport was unknown. The TS-REF has been adopted by Public Health England and the UK Government (Department for Digital, Culture, Media and Sport), for use within both elite and community sports, to both determine the risk of SARS-CoV-2 transmission during specific sporting activities (eg, rugby tackle), and to identify and isolate increased risk contacts during sport. The TS-REF classified increased risk contacts as player-to-player interactions ‘within 1 m, directly face to face, for 3 or more seconds’

    Beta-lactam-induced immediate hypersensitivity reactions: A genome-wide association study of a deeply phenotyped cohort

    Background β-lactam antibiotics are associated with a variety of immune-mediated or hypersensitivity reactions, including immediate (Type I) reactions mediated by antigen-specific IgE. Objective To identify genetic predisposing factors for immediate reactions to β-lactam antibiotics. Methods Patients with a clinical history of immediate hypersensitivity reactions to either penicillins or cephalosporins, which were immunologically confirmed, were recruited from allergy clinics. A genome-wide association study (GWAS) was conducted on 662 patients (the discovery cohort) with a diagnosis of immediate hypersensitivity and the main finding was replicated in a cohort of 98 Spanish cases, recruited using the same diagnostic criteria as the discovery cohort. Results GWAS identified rs71542416 within the Class II HLA region as the top hit (P = 2x10-14); this was in linkage disequilibrium with HLA-DRB1*10:01 (OR = 2.93 P = 5.4x10-7) and HLA-DQA1*01:05 (OR=2.93, P=5.4x10-7). Haplotype analysis identified that HLA-DRB1*10:01 was a risk factor even without the HLA-DQA1*01:05 allele. The association with HLA-DRB1*10:01 was replicated in another cohort, with the meta-analysis of the discovery and replication cohorts showing that HLA-DRB1*10:01 increased the risk of immediate hypersensitivity at a genome-wide level (OR = 2.96 P=4.1x10-9). No association with HLA-DRB1*10:01 was identified in 268 patients with delayed hypersensitivity reactions to β-lactams. Conclusion HLA-DRB1*10:01 predisposed to immediate hypersensitivity reactions to penicillins. Further work to identify other predisposing HLA and non-HLA loci is required. Clinical implications This novel insight into the mechanisms of immediate reactions associated with penicillins may be of use in risk stratifying patients where penicillin cannot be excluded as an etiological agent

    Wdpcp, a PCP Protein Required for Ciliogenesis, Regulates Directional Cell Migration and Cell Polarity by Direct Modulation of the Actin Cytoskeleton

    Planar cell polarity (PCP) regulates cell alignment required for collective cell movement during embryonic development. This requires PCP/PCP effector proteins, some of which also play essential roles in ciliogenesis, highlighting the long-standing question of the role of the cilium in PCP. Wdpcp, a PCP effector, was recently shown to regulate both ciliogenesis and collective cell movement, but the underlying mechanism is unknown. Here we show Wdpcp can regulate PCP by direct modulation of the actin cytoskeleton. These studies were made possible by recovery of a Wdpcp mutant mouse model. Wdpcp-deficient mice exhibit phenotypes reminiscent of Bardet-Biedl/Meckel-Gruber ciliopathy syndromes, including cardiac outflow tract and cochlea defects associated with PCP perturbation. We observed Wdpcp is localized to the transition zone, and in Wdpcp-deficient cells, Sept2, Nphp1, and Mks1 were lost from the transition zone, indicating Wdpcp is required for recruitment of proteins essential for ciliogenesis. Wdpcp is also found in the cytoplasm, where it is localized in the actin cytoskeleton and in focal adhesions. Wdpcp interacts with Sept2 and is colocalized with Sept2 in actin filaments, but in Wdpcp-deficient cells, Sept2 was lost from the actin cytoskeleton, suggesting Wdpcp is required for Sept2 recruitment to actin filaments. Significantly, organization of the actin filaments and focal contacts were markedly changed in Wdpcp-deficient cells. This was associated with decreased membrane ruffling, failure to establish cell polarity, and loss of directional cell migration. These results suggest the PCP defects in Wdpcp mutants are not caused by loss of cilia, but by direct disruption of the actin cytoskeleton. Consistent with this, Wdpcp mutant cochlea has normal kinocilia and yet exhibits PCP defects. Together, these findings provide the first evidence, to our knowledge, that a PCP component required for ciliogenesis can directly modulate the actin cytoskeleton to regulate cell polarity and directional cell migration

    Advances in genome sequencing and assembly technologies are generating many high-quality genome sequences, but assemblies of large, repeat-rich polyploid genomes, such as that of bread wheat, remain fragmented and incomplete. We have generated a new wheat whole-genome shotgun sequence assembly using a combination of optimized data types and an assembly algorithm designed to deal with large and complex genomes. The new assembly represents >78% of the genome with a scaffold N50 of 88.8 kb that has a high fidelity to the input data. Our new annotation combines strand-specific Illumina RNA-seq and Pacific Biosciences (PacBio) full-length cDNAs to identify 104,091 high-confidence protein-coding genes and 10,156 noncoding RNA genes. We confirmed three known and identified one novel genome rearrangements. Our approach enables the rapid and scalable assembly of wheat genomes, the identification of structural variants, and the definition of complete gene models, all powerful resources for trait analysis and breeding of this key global crop
