Shared determinants between virus-infected and trinitrophenyl-conjugated H-2-identical target cells detected in cell-mediated lympholysis

Infection of H-2-identical mice with either lymphocytic choriomeningitis (LCM) virus, vaccinia virus, or paramyxo (Sendai) virus resulted in the generation of specifically sensitized cytotoxic T lymphocytes (CTL). CTL generated in vitro against 2,4,6-trinitrophenyl (TNP)-conjugated syngeneic stimulator cells were specifically cytotoxic for TNP-conjugated H-2K(D) region identical targets. Both LCM and vaccinia-induced CTL, however, were found to be strongly cytotoxic towards TNP-conjugated, H-2K(D) region-identical target cells. In contrast, Sendai virus-induced CTL did not lyse TNP-conjugated, syngeneic target cells. Inhibition experiments using cold targets suggested that shared antigenic determinants can be detected on either LCM virus-infected and TNP-conjugated targets, which are not present on the cell surface 6f normal target cells

Experience of Multiplicity, Multiplicity’s Experience. From a Critical Genealogy of Homo Economicus to a Contemporary Machine Phenomenology

In Experience. New Foundations for the Human Sciences, sociologist Scott Lash addresses the human sciences in general. He proposes a new perspective on the methodological status of experience. His book covers a vast array of fields and topics: rooted in sociology and cultural studies, Lash takes his readers on a profound, sometimes swift journey from Greek antiquity to the Methodenstreit in 19th century sociology, from Kant’s transcendentalism to Arendt’s political thought and from the rise of cybernetics to Chinese art and aesthetics – always focussed on the historical conditions, role, and function of experience. Lash’s aim is twofold: firstly he is presenting a critique of the homo economicus, the currently dominant ideology of the Western world, by a genealogy of objective experience and formalism in the social sciences and economics. Secondly, he develops, with Aristotle, Hannah Arendt, William James but also Chinese thought, a perspective on subjective experience suitable for our present

Unfinished Business, Continuous Growth and Persisting Interests. Making Sense of Foucault’s Last Years

In Foucault’s Last Decade Stuart Elden portrays the intellectual history of the last ten years of Michel Foucault’s life. By referring to published and unpublished sources (some only newly available to the public) as well as the testimonies of friends and colleagues, he convincingly reconstructs the development from Discipline and Punish via biopolitics and governmentality to Foucault’s interest in sexuality and antique ethics. Centering on Foucault’s main works and courses at the Collège de France from 1974 to 1984, Elden brilliantly discusses key concepts at their first emergence and concisely traces their development in regard to the entirety of Foucault’s work. He also promptly discusses various issues that shaped Foucault’s work, intellectually and institutionally

Insulin selectively activates STAT5b, but not STAT5a, via a JAK2-independent signalling pathway in Kym-1 rhabdomyosarcoma cells

AbstractThe STAT multigene family of transcriptional regulators conveys signals from several cytokines and growth factors upon phosphorylation by janus kinases (JAK). Activation of STAT5 is typically mediated by JAK2, but more recent data indicate a direct activation by the insulin receptor kinase. STAT5 exists in two closely homologous isoforms, STAT5a and b. We here describe the selective tyrosine phosphorylation of STAT5b in Kym-1 cells in response to insulin. Blocking insulin signalling by HNMPA-(AM)3, an insulin receptor kinase inhibitor, resulted in the loss of insulin-induced STAT5b tyrosine phosphorylation, whereas the inhibition of JAK2 by the JAK selective inhibitor tyrphostin AG490 had no effect. By contrast, in the same cells, IFNÎł-induced STAT5b activation was JAK2-dependent, indicating that this signal pathway is functional in Kym-1 cells. We conclude from this rhabdomyosarcoma model that STAT5b, but not STAT5a is a direct target of the insulin receptor kinase

The cytolytic T lymphocyte response to the murine cytomegalovirus

During the acute cytolytic T lymphocyte (CTL) response of mice to infection with the murine cytomegalovirus two independent populations of activated interleukin-receptive CTL precursors can be demonstrated. One population is specific for cell membrane-incorporated viral structural antigens, whereas the second population detects an antigen, whose appearance is correlated with the synthesis of viral immediate early proteins. Since this new type of antigen is only defined by lymphocyte recognition, it is referred to as the lymphocyte-detected immediate early antigen (LYDIEA). Expression of immediate early antigen precedes the production of viral progeny and, therefore, it is possible that LYDIEA-specific CTL could serve as indicator cells for the very first activities of the viral genome, even during nonproductive infection
