    Differential expression of aquaporin 3 in Triturus italicus from larval to adult epidermal conversion

    By using immunohistochemical techniques applied to confocal microscopy, the presence of aquaporin 3 water channel in the epidermis of Triturus italicus (Amphibia, Urodela) has been shown. We analysed the expression of aquaporin 3 (AQP3) during the larval, pre-metamorphic and adult phases; we also showed the localization of the water-channel protein AQP3 in free-swimming conditions and during aestivation in parallel with histological analysis of the skin, focusing on the possible relationship between protein expression and terrestrial habitats. Our results indicate that aquaporin is produced as the epidermis modifies during the functional maturation phase starting at the climax. Moreover, our data suggest an increase in enzyme expression in aestivating newts emphasizing the putative functional importance of differential expression related to a distinct phase of the biological cycle

    Host lifestyle affects human microbiota on daily timescales

    Background: Disturbance to human microbiota may underlie several pathologies. Yet, we lack a comprehensive understanding of how lifestyle affects the dynamics of human-associated microbial communities. Results: Here, we link over 10,000 longitudinal measurements of human wellness and action to the daily gut and salivary microbiota dynamics of two individuals over the course of one year. These time series show overall microbial communities to be stable for months. However, rare events in each subjects’ life rapidly and broadly impacted microbiota dynamics. Travel from the developed to the developing world in one subject led to a nearly two-fold increase in the Bacteroidetes to Firmicutes ratio, which reversed upon return. Enteric infection in the other subject resulted in the permanent decline of most gut bacterial taxa, which were replaced by genetically similar species. Still, even during periods of overall community stability, the dynamics of select microbial taxa could be associated with specific host behaviors. Most prominently, changes in host fiber intake positively correlated with next-day abundance changes among 15% of gut microbiota members. Conclusions: Our findings suggest that although human-associated microbial communities are generally stable, they can be quickly and profoundly altered by common human actions and experiences.National Science Foundation (U.S.) (Grant 0821391

    Biomechanically tunable nano-silica/p-hema structural hydrogels for bone scaffolding

    Innovative tissue engineering biomimetic hydrogels based on hydrophilic polymers have been investigated for their physical and mechanical properties. 5% to 25% by volume loading PHEMA-nanosilica glassy hybrid samples were equilibrated at 37◦C in aqueous physiological isotonic and hypotonic saline solutions (0.15 and 0.05 M NaCl) simulating two limiting possible compositions of physiological extracellular fluids. The glassy and hydrated hybrid materials were characterized by both dynamo-mechanical properties and equilibrium absorptions in the two physiological-like aqueous solutions. The mechanical and morphological modifications occurring in the samples have been described. The 5% volume nanosilica loading hybrid nanocomposite composition showed mechanical characteristics in the dry and hydrated states that were comparable to those of cortical bone and articular cartilage, respectively, and then chosen for further sorption kinetics characterization. Sorption and swelling kinetics were monitored up to equilibrium. Changes in water activities and osmotic pressures in the water-hybrid systems equilibrated at the two limiting solute molarities of the physiological solutions have been related to the observed anomalous sorption modes using the Flory-Huggins interaction parameter approach. The bulk modulus of the dry and glassy PHEMA-5% nanosilica hybrid at 37◦C has been observed to be comparable with the values of the osmotic pressures generated from the sorption of isotonic and hypotonic solutions. The anomalous sorption modes and swelling rates are coherent with the difference between osmotic swelling pressures and hybrid glassy nano-composite bulk modulus: the lower the differences the higher the swelling rate and equilibrium solution uptakes.     Nature and statistical properties of quasar associated absorption systems in the XQ-100 Legacy Survey

    We statistically study the physical properties of a sample of narrow absorption line (NAL) systems looking for empirical evidences to distinguish between intrinsic and intervening NALs without taking into account any a priori definition or velocity cut-off. We analyze the spectra of 100 quasars with 3.5 < zem\rm_{em} < 4.5, observed with X-shooter/VLT in the context of the XQ-100 Legacy Survey. We detect a \sim 8 σ\sigma excess in the number density of absorbers within 10,000 km/s of the quasar emission redshift with respect to the random occurrence of NALs. This excess does not show a dependence on the quasar bolometric luminosity and it is not due to the redshift evolution of NALs. It extends far beyond the standard 5000 km/s cut-off traditionally defined for associated absorption lines. We propose to modify this definition, extending the threshold to 10,000 km/s when also weak absorbers (equivalent width < 0.2 \AA) are considered. We infer NV is the ion that better traces the effects of the quasar ionization field, offering the best statistical tool to identify intrinsic systems. Following this criterion we estimate that the fraction of quasars in our sample hosting an intrinsic NAL system is 33 percent. Lastly, we compare the properties of the material along the quasar line of sight, derived from our sample, with results based on close quasar pairs investigating the transverse direction. We find a deficiency of cool gas (traced by CII) along the line of sight associated with the quasar host galaxy, in contrast with what is observed in the transverse direction.Comment: 18 pages, 13 figures, 5 table

    Autophagy-mediated neuroprotection induced by octreotide in an ex vivo model of early diabetic retinopathy

    Neuronal injury plays a major role in diabetic retinopathy (DR). Our hypothesis was that the balance between neuronal death and survival may depend on a similar equilibrium between apoptosis and autophagy and that a neuroprotectant may act by influencing this equilibrium. Ex vivo mouse retinal explants were treated with high glucose (HG) for 10days and the somatostatin analog octreotide (OCT) was used as a neuroprotectant. Chloroquine (CQ) was used as an autophagy inhibitor. Apoptotic and autophagic markers were evaluated using western blot and immunohistochemistry. HG-treated explants displayed a significant increase of apoptosis paralleled by a significant decrease of the autophagic flux, which was likely to be due to increased activity of the autophagy regulator mTOR (mammalian target of rapamycin). Treatment with OCT rescued HG-treated retinal explants from apoptosis and determined an increase of autophagic activity with concomitant mTOR inhibition. Blocking the autophagic flux with CQ completely abolished the anti-apoptotic effect of OCT. Immunohistochemical observations showed that OCT-induced autophagy is localized to populations of bipolar and amacrine cells and to ganglion cells. These observations revealed the antithetic role of apoptosis and autophagy, highlighting their equilibrium from which neuronal survival is likely to depend. These data suggest the crucial role covered by autophagy, which could be considered as a molecular target for DR neuroprotective treatment strategies

    Remission in ankylosing spondylitis treated with anti-TNF-α drugs: a national multicentre study

    Objective: The primary objective of this retrospective study was to investigate the possibility of achieving partial remission (PR) in AS patients treated with anti-TNF-α antagonists, such as adalimumab (ADA), etanercept (ETA) and infliximab (INF), in a real clinical practice setting. Predictors of PR were also evaluated. Methods: A retrospective study was conducted in patients with AS treated with ADA, ETA and INF from 2000 to 2012. Kaplan-Meier survival curves were plotted to determine the rates of PR during the treatment with anti-TNF-α drugs. Results: A total of 283 patients with AS were treated with ADA (18.7%), ETA (26.8%) and INF (54.4%) as first anti-TNF-α drugs, with a PR rate of 57.6%. The probability of obtaining PR with ADA, ETA or INF was not significantly different among all anti-TNF-α patients. AS patients treated with a second anti-TNF-α drug had a PR rate of 40.5%, but after switching for lack of response, the probability of obtaining PR with a second anti-TNF-α drug was significantly lower from that of the first anti-TNF-α drug (P = 0.0039). The probability of obtaining PR in patients with enthesitis (P = 0.04) or psoriasis (P = 0.0016) or low levels of CRP (P = 0.0225) was significantly lower compared with that of patients without these manifestations at baseline. Conclusion: Our real-life study on PR confirmed the effectiveness of ADA, ETA or INF as first or second anti-TNF-α drugs. The presence at baseline of enthesitis or psoriasis or low CRP values yielded a lower probability of obtaining PR. © The Author 2013.     Equation of State of Oscillating Brans-Dicke Scalar and Extra Dimensions

    We consider a Brans-Dicke scalar field stabilized by a general power law potential with power index nn at a finite equilibrium value. Redshifting matter induces oscillations of the scalar field around its equilibrium due to the scalar field coupling to the trace of the energy momentum tensor. If the stabilizing potential is sufficiently steep these high frequency oscillations are consistent with observational and experimental constraints for arbitrary value of the Brans-Dicke parameter ω\omega. We study analytically and numerically the equation of state of these high frequency oscillations in terms of the parameters ω\omega and nn and find the corresponding evolution of the universe scale factor. We find that the equation of state parameter can be negative and less than -1 but it is not related to the evolution of the scale factor in the usual way. Nevertheless, accelerating expansion is found for a certain parameter range. Our analysis applies also to oscillations of the size of extra dimensions (the radion field) around an equilibrium value. This duality between self-coupled Brans-Dicke and radion dynamics is applicable for ω=1+1/D\omega= -1 + 1/D where D is the number of extra dimensions.Comment: 10 two-column pages, RevTex4, 8 figures.     Phenomenology of a realistic accelerating universe using only Planck-scale physics

    Modern data is showing increasing evidence that the Universe is accelerating. So far, all attempts to account for the acceleration have required some fundamental dimensionless quantities to be extremely small.
