Publisher Copyright: © 2023 The AuthorsThe caregiver's touch behavior during early infancy is linked to multiple developmental outcomes. However, social touch remains a challenging construct to operationalize, and although observational tools have been a gold standard for measuring touch in caregiver-infant interactions, no systematic review has been conducted before. We followed the PRISMA guidelines and reviewed the literature to describe and classify the main characteristics of the available observational instruments. Of the 3042 publications found, we selected 45 that included an observational measure, and from those we identified 12 instruments. Most of the studies were of infants younger than six months of age and assessed touch in two laboratory tasks: face-to-face interaction and still-face procedure. We identified three approaches for evaluating the caregiver's touch behavior: strictly behavioral (the observable touch behavior), functional (the functional role of the touch behavior), or mixed (a combination of the previous two). Half of the instruments were classified as functional, 25% as strictly observational, and 25% as mixed. The lack of conceptual and operational uniformity and consistency between instruments is discussed.publishersversionpublishe

    Etoposide e dexametasona como primeira linha em idosos com comorbidades portadores de Linfoma Difuso de Grandes Células B

    -Pacientes idosos com linfoma difuso de grandes células B (LDGCB) são frequentemente excluídos de estudos clínicos. A utilização de terapias curativas muitas vezes é impossibilitada em virtude das comorbidades apresentadas por esta população ao diagnóstico. Nós adotamos um protocolo alternativo de quimioterapia oral combinando um inibidor da topoisomerase II e dexametasona. Apresentamos os resultados parciais com este protocolo em três pacientes portadores de LDGCB com idade superior a 80 anos e comorbidades severas. Todos alcançaram remissão completa com baixa toxicidade. Esses resultados demonstram que protocolos curativos alternativos devem ser testados em pacientes idosos portadores LDGCB a despeito da presença de comorbidades severas

Células endoteliais maduras (ECs) irão compor a parede dos vasos, enquanto células endoteliais oriundas da medula óssea (MO) migram para locais de neoformação vascular com subseqüente diferenciação em células endoteliais maduras. Processos fisiológicos e patológicos como câncer têm a angiogênese como importante componente. Estudos correlacionam a angiogênese com a agressividade tumoral em alguns tipos de tumores incluindo hematológicos, o que ressalta a importância da terapia antiangiogênica como alvo de pesquisas cada vez mais profundas. Neste artigo de revisão enfocamos o papel da angiogênese nas neoplasias hematológicas com ênfase em possíveis alvos terapêuticos.Vascular neoformation is an essential event in the development of the human body.     Deciphering the interaction of lactoferrin with V-ATPase towardsa deeper understanding of its mechanisms of action

    Lactoferrin (Lf), a bioactive milk protein, exhibits strong anticancer and antifungal activities. The search for Lf targets and mechanisms of action is of utmost importance to enhance its effective applications. A common feature among Lf-treated cancer and fungal cells is the inhibition of a proton pump called V-ATPase. Lf-driven V-ATPase inhibition leads to cytosolic acidification, ultimately causing cell death of cancer and fungal cells. Given that a detailed elucidation of how Lf and V-ATPase interact is still missing, in this work we aimed to fill this gap by employing a multilevel computational approach. Molecular dynamics (MD) simulations of both proteins were performed to obtain a robust sampling of their conformational landscape, followed by clustering and protein-protein docking. Subsequently, MD simulations of the docked complexes and free binding energy calculations were carried out to evaluate the dynamic binding process and build the final ranking. This computational pipeline allowed the unraveling of the putative mechanism by which Lf inhibits V-ATPase and the identification of key binding residues that will certainly aid in the rational design of follow-up experimental studies, bridging in this way computational and experimental biochemistry.info:eu-repo/semantics/publishedVersio

    New sol-gel-derived magnetic bioactive glass-ceramics containing superparamagnetic hematite nanocrystals for hyperthermia application

    Although the three main phases of iron oxide – hematite, maghemite, and magnetite – exhibit superparamagnetic properties at the nanoscale, only maghemite and magnetite phases have been explored in magnetic bioactive glass-ceramics aimed at applications in cancer treatment by hyperthermia. In this work, it is reported for the first time the superparamagnetic properties of hematite nanocrystals grown in a 58S bioactive glass matrix derived from sol-gel synthesis. The glass-ceramics are based on the (100-x)(58SiO2-33CaO-9P2O5)-xFe2O3 system (x = 10, 20 and 30 wt%). A thermal treatment leads to the growth of hematite (α-Fe2O3) nanocrystals, conferring superparamagnetic properties to the glass-ceramics, which is enough to produce heat under an external alternating magnetic field. Besides, the crystallization does not inhibit materials bioactivity, evidenced by the formation of calcium phosphate onto the glass-ceramic surface upon soaking in simulated body fluid. Moreover, their cytotoxicity is similar to other magnetic bioactive glass-ceramics reported in the literature. Finally, these results suggest that hematite nanocrystals' superparamagnetic properties may be explored in multifunctional glass-ceramics applied in bone cancer treatment by hyperthermia allied to bone regeneration

    Acoustics reflex abnormalities in auditory processing disorder

    Introduction: The acoustic reflex threshold is defined as the lowest sound intensity capable of starting the middle ear protection mechanism due to intense sounds (Metz, 1952; Jerger, 1970), being necessary the structural and functional integrity of the periferic and central auditory system. These structures are also responsable for the central processing of auditory information. Aim: We intend to verify whether acoustic reflex abnormalities can also appear in auditory processing (AP) disorders, resulting in symptoms related to speech and language disorders. Study design: Prospective randomized. Material and method: Data were analyzed from one hundred AP assessments, using Pereira (1997) method. Patients, male and female, with ages ranging from 07 to 18 years, had normal hearing thresholds and normal tympanograms patterns. The difference between the acoustic reflex and hearing thresholds defined the acoustic reflex level (ARL), considered normal between 70-90dB and altered when above of this range or when absent in one or more frequencies (Carvallo, 1996; 1997; Metz, 1952). Results: Disorders of AP were found in 97% of the patients. In this group, 62% showed ARL abnormalities, being statistically significant. Furthermore, patients with AP disorders showed ARL alterations, more frequently on severe degree disorders, on patients with combined auditory gnosis impairments and on patients with more than one auditory ability altered. Conclusion: Patients with acoustic reflex alterations and normal audiometry should perform the AP assessment, as these symptoms could unmask pathologies of the central nervous system.Introdução: O limiar do reflexo acústico corresponde à menor intensidade de um som capaz de desencadear o mecanismo de proteção da orelha média frente a sons intensos (Metz, 1952; Jerger, 1970), sendo necessária a integridade estrutural e funcional do sistema auditivo periférico e central em nível de tronco encefálico. Algumas dessas estruturas são também responsáveis pelo processamento central da informação auditiva. Objetivo: Procuramos verificar se alterações do reflexo acústico também estariam presentes na Desordem do Processamento Auditivo Central (DPA), levando a queixas relacionadas aos distúrbios da comunicação. Forma de estudo: Prospectivo randomizado. Material e método: Foram analisados cem protocolos de avaliações do Processamento Auditivo Central (PAC), realizadas segundo Pereira (1997), de indivíduos dos sexos masculino e feminino, de 7 a 18 anos, com limiares de audibilidade normais e timpanograma tipo A. A diferença entre os limiares do reflexo acústico e os de audibilidade forneceu os níveis de reflexos acústicos (NRA), considerados normais entre 70-90dB e alterados quando acima deste intervalo ou quando ocorreu ausência em uma ou mais freqüências (Carvallo, 1996; 1997; Metz, 1952). Resultados: Das crianças avaliadas 97% apresentaram algum tipo de DPA. Destas, 62% tiveram NRA alterados, numa relação estatisticamente significante. Além disso, pacientes com DPA mostraram NRA alterados, mais freqüentemente, nas desordens de grau severo, naquelas com prejuízos gnósicos auditivos combinados e naquelas com mais de uma habilidade auditiva alterada.     Meiotic segregation and interchromosomal effect in the sperm of a double translocation carrier: a case report

    <p>Abstract</p> <p>Background</p> <p>Infertility is a natural mechanism of selection intended to prevent the delivery of a child with malformations or mental retardation. Male infertility is closely related to chromosomal abnormalities. This study was focused on the analysis of meiotic segregation involving a Robertsonian translocation, 45,XY,der(13;13) [56]/45,XY,der(13;14) [44] and the evaluation of possible interchromosomal effects.</p> <p>Results</p> <p>Hybridisation with LSI 13q14 and subtelomere 14q probes and WCP13 SpectrumGreen and WCP14 SpectrumOrange probes showed a high proportion of unbalanced gametes, corresponding to 71.2% of the spermatozoa. The disomic frequencies of the sexual chromosomes and chromosome 18 of the patient were higher (5.28% and 2.55%, respectively) than those of the control (0.6% and 0.59%, respectively).</p> <p>Conclusion</p> <p>Meiotic segregation studies in sperm are an important tool for genetic counselling of chromosomal aberrations, allowing for a prediction of the risks and consequent implications for the reproductive life. The patient with this rare translocation exhibited meiotic segregation fidelity, and a high rate of unbalanced gametes with disomic spermatozoa.</p

    Dynein Function and Protein Clearance Changes in Tumor Cells Induced by a Kunitz-Type Molecule, Amblyomin-X

    Amblyomin-X is a Kunitz-type recombinant protein identified from the transcriptome of the salivary glands of the tick Amblyomma cajennense and has anti-coagulant and antitumoral activity. the supposed primary target of this molecule is the proteasome system. Herein, we elucidated intracellular events that are triggered by Amblyomin-X treatment in an attempt to provide new insight into how this serine protease inhibitor, acting on the proteasome, could be comparable with known proteasome inhibitors. the collective results showed aggresome formation after proteasome inhibition that appeared to occur via the non-exclusive ubiquitin pathway. Additionally, Amblyomin-X increased the expression of various chains of the molecular motor dynein in tumor cells, modulated specific ubiquitin linkage signaling and inhibited autophagy activation by modulating mTOR, LC3 and AMBRA1 with probable dynein involvement. Interestingly, one possible role for dynein in the mechanism of action of Amblyomin-X was in the apoptotic response and its crosstalk with autophagy, which involved the factor Bim; however, we observed no changes in the apoptotic response related to dynein in the experiments performed. the characteristics shared among Amblyomin-X and known proteasome inhibitors included NF-kappa B blockage and nascent polypeptide-dependent aggresome formation. Therefore, our study describes a Kunitz-type protein that acts on the proteasome to trigger distinct intracellular events compared to classic known proteasome inhibitors that are small-cell-permeable molecules. in investigating the experiments and literature on Amblyomin-X and the known proteasome inhibitors, we also found differences in the structures of the molecules, intracellular events, dynein involvement and tumor cell type effects.     Circuito de exercícios sensoriais para o treinamento do equilíbrio funcional e possibilidade de quedas em pessoas idosas

    Exercícios físicos auxiliam na melhora do equilíbrio, entretanto há carência de protocolos específicos para trabalhar preventivamente o equilíbrio funcional (EF) e a possibilidade de quedas (PQ) em idosos da comunidade. O objetivo deste estudo foi avaliar o efeito de um Circuito de exercícios sensoriais (CE) sobre o EF e a PQ em mulheres idosas. Completaram o estudo 57 idosas, divididas em grupo experimental (G1= 32; 65,61±4,72 anos) e grupo controle (G2 = 25; 68,41±4,99 anos). G1 praticou 12 semanas de CE composto por 13 estações em forma de circuito. Foram realizadas 2 aulas semanais de 50 minutos de duração, compostas por aquecimento, exercícios sensoriais e volta à calma. O grau de dificuldade foi progressivo ao longo do treinamento. EF e PQ foram avaliados antes e depois da intervenção, respectivamente pela Escala de Equilíbrio de Berg (EEB) e pelo Índice de Possibilidade de Quedas (IPQ). Utilizou-se estatística não paramétrica para análise dos dados, adotando-se em todos os testes nível de significância 5% (p≤0,05). G1 apresentou incremento significativo no EF (p = 0,001) e redução significativa na PQ (p = 0,013). Estes resultados indicam que a prática do Circuito de exercícios sensoriais é capaz de melhorar o equilíbrio e diminuir a possibilidade de quedas em idosas.Eje 5: Las prácticas de la Educación Física vinculadas con la integración e inclusión social.
