    Vitamin C and E supplementation prevents mitochondrial damage of ileum myocytes caused by intense and exhaustive exercise training

First published August 20, 2009; doi: 10.1152/japplphysiol.91166.2008.Intense and exhaustive exercise (IEE) is associated with oxidative stress in skeletal muscle, and we recently reported that intestine is sensitive to IEE. in the present study, we investigated the possible relationship between the effects of IEE on morphology and oxidative markers in the ileum and isolated mitochondria. C57BL/6 mice were ascribed either to a control group comprising two subgroups, one sedentary and another exercised for 10 days (E10), or to a corresponding supplemented control group again comprising two subgroups, one sedentary and another exercised for 10 days (E10-V). the IEE program consisted of a single daily treadmill running session at 85% of V(max), until animal exhaustion. Vitamins C (10 mg/kg) and E (10 mg/kg) were concurrently intraperitoneally administered 2 h before the exercise sessions. IEE was shown to cause 1) impairment of ileum internal membrane mitochondria verified by ultramicrography analysis; 2) increase in ileum carbonyl content (117%) and reduction in antioxidant capacity (36%); 3) increase in mitochondria carbonyl content (38%), increase in the percentage of ruptured mitochondria 25.3%), increase in superoxide dismutase activity (186%), and reduction in citrate synthase activity (40.4%) compared with control animals. Observations in the vitamin-supplemented exercised animals (E10-V) were 1) healthy appearance of myocyte mitochondria; 2) decrease in ileum carbonyl content (66%) and increase in antioxidant capacity (53%); 3) decrease in mitochondria carbonyl content (43%), decrease in the percentage of ruptured mitochondria (30%), slight increase in superoxide dismutase activity (7%), and significant increase in citrate synthase activity (121%) compared with E10 animals.     LaMMos - Latching Mechanism based on Motorized-screw for Reconfigurable Robots and Exoskeleton Suits

    Reconfigurable robots refer to a category of robots that their components (individual joints and links) can be assembled in multiple configurations and geometries. Most of existing latching mechanisms are based on physical tools such as hooks, cages or magnets, which limit the payload capacity. Therefore, robots re- quire a latching mechanism which can help to reconfigure itself without sacrificing the payload capability. This paper presents a latching mechanism based on the flexible screw attaching principle. In which, actuators are used to move the robot links and joints while connecting them with a motorized-screw and dis- connecting them by unfastening the screw. The brackets used in our mechanism configuration helps to hold maximum force up to 5000N. The LaMMos - Latching Mechanism based on Motorized- screw has been applied to the DeWaLoP - Developing Water Loss Prevention in-pipe robot. It helps the robot to shrink its body to crawl into the pipe with minimum diameter, by recon- figuring the leg positions. And it helps to recover the legs positions to original status once the robot is inside the pipe. Also, LaMMos add stiffness to the robot legs by dynamically integrate them to the structure. Additionally, we present an application of the LaMMos mechanism to exoskeleton suits, for easing the mo- tors from the joints when carrying heavy weights for long periods of time. This mechanism offers many interesting opportunities for robotics research in terms of functionality, pay- load and size.Comment: 14 pages, 15 figure

    Derivative Expansion and the Effective Action for the Abelian Chern-Simons Theory at Higher Orders

    We study systematically the higher order corrections to the parity violating part of the effective action for the Abelian Chern-Simons theory in 2+1 dimensions, using the method of derivative expansion. We explicitly calculate the parity violating parts of the quadratic, cubic and the quartic terms (in fields) of the effective action. We show that each of these actions can be summed, in principle, to all orders in the derivatives. However, such a structure is complicated and not very useful. On the other hand, at every order in the powers of the derivatives, we show that the effective action can also be summed to all orders in the fields. The resulting actions can be expressed in terms of the leading order effective action in the static limit. We prove gauge invariance, both large and small of the resulting effective actions.     It Takes Two to Tango, Part II : Synthesis of A-Ring Functionalised Quinones Containing Two Redox-Active Centres with Antitumour Activities

    In 2021, our research group published the prominent anticancer activity achieved through the successful combination of two redox centres (ortho-quinone/para-quinone or quinone/seleniumcontaining triazole) through a copper-catalyzed azide-alkyne cycloaddition (CuAAC) reaction. The combination of two naphthoquinoidal substrates towards a synergetic product was indicated, but not fully explored. Herein, we report the synthesis of 15 new quinone-based derivatives prepared from click chemistry reactions and their subsequent evaluation against nine cancer cell lines and the murine fibroblast line L929. Our strategy was based on the modification of the A-ring of paranaphthoquinones and subsequent conjugation with different ortho-quinoidal moieties. As anticipated, our study identified several compounds with IC50 values below 0.5 µM in tumour cell lines. Some of the compounds described here also exhibited an excellent selectivity index and low cytotoxicity on L929, the control cell line. The antitumour evaluation of the compounds separately and in their conjugated form proved that the activity is strongly enhanced in the derivatives containing two redox centres. Thus, our study confirms the efficiency of using A-ring functionalized para-quinones coupled with ortho-quinones to obtain a diverse range of two redox centre compounds with potential applications against cancer cell lines.     Genômica e proteômica de Anaplasma marginale: contribuições para o controle da riquétsia.

    A anaplasmose bovina é causada pela riquétsia intra-eritrocítica Anaplasma marginale, responsável por importantes prejuízos econômicos, por causa da alta morbidade e mortalidade em rebanhos bovinos suscetíveis. A vacinação tem sido uma forma econômica e eficiente de controlar a enfermidade. No entanto, os métodos de imunização tradicionais apresentam efeitos adversos em algumas categorias de animais. Nas últimas décadas, os estudos sobre imunização contra Anaplasma concentraram-se nas proteínas de superfície MSP1a, 1b, 2, 3, 4 e 5. No entanto, até o momento, os resultados foram pouco promissores, apontando a necessidade de ampliar o conhecimento sobre o rol das proteínas de membrana da riquétsia e das relações estruturais entre elas. Nesse contexto, os estudos do genoma e do proteoma da riquétsia têm contribuído com essa finalidade. Pela análise genômica, 14 genes para novas proteínas de membrana externa foram identificados (omp 1-14), dentre os quais, omp2, 3 e 6 não são transcritos. Esses genes ostraram-se altamente conservados entre isolados da riquétsia. As proteínas OMP4, 7, 10 e 14 foram reconhecidas por soros de bovinos imunizados com membrana de A. marginale, mostrando potencial para desenvolvimento de imunógenos. Além disso, mediante análise proteômica, foi possível detectar novas proteínas de membrana, negligenciadas pela anotação genômica. Dentre elas estão AM097 - conjugal transfer protein, AM956 - PepA citosol amino peptidase, AM254 - fator de elongação Tu e quatro proteínas de função desconhecida: AM127, 197, 387 e 854, as quais também foram reconhecidas por soros de bovinos imunizados com membrana de A. marginale.bitstream/CNPGC-2009-09/12410/1/DOC161.pd

    Uso de preparados homeopáticos no controle de ectoparasitas em novilhas de corte e leite

    Na produção bovina, um dos entraves mais importantes são as infestações por ectoparasitas, não apenas pelos mesmos afetarem o bem-estar dos animais, mas também por causarem perdas econômicas significativas. O objetivo desse estudo foi avaliar a utilização de preparados homeopáticos no manejo ecológico de infestações causadas por Haematobia irritans (mosca-do-chifre), Dermatobia hominis (berne) e por Boophilus microplus (carrapato-do-boi) em novilhas mantidas sob sistema de Pastoreio Racional Voisin. O experimento foi realizado na Fazenda Experimental da Universidade Federal de Santa Catarina nos meses de outubro de 2014 a fevereiro de 2015. Foi realizada a cada quinze dias, a contagem desses ectoparasitas e aplicado preparados homeopáticos de triturado destes ectoparasitas na potência CH6. Não foram encontradas diferenças significativas entre os grupos controle e tratamento nas contagens de ectoparasitas nos animais avaliados (P>0,05), porém, houve uma diminuição na população de bernes e carrapatos.In beef production, one of the most important barriers are infestations by ectoparasites, not only the same effect upon the welfare of animals but also to cause significant economic losses. The aim of this study was to evaluate the use of homeopathic preparations in ecological management of infestations caused by Haematobia irritans (fly-do-horn), Dermatobia hominis (warble) and Boophilus microplus (cattle tick) in heifers kept in system Pastoreio Racional Voisin. The experiment was conducted at the Experimental Farm of the Federal University of Santa Catarina from October 2014 to February 2015. It was held every fortnight, the count of these ectoparasites and applied prepared ground of homeopathic these ectoparasites in CH6 power.
