    Analysis of Multidrug Transporter in Living Cells. Inhibition of P-glycoprotein-mediated Efflux of Anthracyclines by Ionophores

    One of the major obstacles of chemotherapy is that, after repeated treatments, cellular resistance to the drug appears. The problem is that the tumor cells become resistant not only to the drugs which have been used during the treatment but also to other drugs which are structurally and functionally unrelated. This is termed ‘multidrug resistance’ (MDR). MDR is frequently associated with decreased drug accumulation resulting from enhanced drug efflux. This is correlated with the presence of a membrane protein, P-glycoprotein, which pumps a wide variety of drugs out of cells thus reducing their toxicity. The search for molecules able to reverse MDR is very important. We here report that mobile ionophores such as valinomycin, nonactin, nigericin, monensin, calcimycin, lasalocid inhibit the efflux of anthracycline by P-glycoprotein whereas, channel-forming ionophores such as gramicidin do not. Cyclosporin which is also a strong Ca2+ chelating agent also inhibits the P-glycoprotein-mediated efflux of anthracycline

    A New Complex of Palladium(II) With 2-Furoic Hydrazide: Synthesis, Characterization, Theoretical Calculations and Biological Studies

    A new complex of palladium was isolated with 2-furoic hydrazide (FH) and characterized by spectroscopic methods. The results show that the ligand is coordinated to palladium by the basic nitrogen of NH2 group and has a general structure of type cis-[Pd(FH)2Cl2]. The structure of palladium(II) complex was optimized and theoretical data show good agreement with the experimental results. The cytotoxic activity was evaluated in a chronic myelogenous leukemia cell line, which revealed that the compound is less active than cisplatin or carboplatin. At 300 g mL−1, the complex presented antimicrobial activity more efficient than ampicillin, chloranfenicol and kanamicyn. (doi: 10.5562/cca2151

    Introdução. O diabetes mellitus é uma das principais doenças crônicas que acomete a população, principalmente os idosos. Dentre as suas principais complicações está o pé diabético. Objetivo. Conhecer o perfil de pacientes idosos diabéticos com úlcera de pé, internados em um hospital de emergência de São Luís-MA. Métodos. Trata-se de um estudo descritivo, exploratório e quantitativo. Para a coleta de dados utilizou-se um questionário com perguntas abertas e fechadas referentes ao tempo de doença, conhecimento sobre a doença, tratamento, comorbidades e dados socioeconômicos. A amostra foi constituída por 20 idosos diabéticos, com idade entre 71 a 80 anos. Resultados. Observou-se que 50% eram do sexo masculino, 55% não sabiam ler e escrever, 50% percebiam renda de até 1 salário mínimo, 35% tinham menos de 5 anos de diagnóstico da doença. Sobre o conhecimento da doença, 60% tinham pouca informação e os fatores de risco para formação de pé diabético foram: pele seca, unhas grossas, rachaduras e ranhuras. De acordo com a escala de Wagner as feridas mais frequentes foram classificadas nos graus II e IV. Quanto aos hábitos de vida, 50% eram ex-fumantes, 55% ex-usuários de álcool, 95% eram sedentários e 60% não faziam dieta. A hipertensão arterial foi a comorbidade mais frequente. Conclusão. Os idosos com úlceras de pé possuíam pouco poder aquisitivo e baixa escolaridade além de pouco conhecimento e orientação sobre a doença, o que pode contribuir significativamente para o aparecimento de complicações como as úlceras de pé

    Tetracyclines exhibits activity to a broad range of Gram-negative and Gram-positive bacteria and this fact allied to the low toxicity, low cost, and the advantage of administration by oral route led to their indiscriminate use, which caused the appearance of bacterial resistance to these agents, wich has restricted its clinical utility, though new applications have emerged. On the other hand, the glycylcyclines, semi-synthetic products are similar to tetracyclines, which are active against many bacteria resistant to tetracycline and other classes of antibiotics. The purpose of this paper is to give an overview of this important class of antibiotics focusing on its coordination chemistry and possible applications

    A novel oxidovanadium(V) compound with an isonicotinohydrazide ligand: A combined experimental and theoretical study and cytotoxity against K562 cells

    The interaction of oxidovanadium(V) with INHOVA (the condensation product of isoniazid and o-vanillin) lead to the formation of the ester-like complex [VO(INHOVA)EtO(OH2)]Cl·H2O (1). Crystals suitable for X-ray diffraction methods were obtained. The complex crystallizes as a dimer in the space group P21/c of the monoclinic system. A detailed analysis, including solid-state vibrational spectroscopy and electronic spectroscopy in DMSO solution, was performed for both INHOVA and complex (1). A complete theoretical study based on DFT was also carried out. The calculations were of valuable assistance in the spectra assignments and interpretation. The electrochemical characterization allows determining the redox behavior of INHOVA and complex (1). Cytotoxicity was assayed against the chronic myelogenous leukemia K562 cell line. The IC50 values obtained denote that both the ligand and complex (1) are good candidates for further studies.Centro de Química Inorgánic

    Platinum and palladium complexes with 2,4-Dinitrophenylhydrazine: synthesis and cytotoxic activity

    New complexes of platinum and palladium were isolated with 2,4-dinitrophenylhydrazine (2,4-DNPH). These complexes were characterized and the results shown that the ligand is coordinated to platinum or palladium by the basic nitrogen of NH2 group and have the general structure cis-[M(2,4- DNPH)2Cl2 ].H2O where M = Pt or Pd. The thermal stability of the complexes was followed in the temperature range 25-850 ºC. The antitumor activity of the synthesized compounds has been studied and the compound cis-[Pt(2,4-DNPH)2Cl2 ], was found to display cytotoxicity (IC50 = 4.6 μmol/L) against K562 tumoral cell line.     A destinação dos Resíduos Sólidos de Serviços de Saúde (RSS) sem conformidade com a legislação ocasiona danos ao meio ambiente e à saúde humana.  Diante disso, este estudo buscou analisar o gerenciamento dos RSS, da Unidade de Pronto Atendimento – UPA, localizada no bairro Santa Mônica, município de Tucuruí-Pará. Verificando concordância com os procedimentos implementados no local para com as exigências da Resolução da Diretoria Colegiada (RDC), Nº 222, de 28 de março de 2018. E também, realizou-se uma análise da percepção dos profissionais de saúde da unidade, quanto ao manejo dos RSS e suas adequações, afim de proporcionar compreensão dos seus riscos e propor melhorias diante aos resultados encontrados
