Time-of-flight secondary ion mass spectrometry (TOF-SIMS) with the Bi3+_{3}^{+} liquid metal ion gun was used to investigate the content of lipids and amino acids (AAs) in extracellular vesicles (EVs). We induced metabolic changes in human pancreatic β-cells by stimulation with high glucose concentrations (35 mM) and tested the hypothesis of hyperglycemia (HG) has a detrimental effect on lipids and AAs in released EV subpopulations: ectosomes and exosomes. As a result of HG treatment, selected fatty acids (FAs) such as arachidonic, myristic and palmitic acids, changed their abundance in ectosomes and exosomes. Also, intensities of the characteristic peaks for cholesterol (m/z 95.09; 147.07; 161.11; 369.45) along with the molecular ion m/z 386.37 [C27_{27}H46_{46}O+^{+}] under HG conditions, both for ectosomes and exosomes, have changed significantly. Comparative analysis of HG EVs and normoglycemic (NG) ones showed statistically significant differences in the signal intensities of four AAs: valine (m/z 72.08 and 83.05), isoleucine (m/z 86.10), phenylalanine (m/z 120.08 and 132.05) and tyrosine (m/z 107.05 and 136.09). We confirmed that ToF-SIMS is a useful technique to study selected AAs and lipid profiles in various EV subpopulations. Our study is the first demonstration of changes in FAs and AAs in exosomes and ectosomes derived from β-cells under the influence of HG

The North European Platform suture zone in Poland

The authors interpret the structure of the Central Carpathian-North European plates suture zone in Poland, where three main Carpathian tectonic units: the Central Carpathian, Pieniny Klippen Belt (PKB) and Outer Carpathian are present. In general, the PKB follows this zone. Several deep bore-holes were drilled in this region and the seismic lines were tied to bore-hole data and geological maps. The Polish PKB belongs to the complex geological structure stretching from Vienna in Austria to Romania. The rocks included in the PKB tectonic components were deposited within the paleogeographic realm known as the Alpine Tethys, mainly during the Jurassic-Early Cretaceous times. Both strike-slip and thrust components occur within the Polish section of the PKB. The strongly tectonized, few kilometer wide PKB zone is limited by a flower structure marked by two major faults, linked to the strike-slip zone. These faults reach the North European Platform (part of the North European Plate). The flysch sequences, arranged into a series of north-vergent thrust-sheets, constitute the main component of the PKB in the survey zone. They contain olistoliths, which are mainly Jurassic-Early Cretaceous in age. The PKB tectonic components of different age, strike-slip, thrust as well as toe-thrusts and olistostromes are mixed together, giving the present-day mélange character of this belt, where individual units are hard to distinguish. Two olistostrome belts (mélange units) exist within the PKB structure. The seismic lines show the Central Carpathian Paleogene rocks covering the Paleozoic Central Carpathian Basement south of the PKB. The Subtatric covers the High-Tatric autochthonic and allochthone rocks. The Central Carpathian Plate is thrust over the North European Platform in the Podhale region. The allochthonous Outer Carpathians consist of several nappes (thrust-sheets) verging northward. They are thrust over each other and over the North European Platform which dips gently southward

SELECTED ASPECTS OF THE USE OF GASEOUS FUELS BLENDS TO IMPROVE EFFICIENCY AND EMISSION OF SI ENGINE

    Get PDF
The paper presents results of SI engine tests, carried on for different gaseous fuels. The analysis carried out made it possible to define the correlation between fuel composition and engine operating parameters. Tests cover various gaseous mixtures: methane with hydrogen and LPG with DME. The first group, considered as low carbon content fuels, can be characterized by low CO2 emissions. The flammability of hydrogen added in those mixtures realizes the function of combustion process activator and improves the energy conversion. The second group of fuels is constituted by LPG and DME mixtures. DME mixes perfectly with LPG and differently than in the case of other hydrocarbon fuels also consisting of oxygen, which makes the stoichiometric mixture less oxygen demanding. In the case of this fuel, improvement in engine volumetric and overall engine efficiency has been noticed, compared to LPG. During the tests, standard CNG/LPG feeding systems have been used, which underlines the utility value of the research

The use of seismostratigraphy for exploration of Miocene gas-bearing reservoir facies in the NE part of the Carpathian Foreland Basin (Poland)

    Get PDF
Sedimentological and seismostratigraphic interpretation of a dip cross-section located in the northwestern part of the Wielkie Oczy Graben, across the Markowice - Lubliniec elevation and farther to the NE resulted in the identification of the base-Sarmatian angular unconformity (UNO) and allowed to subdivide the Machów Formation into seven genetic sequences composed mainly of deltaic deposits. The unconformity reflects extension episode superimposed on regional south-westward rotation of the basin floor and is overstepped and upplapped towards the NE by deltaic bodies fed from an southerly (orogenic) source. The upplaping pinchouts may form combined, structural-stratigraphic traps for methane generated in delta front-prodelta heteroliths. Unconformity UNO is farther to the SE overlain locally by gas-bearing massive sandstones of intrabasinal or northerly provenience, and similar sandstones may be expected to occur in the study area below the belt of upplapping pinch-outs

The establishment of new businesses as a seasonal phenomenon : a Polish example

PURPOSE: The aim of the article is to analyse the phenomenon of seasonality in the context of registration of new economic entities in the form of a sole proprietorship based on the data from the Central Registration and Information on Business (CEIDG) in the years 2012-2021.DESIGN/METHODOLOGY/APPROACH: The research method and tools applied in the article are subordinated to the identification of the occurence of statistically significant differences between average values of numbers of applications filed with CEIDG in the assumption category on a month-to-month basis. As part of the research process post hoc tests, preceded by the one-way analysis of variance, ANOVA, were used. Due to the imperfection of statistical tests and the nature of deterministic methods for the verification of the research hypothesis the Kruskal–Wallis was also used. The ANOVA was preceded by the verification of meeting the distribution normality criterion (by means of the Kolmogorowv-Smirnov test and the Shapiro-Wilk test) and of the variance uniformity (the Levine test).FINDINGS: A stable economic situation in a macroeconomic aspect together with an unchanging regulatory, economic and financial, technical and organisational as well as market environment translates to a statistical repeatability of market behaviours. In this context, a projection of the number of applications filed with CEIDG including seasonal fluctuations was developed.PRACTICAL IMPLICATIONS: The knowledge of seasonality in terms of undertaking economic activity may prove helpful in selecting the right economic policy tools, in organising entrepreneur administrative service processes as well as useful for companies providing outsourcing services (inter alia, within the scope of marketing, bookkeeping and human resources service or recapitalisation) for new businesses.ORIGINALITY/VALUE: This type of analyses have not been conducted to date on such a wide and diversified time spectre. In addition, the situation related to the COVID-19 pandemic, and to be exact, the observation of correlation between future decisions of entrepreneurs with respect to continuing or establishing business activity and a specified seasonality also seems extremely significant.peer-reviewe

Potential environmental life cycle impacts of fuel cell electric vehicles powered by hydrogen produced from polish coke oven gas

    Get PDF
This study analysed the greenhouse gas (GHG) emissions of hydrogen fuel cell vehicles'(FCEVs') life cycles. These included models running on hydrogen derived from coke oven gas (COG), which is a by-product of the coking process of coal and includes hydrogen, methane, and other gases. FCEVs and hydrogen have the potential to drive future mobility. Hydrogen can be separated from the COG in the process of pressure swing adsorption to obtain a purity of hydrogen that meets the requirements of a hydrogen FCEV. An environmental life cycle assessment (LCA) of FCEV powered by hydrogen produced from Polish COG was conducted. The direction of hydrogen production strategies in Poland was also presented. The analyses included the entire life cycle of FCEVs with the production of hydrogen from COG in a Polish coke plant. A comparative analysis of FCEVs and other alternative fuels was conducted, and the main determinants of GHG emissions of FCEV were given. Importantly, this is the first attempt at an environmental assessment of FCEVs in Poland.Web of Science17116115
