    Composition and properties of bovine colostrum: a review

    International audienceAbstractColostrum is the initial milk secreted by mammals following parturition, the composition and physicochemical properties of which are highly dynamic and variable. The composition and physicochemical properties of colostrum during the initial post-partum period has not been systematically reviewed for many years, although the topic remains of interest both to milk producers and processors. In this article, the current understanding of the composition of colostrum, i.e. carbohydrates, proteins, growth factors, enzymes, enzyme inhibitors, nucleotides and nucleosides, cytokines, fats, vitamins and minerals, is reviewed. In addition, the physicochemical properties, i.e. pH and buffering capacity, colour, density and specific gravity, osmotic pressure, somatic cell count, properties of casein micelles, ethanol stability and rennet coagulation properties are discussed, as well as the effects of heat-treating colostrum

    Identifying Otosclerosis with Aural Acoustical Tests of Absorbance, Group Delay, Acoustic Reflex Threshold, and Otoacoustic Emissions

    Background: Otosclerosis is a progressive middle-ear disease that affects conductive transmission through the middle ear. Ear-canal acoustic tests may be useful in the diagnosis of conductive disorders. This study addressed the degree to which results from a battery of ear-canal tests, which include wideband reflectance, acoustic stapedius muscle reflex threshold (ASRT), and transient evoked otoacoustic emissions (TEOAEs), were effective in quantifying a risk of otosclerosis and in evaluating middle-ear function in ears after surgical intervention for otosclerosis. Purpose: To evaluate the ability of the test battery to classify ears as normal or otosclerotic, measure the accuracy of reflectance in classifying ears as normal or otosclerotic, and evaluate the similarity of responses in normal ears compared with ears after surgical intervention for otosclerosis. Research Design: A quasi-experimental cross-sectional study incorporating case control was used. Three groups were studied: one diagnosed with otosclerosis before corrective surgery, a group that received corrective surgery for otosclerosis, and a control group. Study Sample: The test groups included 23 ears (13 right and 10 left) with normal hearing from 16 participants (4 male and 12 female), 12 ears (7 right and 5 left) diagnosed with otosclerosis from 9 participants (3 male and 6 female), and 13 ears (4 right and 9 left) after surgical intervention from 10 participants (2 male and 8 female). Data Collection and Analysis: Participants received audiometric evaluations and clinical immittance testing. Experimental tests performed included ASRT tests with wideband reference signal (0.25–8 kHz), reflectance tests (0.25–8 kHz), which were parameterized by absorbance and group delay at ambient pressure and at swept tympanometric pressures, and TEOAE tests using chirp stimuli (1–8 kHz). ASRTs were measured in ipsilateral and contralateral conditions using tonal and broadband noise activators. Experimental ASRT tests were based on the difference in wideband-absorbed sound power before and after presenting the activator. Diagnostic accuracy to classify ears as otosclerotic or normal was quantified by the area under the receiver operating characteristic curve (AUC) for univariate and multivariate reflectance tests. The multivariate predictor used a small number of input reflectance variables, each having a large AUC, in a principal components analysis to create independent variables and followed by a logistic regression procedure to classify the test ears. Results:Relative to the results in normal ears, diagnosed otosclerosis earsmore frequently showed absent TEOAEs and ASRTs, reduced ambient absorbance at 4 kHz, and a different pattern of tympanometric absorbance and group delay (absorbance increased at 2.8 kHz at the positive-pressure tail and decreased at 0.7–1 kHz at the peak pressure, whereas group delay decreased at positive and negative-pressure tails from 0.35–0.7 kHz, and at 2.8–4 kHz at positive-pressure tail). Using a multivariate predictor with three reflectance variables, tympanometric reflectance (AUC 5 0.95) was more accurate than ambient reflectance (AUC 5 0.88) in classifying ears as normal or otosclerotic. Conclusions: Reflectance provides a middle-ear test that is sensitive to classifying ears as otosclerotic or normal, which may be useful in clinical applications

    Radiation-induced Assembly of Rad51 and Rad52 Recombination Complex Requires ATM and c-Abl

    Cells from individuals with the recessive cancer-prone disorder ataxia telangiectasia (A-T) are hypersensitive to ionizing radiation (I-R). ATM (mutated in A-T) is a protein kinase whose activity is stimulated by I-R. c-Abl, a nonreceptor tyrosine kinase, interacts with ATM and is activated by ATM following I-R. Rad51 is a homologue of bacterial RecA protein required for DNA recombination and repair. Here we demonstrate that there is an I-R-induced Rad51 tyrosine phosphorylation, and this induction is dependent on both ATM and c-Abl. ATM, c-Abl, and Rad51 can be co-immunoprecipitated from cell extracts. Consistent with the physical interaction, c-Abl phosphorylates Rad51 in vitro and in vivo. In assays using purified components, phosphorylation of Rad51 by c-Abl enhances complex formation between Rad51 and Rad52, which cooperates with Rad51 in recombination and repair. After I-R, an increase in association between Rad51 and Rad52 occurs in wild-type cells but not in cells with mutations that compromise ATM or c-Abl. Our data suggest signaling mediated through ATM, and c-Abl is required for the correct post-translational modification of Rad51, which is critical for the assembly of Rad51 repair protein complex following I-R

    Assessing Threats and Conservation Status of Historical Centers of Oak Richness in California

    Oak trees are emblematic of California landscapes, they serve as keystone cultural and ecological species and as indicators of natural biological diversity. As historically undeveloped landscapes are increasingly converted to urban environments, endemic oak woodland extent is reduced, which underscores the importance of strategic placement and reintroduction of oaks and woodland landscape for the maintenance of biodiversity and reduction of habitat fragmentation. This paper investigated the effects of human urban development on oak species in California by first modeling historical patterns of richness for eight oak tree species using historical map and plot data from the California Vegetation Type Mapping (VTM) collection. We then examined spatial intersections between hot spots of historical oak richness and modern urban and conservation lands and found that impacts from development and conservation vary by both species and richness. Our findings suggest that the impact of urban development on oaks has been small within the areas of highest oak richness but that areas of highest oak richness are also poorly conserved. Third, we argue that current policy measures are inadequate to conserve oak woodlands and suggest regions to prioritize acquisition of conservation lands as well as examine urban regions where historic centers of oak richness were lost as potential frontiers for oak reintroduction. We argue that urban planning could benefit from the adoption of historical data and modern species distribution modelling techniques primarily used in natural resources and conservation fields to better locate hot spots of species richness, understand where habitats and species have been lost historically and use this evidence as incentive to recover what was lost and preserve what still exists. This adoption of historical data and modern techniques would then serve as a paradigm shift in the way Urban Planners recognize, quantify, and use landscape history in modern built environments

    Cognitive representations of disability behaviours in people with mobility limitations : consistency with theoretical constructs

    Disability is conceptualised as behaviour by psychological theory and as a result of bodily impairment by medical models. However, how people with disabilities conceptualise those disabilities is unclear. The purpose of this study was to examine disability representations in people with mobility disabilities. Thirteen people with mobility disabilities completed personal repertory grids (using the method of triads) applied to activities used to measure disabilities. Ten judges with expertise in health psychology then examined the correspondence between the elicited disability constructs and psychological and medical models of disability. Participants with mobility disabilities generated 73 personal constructs ofdisability. These constructs were judged consistent with the content of two psychological models, namely the theory of planned behaviour and social cognitive theory and with the main medical model of disability, the International Classification of Functioning Disability and Health.Individuals with activity limitations conceptualise activities in a manner that is compatible with both psychological and medical models. This ensures adequate communication in contexts where the medical model is relevant, e.g. clinical contexts, as well as in everyday conversation about activities and behaviours. Finally, integrated models of disability may be of value for theory driven interdisciplinary approaches to disability and rehabilitation

    Characterizing Fishing Effort and Spatial Extent of Coastal Fisheries

    Biodiverse coastal zones are often areas of intense fishing pressure due to the high relative density of fishing capacity in these nearshore regions. Although overcapacity is one of the central challenges to fisheries sustainability in coastal zones, accurate estimates of fishing pressure in coastal zones are limited, hampering the assessment of the direct and collateral impacts (e.g., habitat degradation, bycatch) of fishing. We compiled a comprehensive database of fishing effort metrics and the corresponding spatial limits of fisheries and used a spatial analysis program (FEET) to map fishing effort density (measured as boat-meters per km2) in the coastal zones of six ocean regions. We also considered the utility of a number of socioeconomic variables as indicators of fishing pressure at the national level; fishing density increased as a function of population size and decreased as a function of coastline length. Our mapping exercise points to intra and interregional ‘hotspots’ of coastal fishing pressure. The significant and intuitive relationships we found between fishing density and population size and coastline length may help with coarse regional characterizations of fishing pressure. However, spatially-delimited fishing effort data are needed to accurately map fishing hotspots, i.e., areas of intense fishing activity. We suggest that estimates of fishing effort, not just target catch or yield, serve as a necessary measure of fishing activity, which is a key link to evaluating sustainability and environmental impacts of coastal fisheries

    Characteristics of Mantle Fabrics beneath the South-Central United States: Constraints from Shear-Wave Splitting Measurements

    New shear-wave splitting measurements at permanent broadband seismic stations in the south-central United States reveal the orientation and degree of polarization of mantle fabrics, and provide constraints on models for the formation of these fabrics. For stations on the stable North American craton, correspondence between observed polarization direction of the fast wave and the trend of Proterozoic and Paleozoic structures associated with rifts and orogenic belts implies a lithospheric origin for the observed anisotropy. The largest splitting times (up to 1.6 s) are observed at stations located in the ocean-continent transition zone, in which the fast directions are parallel to the Gulf of Mexico continental margin. The parallelism and the geometry of the keel of the craton beneath the study area suggest that asthenospheric flow around the keel of the North American craton, lithospheric fabrics developed during Mesozoic rifting, or a combination of these factors are responsible for the observed anisotropy on stations above the transitional crust

    Passive Rifting of Thick Lithosphere in the Southern East African Rift: Evidence from Mantle Transition Zone Discontinuity Topography

    To investigate the mechanisms for the initiation and early-stage evolution of the nonvolcanic southernmost segments of the East African Rift System (EARS), we installed and operated 35 broadband seismic stations across the Malawi and Luangwa rift zones over a 2 year period from mid-2012 to mid-2014. Stacking of over 1900 high-quality receiver functions provides the first regional-scale image of the 410 and 660 km seismic discontinuities bounding the mantle transition zone (MTZ) within the vicinity of the rift zones. When a 1-D standard Earth model is used for time-depth conversion, a normal MTZ thickness of 250 km is found beneath most of the study area. In addition, the apparent depths of both discontinuities are shallower than normal with a maximum apparent uplift of 20 km, suggesting widespread upper mantle high-velocity anomalies. These findings suggest that it is unlikely for a low-velocity province to reside within the upper mantle or MTZ beneath the nonvolcanic southern EARS. They also support the existence of relatively thick and strong lithosphere corresponding to the widest section of the Malawi rift zone, an observation that is consistent with strain localization models and fault polarity and geometry observations. We postulate that the Malawi rift is driven primarily by passive extension within the lithosphere attributed to the divergent rotation of the Rovuma microplate relative to the Nubian plate, and that contributions of thermal upwelling from the lower mantle are insignificant in the initiation and early-stage development of rift zones in southern Africa
