    Design and development of an AGV using RF module

    Automated guided vehicles play an important role in the flexible manufacturing system. The project mainly deals with the design and development of an AGV and controlling it with computer through RF signals. This includes several steps • Design of microcontroller on robot. Here ATMEGA 16 (AVR series of microcontroller) is used as it is enabled with all sorts of required features. • Design of driving systems. Stepper motors are used. • Design of communication systems. RF modules are to be used to communicate data between computer and AGV. Driving system Stepper motors are used to drive the AGV. They are controlled by ATMEGA 16 microcontroller and a driver chip to amplify the current to the motor. Communication The details of position and feedback should be sent to the central computer and instructions received to proceed to do further tasks. The communication has to be wireless to make things easier. Available RF modules are to be used which are serial in nature. But before that, the parallel data has to be converted into the serial data

    Concomitant yield optimization of tannase and gallic acid by Bacillus licheniformis KBR6 through submerged fermentation : an industrial approach

    The present study is concerned with the evaluation of tannase and gallic acid production effi cacy of Bacillus licheniformis KBR6 under diff erent environmental conditions through submerged fermentation. Results have shown that diff erent environmental conditions and mineral sources have diff erential infl uences on tannase and gallic acid production. Highest tannase and gallic acid yield was observed at incubation period of 18 h and 22 h, respectively. At tannic acid concentration of 15 g/l, maximum cell mass (0.75 g/l), cell yield coeffi cient (0.08 g/g), specifi c growth rate (37.5 mg/g/h), tannase yield (16.3 U/g) and specifi c tannase production rate (0.80 U/g/h) were observed, however, at higher tannic acid concentration a decrease in tannase yield and production rate were observed, but gallic acid production increased with increasing tannic acid concentration. Additional carbohydrate sources like glucose, fructose, and lactose showed positive infl uence on enzyme yield. Among the studied nitrogen sources urea and NH4Cl, and of the phosphate sources KH2PO4 showed favourable eff ects on cell growth and simultaneous enzyme and gallic acid production. Temperature of 35 °C was found to be optimum for tannase and gallic acid production. Of all the studied metal ions Ca2+, Mg2+ and Na+ showed positive eff ect whereas, Co2+, Ag2+, Pb2+, Hg2+ showed inhibitory eff ects

    Parametric optimalization of submerged fermantation conditions for xylanase production Bacillus cereus BSA1 through Taguchi Methodology

    Extracellular xylanase production of Bacillus cereus BSA1 was optimized under submerged fermentation using Taguchi orthogonal array (OA). An L18 layout of OA was constructed at three-levels of six factors, i.e. temperature, pH, and xylan, Na2HPO4, NH4NO3 and NaCl concentrations, influencing the xylanase synthesis. The enzyme production was studied in 18 parallel batch systems using different levels of each factor. The results were processed with Qualitek-4 software using ‘bigger is better’ quality character, and combination of 35 oC; pH 6.0; and xylan 0.5; NH4NO3 0.5, Na2HPO4, 0.1; NaCl 0.05 concentrations (in w/v %) with a predictive xylanase production of 7.404 U/ml was obtained. Fermentation experiment was performed for further validating the statistical output, and it resulted 10.24% in the xylanase yield (from 6.44 U/ml to 7.10 U/ml) as compared to one variable at a time (OVAT) design.     Ethnic Preparation of Haria, a Rice-Based Fermented Beverage, in the Province of Lateritic West Bengal, India

    Haria is a rice-based fermented beverage that is popular among tribal and low income people in lateritic West Bengal and East-Central India. The principal ingredient of this beverage is low grade boiled rice (Oryza sativa L.), which is mixed with a traditional starter, called bakhar, and fermented within a heat-sterilized earthen pot for 3-4 days. The main aim of this study was to investigate the ethnobotanical importance and traditional process of haria preparation. The method adopted for this study was based on interactive questionnaires and laboratory experiments. It was found that the pH decreased during the course of fermentation with increased titratable acidity of 1.42%. The alcohol content was 2-3% (v/v) in the consumable beverages. This documentation will be useful for further exploitation of haria as a health drink

    MBL2 variations and malaria susceptibility in Indian populations

    Human Mannose-binding Lectin (MBL) encoded by the MBL2 gene is a pattern recognition protein and has been associated with many infectious diseases, including malaria. We sought to investigate the contribution of functional MBL2 gene variations to Plasmodium falciparum malaria in well-defined cases and in matched controls. We resequenced the 8.7 kb of the entire MBL2 gene in 434 individuals clinically classified with malaria from regions of India where malaria is endemic. The study cohort included 176 patients with severe malaria, 101 patients with mild malaria, and 157 ethnically matched asymptomatic individuals. In addition, 830 individuals from 32 socially, linguistically, and geographically diverse endogamous populations of India were investigated for the distribution of functional MBL2 variants. The MBL2 −221C (X) allelic variant is associated with increased risk of malaria (mild malaria odds ratio [OR] = 1.9, corrected P value [PCorr] = 0.0036; severe malaria OR = 1.6, PCorr = 0.02). The exon1 variants MBL2*B (severe malaria OR = 2.1, PCorr = 0.036; mild versus severe malaria OR = 2.5, PCorr = 0.039) and MBL2*C (mild versus severe malaria OR = 5.4, PCorr = 0.045) increased the odds of having malaria. The exon1 MBL2*D/*B/*C variant increased the risk for severe malaria (OR = 3.4, PCorr = 0.000045). The frequencies of low MBL haplotypes were significantly higher in severe malaria (14.2%) compared to mild malaria (7.9%) and asymptomatic (3.8%). The MBL2*LYPA haplotypes confer protection, whereas MBL2*LXPA increases the malaria risk. Our findings in Indian populations demonstrate that MBL2 functional variants are strongly associated with malaria and infection severity

    Characterization of an acidophilic α-amylase from Aspergillus niger RBP7 and study of catalytic potential in response to nutritionally important heterogeneous compound

    An acidophilic α-amylase from Aspergillus niger RBP7 was purified after solid state fermentation on potato peel substrate. Molecular mass of the purified α-amylase was 37.5 kDa and it exhibited 1.4 mg/ml and 0.992 μ/mol/min Km and Vmax values, respectively. The enzyme was stable in the pH range from 2.0 to 6.0, at high NaCl concentration (3 M) and at temperatures between 40 °C and 70 °C. The enzyme showed an optimal activity at pH 3.0 and at 45 °C. The enzyme was inhibited by Hg2+ and was stable in the presence of different surfactants (Tween 60, Tween 80, and SDS at 1% level) and different inhibitory reagents (β-mercaptoethanol, phenylmethylsulfonyl fluoride, and sodium azide). This acidophilic amylase enzyme can digest heterogeneous food materials, i.e. the mixture of rice, fish, bread and curry with comparable activity to the commercial diastase enzymes available

    The development and validation of a scoring tool to predict the operative duration of elective laparoscopic cholecystectomy

    Background: The ability to accurately predict operative duration has the potential to optimise theatre efficiency and utilisation, thus reducing costs and increasing staff and patient satisfaction. With laparoscopic cholecystectomy being one of the most commonly performed procedures worldwide, a tool to predict operative duration could be extremely beneficial to healthcare organisations. Methods: Data collected from the CholeS study on patients undergoing cholecystectomy in UK and Irish hospitals between 04/2014 and 05/2014 were used to study operative duration. A multivariable binary logistic regression model was produced in order to identify significant independent predictors of long (> 90 min) operations. The resulting model was converted to a risk score, which was subsequently validated on second cohort of patients using ROC curves. Results: After exclusions, data were available for 7227 patients in the derivation (CholeS) cohort. The median operative duration was 60 min (interquartile range 45–85), with 17.7% of operations lasting longer than 90 min. Ten factors were found to be significant independent predictors of operative durations > 90 min, including ASA, age, previous surgical admissions, BMI, gallbladder wall thickness and CBD diameter. A risk score was then produced from these factors, and applied to a cohort of 2405 patients from a tertiary centre for external validation. This returned an area under the ROC curve of 0.708 (SE = 0.013, p  90 min increasing more than eightfold from 5.1 to 41.8% in the extremes of the score. Conclusion: The scoring tool produced in this study was found to be significantly predictive of long operative durations on validation in an external cohort. As such, the tool may have the potential to enable organisations to better organise theatre lists and deliver greater efficiencies in care

    Multiple novel prostate cancer susceptibility signals identified by fine-mapping of known risk loci among Europeans

    Genome-wide association studies (GWAS) have identified numerous common prostate cancer (PrCa) susceptibility loci. We have fine-mapped 64 GWAS regions known at the conclusion of the iCOGS study using large-scale genotyping and imputation in 25 723 PrCa cases and 26 274 controls of European ancestry. We detected evidence for multiple independent signals at 16 regions, 12 of which contained additional newly identified significant associations. A single signal comprising a spectrum of correlated variation was observed at 39 regions; 35 of which are now described by a novel more significantly associated lead SNP, while the originally reported variant remained as the lead SNP only in 4 regions. We also confirmed two association signals in Europeans that had been previously reported only in East-Asian GWAS. Based on statistical evidence and linkage disequilibrium (LD) structure, we have curated and narrowed down the list of the most likely candidate causal variants for each region. Functional annotation using data from ENCODE filtered for PrCa cell lines and eQTL analysis demonstrated significant enrichment for overlap with bio-features within this set. By incorporating the novel risk variants identified here alongside the refined data for existing association signals, we estimate that these loci now explain ∼38.9% of the familial relative risk of PrCa, an 8.9% improvement over the previously reported GWAS tag SNPs. This suggests that a significant fraction of the heritability of PrCa may have been hidden during the discovery phase of GWAS, in particular due to the presence of multiple independent signals within the same regio
