    Concentrate levels of crossbred bulls slaughtered at 16 or 22 months: Performance and carcass characteristics

    This study was performed to assess the animal performance of 97 bulls, 1/2 Puruna~ vs. 1/2 Canchim, that were slaughtered at 16 (16M) or 22 (22M) months with three concentrate levels (0.8, 1.2, and 1.6%) of body weight (BW). The initial body weight was lower for the 16M bulls. The final body weight and hot carcass weights were similar between the two slaughter ages. The hot carcass dressing was higher for 22M bulls. The average daily gain was higher for 16M bulls. The feed intake and dry matter feed conversion were similar between 16 and 22M bulls. The carcass length, marbling, and bone percentage were lower for 16M bulls. However, the Longissimus dorsi muscle area was higher for 16M bulls. The leg length, cushion thickness, fat thickness, conformation, colour, texture, muscle percent, and fat percent were similar between 16 and 22M bulls. The final and hot carcass weights were lower for bulls that were fed with 0.8% of BW. The carcass dressing was similar for the three concentrate levels. The average daily gain was higher for bulls that were fed with 1.6% of BW of concentrate and lower for those that were fed with 0.8%. The concentrate levels had no effect on carcass characteristics, feed intake, and dry matter conversion

    Number of consumers and days of display necessary for the assessment of meat colour acceptability

    Visual assessment is regarded as the gold standard to evaluate meat colour shelf-life, but it is costly and time consuming. To address this issue, this paper aims to evaluate the number of consumers and days of display that are necessaries in order to assess the colour shelf-life of meat, presented with different methods, all using images. Photographs of thirty-six lamb steaks were taken just after cutting (day 0) and on each of the following days until the 14th day of display under standardized conditions. Images were presented in three different manners: 1) with days of display and animals in random order (Random); 2) days of display in sequential and animals in random order (Sequential); and, 3) days of display and animals in sequential order (Animal); they were presented to 211 consumers who evaluated visual acceptability on a 9-point scale. At day zero, visual acceptability scores were the highest in Animal, followed by Sequential, and then by the Random (P <.05) method. Scores decreased over time for all methods tested (P <.05). The Random method presented the highest standard deviation; however, an increase in standard deviation among consumers along days of display was observed for all methods tested (P <.05). Shelf-life determined by regression varied according to the method of presentation (7.83, 7.00 and 7.54 days for Random, Sequential and Animal, respectively). A minimum number of 4 day points before and 4 day points after neutral scores had been reached (scores = 5.0) were necessary in order to obtain a robust model. The minimum number of required consumers (a = 0.05; d = 0.1 and Ăź = 0.2 or 0.1) varied according to methodology: it was 81 to 109 consumers for Random, 69 to 92 for Sequential, and 55 to 74 for Animal. Our study indicates that an optimal number of days and evaluators can be calculated depending on the manner of sample presentation. These findings should be taken into account in further studies that aim to balance data reliability with the cost involved in meat colour analyses

    Essential oils in the diet of young bulls: Effect on animal temperament.

    Plant extracts and essential oils can be alternative products to antibiotics, because several plants produce secondary metabolites with antimicrobial properties. Additionally, they may act on the olfactory system, which sends signals to the central nervous system releasing endorphins that may affect the feeling of an animal''s welfare while altering an animal''s temperament. This study was conducted to evaluate the animal temperament of 40 young crossbred bulls (one-half Brown Swiss and one-half Nellore) 10 ± 2.2 mo old with an average BW of 219 ± 11.7 kg. Young bulls were randomly assigned to 1 of 5 diets: control, which had no clove or cinnamon; clove leaf included to supply 3, 500 mg/animal per day; clove leaf included to supply 7, 000 mg/animal per day; essential oil of leaf cinnamon to supply 3, 500 mg/animal per day; or essential oil of leaf cinnamon to supply 7, 000 mg/animal per day. The animal temperament was evaluated by calculating the chute score, exit score, and temperament score for periods (each 28 d during 6 periods) and among diets. The data were submitted to an ANOVA using GLM procedures with SAS version 9.0. The diet and period were considered fixed effects, whereas the animals were considered a random effect. Differences between means were evaluated using a Tukey test of 5% of significance. The addition of clove or cinnamon essential oils did not alter (P > 0.05) animal temperament. The analysis of temperament score (P = 0.55), chute score (P = 0.71), and exit score (P = 0.06) did not show significant differences among the diets. The exit score was similar among the 6 periods (1.57, 1.80, 2.98, 2.02, 2.73, and 2.66, respectively); however, chute score and temperament score were greater (P < 0.001) in the first period compared with other periods. The values for chute score were 1.85, 1.40, 1.35, 1.33, 1.20, 1.17, respectively, and the values for temperament score were 2.31, 1.81, 1.67, 1.69, 1.50, and 1.56, respectively, for periods 1 to 6. The results of this study suggest that clove and cinnamon essential oils can be added as an additive in high-grain finished diets without changing the animal temperament

    Validation of photographs usage to evaluate meat visual acceptability of young bulls finished in feedlot fed with or without essential oils

    Forty ½ Brown Swiss × ½ Nellore crossbred bulls were distributed into three experimental groups: CON – diet without addition of essential oils; CLO – diet with average 5, 000 mg/animal/day of clove essential oils and CIN – diet with average 5, 000 mg/animal/day of cinnamon essential oils to evaluate three methodologies of visual acceptability: with steaks directly in Trays and Sequential and Random photos. Seventeen consumers evaluated visual appearance of meat using a 9-point structured hedonic scale. CON group presented higher shelf-life than essential oils groups. Trays and Sequential scores were similar in the majority of days; thus digital images could be used to evaluate colour evolution. However, Random photos resulted in lower scores and slower acceptability decrease than Trays and Sequential photos (p < 0.05) among the second and fifth day of display. Random photos presented a lower and more constant standard deviation than Trays and Sequential photos (p < 0.01) indicating that this methodology promoted a higher standard situation for meat colour evaluation

    Pharmacological characterization of a new Ca2+ sensitizer

    The benzimidazole molecule was modified to synthesize a Ca(2+) sensitizer devoid of additional effects associated with Ca(2+) overload. Newly synthesized compounds, termed 1, 2, 3, 4, and 5, were evaluated in spontaneously beating and electrically driven atria from reserpine-treated guinea pigs. Compound 3 resulted as the most effective positive inotropic agent, and experiments were performed to study its mechanism of action. In spontaneously beating atria, the inotropic effect of 3 was concentration-dependent (3.0 microM-0.3 mM). Compound 3 was more potent and more active than the structurally related Ca(2+) sensitizers sulmazole and caffeine, but unlike them it did not increase the heart rate. In electrically driven atria, the inotropic activity of 3 was well preserved and it was not inhibited by propranolol, prazosin, ranitidine, pyrilamine, carbachol, adenosine deaminase, or ruthenium red. At high concentrations (0.1-1.0 mM) 3 inhibited phosphodiesterase-III, whereas it did not affect Na(+)/K(+)-ATPase, sarcolemmal Ca(2+)-ATPase, Na(+)/Ca(2+) exchange carrier, or sarcoplasmic reticulum Ca(2+) pump activities of guinea pig heart. In skinned fibers obtained from guinea pig papillary muscle and skeletal soleus muscle, compound 3 (0.1 mM, 1 mM) shifted the pCa/tension relation curve to the left, with no effect on maximal tension and no signs of toxicity. Compound 3 did not influence the basal or raised tone of guinea pig isolated aorta rings, whose cells do not contain the contractile protein troponin. The present results indicate that the inotropic effect of compound 3 seems to be primarily sustained by sensitization of the contractile proteins to Ca(2+)

    Synchronous down-modulation of miR-17 family members is an early causative event in the retinal angiogenic switch

    Six members of the microRNA-17 (miR-17) family were mapped to three different chromosomes, although they share the same seed sequence and are predicted to target common genes, among which are those encoding hypoxia-inducible factor-1α (HIF1A) and VEGFA. Here, we evaluated the in vivo expression profile of the miR-17 family in the murine retinopathy of prematurity (ROP) model, whereby Vegfa expression is highly enhanced at the early stage of retinal neovascularization, and we found simultaneous reduction of all miR-17 family members at this stage. Using gene reporter assays, we observed binding of these miRs to specific sites in the 3′ UTRs of Hif1a and Vegfa. Furthermore, overexpression of these miRs decreased HIF1A and VEGFA expression in vitro. Our data indicate that this miR-17 family elicits a regulatory synergistic down-regulation of Hif1a and Vegfa expression in this biological model. We propose the existence of a coordinated regulatory network, in which diverse miRs are synchronously regulated to target the Hif1a transcription factor, which in turn, potentiates and reinforces the regulatory effects of the miRs on Vegfa to trigger and sustain a significant physiological response

    Temperament and Impulsivity Predictors of Smoking Cessation Outcomes

    Aims: Temperament and impulsivity are powerful predictors of addiction treatment outcomes. However, a comprehensive assessment of these features has not been examined in relation to smoking cessation outcomes.Methods: Naturalistic prospective study. Treatment-seeking smokers (n = 140) were recruited as they engaged in an occupational health clinic providing smoking cessation treatment between 2009 and 2013. Participants were assessed at baseline with measures of temperament (Temperament and Character Inventory), trait impulsivity (Barratt Impulsivity Scale), and cognitive impulsivity (Go/No Go, Delay Discounting and Iowa Gambling Task). The outcome measure was treatment status, coded as “dropout” versus “relapse” versus “abstinence” at 3, 6, and 12 months endpoints. Participants were telephonically contacted and reminded of follow-up face to face assessments at each endpoint. The participants that failed to answer the phone calls or self-reported discontinuation of treatment and failed to attend the upcoming follow-up session were coded as dropouts. The participants that self-reported continuing treatment, and successfully attended the upcoming follow-up session were coded as either “relapse” or “abstinence”, based on the results of smoking behavior self-reports cross-validated with co-oximetry hemoglobin levels. Multinomial regression models were conducted to test whether temperament and impulsivity measures predicted dropout and relapse relative to abstinence outcomes.Results: Higher scores on temperament dimensions of novelty seeking and reward dependence predicted poorer retention across endpoints, whereas only higher scores on persistence predicted greater relapse. Higher scores on the trait dimension of non-planning impulsivity but not performance on cognitive impulsivity predicted poorer retention. Higher non-planning impulsivity and poorer performance in the Iowa Gambling Task predicted greater relapse at 3 and 6 months and 6 months respectively.Conclusion: Temperament measures, and specifically novelty seeking and reward dependence, predict smoking cessation treatment retention, whereas persistence, non-planning impulsivity and poor decision-making predict smoking relapse.This research was funded by the Occupational Medicine Area (Prevention Service); Department of Personality, Assessment and Psychological Treatment, University of Granada (Spain); and Ministerio de Economía y Competitividad grant (MINICO, ref. # PSI2013-45055-P) for the first and second authors

    The dopamine D2/D3 receptor agonist quinpirole increases checking-like behaviour in an operant observing response task with uncertain reinforcement: a novel possible model of OCD.

    Excessive checking is a common, debilitating symptom of obsessive-compulsive disorder (OCD). In an established rodent model of OCD checking behaviour, quinpirole (dopamine D2/3-receptor agonist) increased checking in open-field tests, indicating dopaminergic modulation of checking-like behaviours. We designed a novel operant paradigm for rats (observing response task (ORT)) to further examine cognitive processes underpinning checking behaviour and clarify how and why checking develops. We investigated i) how quinpirole increases checking, ii) dependence of these effects on D2/3 receptor function (following treatment with D2/3 receptor antagonist sulpiride) and iii) effects of reward uncertainty. In the ORT, rats pressed an 'observing' lever for information about the location of an 'active' lever that provided food reinforcement. High- and low-checkers (defined from baseline observing) received quinpirole (0.5mg/kg, 10 treatments) or vehicle. Parametric task manipulations assessed observing/checking under increasing task demands relating to reinforcement uncertainty (variable response requirement and active-lever location switching). Treatment with sulpiride further probed the pharmacological basis of long-term behavioural changes. Quinpirole selectively increased checking, both functional observing lever presses (OLPs) and non-functional extra OLPs (EOLPs). The increase in OLPs and EOLPs was long-lasting, without further quinpirole administration. Quinpirole did not affect the immediate ability to use information from checking. Vehicle and quinpirole-treated rats (VEH and QNP respectively) were selectively sensitive to different forms of uncertainty. Sulpiride reduced non-functional EOLPs in QNP rats but had no effect on functional OLPs. These data have implications for treatment of compulsive checking in OCD, particularly for serotonin-reuptake-inhibitor treatment-refractory cases, where supplementation with dopamine receptor antagonists may be beneficial
