    IgA and IgG antibodies in SARS-CoV-2 vaccinated health workers by a homemade ELISA diagnostic test

    Abstract The SARS-CoV-2 pandemic has accelerated vaccine development and testing, an important step in its eradication. Health workers were included among the first subjects to be vaccinated, therefore it was necessary to check their immunological status after the first and second dose of vaccine. Serum samples belonging to 24 health workers undergoing vaccination for SARS-CoV-2 (Pfizer-BioNTech) were analysed: for 2 of them it was also possible to obtain a serum sample prior to the first dose of vaccine (zero time); antibody dosing was performed in 18 out of 24 after the first and second vaccination dose; in the remaining 6 healthcare workers a serum sample was obtained only after the second dose. In each serum sample, IgA and IgG antibodies to "Spike Receptor Binding Domain" (RBD) and "Nucleocapsid" (N) proteins were searched by ELISA test. Except for the two subjects for whom it was possible to have a serum sample before vaccination, the others were collected on day 18 from the first dose of vaccine and on day 8 from the second dose. Serum samples collected after the first dose of vaccine showed reactivity to anti-RBD IgG in 11 out of 18 healthcare workers and to IgA in 2 subjects. After the second dose all 24 samples showed the presence of anti-S IgG, while 5 revealed a reactivity for anti-S IgA. Three samples showed reactivity towards anti-N IgG. The ELISA test has shown all its effectiveness in controlling post vaccine immunity and in discriminating natural immunity from vaccine induced immunity

    Human Papillomavirus Type Distribution and Correlation with Cyto-Histological Patterns in Women from the South of Italy

    Human papillomavirus (HPV) type-specific distribution was evaluated in genital samples collected from 654 women from the South of Italy undergoing voluntary screening and correlated with cyto-histological abnormalities. HPV DNA was detected in 45.9% of the samples, 41.7% of which had multiple infection and 89.0% had high-risk HPV infection. The prevalence of HPV infection and the rate of multiple infections decreased with age, suggesting natural selection of HPV types with better fitness. In line with other Italian studies, the most common HPV types were HPV-6 and HPV-16, followed by HPV-51, HPV-31, HPV-53, and HPV-66, in women with both normal and abnormal cytology. Cervical intraepithelial lesions grade 2 or 3 were associated with high-risk HPV-16, HPV-18, HPV-31, and HPV-51 infection. These data indicate that prophylactic HPV vaccination is expected to reduce the burden of HPV-related cervical lesions in this population, but also suggest the potential utility of new vaccines with larger type coverage

    In 1931 at Tebtunis in the Fayyum oasis, Carlo Anti and Gilbert Bagnani discovered one of the largest deposit of Egyptian literary papyri ever made and moreover it originally derived from the library of the temple itself. The precise location of this deposit, however, has never been clear, because there was not the possibility to see and study the whole archive, collected by Anti, made by photographic materials and written documents. On the contrary, now Carlo Anti\u2019s archive has recently been reordered at the Istituto Veneto and in the Museum of Archaeological Sciences and Arts (University of Padua)

    Protective effect of epigallocatechin-3-gallate (EGCG) in diseases with uncontrolled immune activation: Could such a scenario be helpful to counteract COVID-19?

    Some coronavirus disease 2019 (COVID-19) patients develop acute pneumonia which can result in a cytokine storm syndrome in response to Severe Acute Respiratory Syndrome coronavirus 2 (SARS-CoV-2) infection. The most effective anti-inflammatory drugs employed so far in severe COVID-19 belong to the cytokine-directed biological agents, widely used in the management of many autoimmune diseases. In this paper we analyze the efficacy of epigallocatechin 3-gallate (EGCG), the most abundant ingredient in green tea leaves and a well-known antioxidant, in counteracting autoimmune diseases, which are dominated by a massive cytokines production. Indeed, many studies registered that EGCG inhibits signal transducer and activator of transcription (STAT)1/3 and nuclear factor kappa-light-chain-enhancer of activated B cells (NF-\u3baB) transcription factors, whose activities are crucial in a multiplicity of downstream pro-inflammatory signaling pathways. Importantly, the safety of EGCG/green tea extract supplementation is well documented in many clinical trials, as discussed in this review. Since EGCG can restore the natural immunological homeostasis in many different autoimmune diseases, we propose here a supplementation therapy with EGCG in COVID-19 patients. Besides some antiviral and anti-sepsis actions, the major EGCG benefits lie in its anti-fibrotic effect and in the ability to simultaneously downregulate expression and signaling of many inflammatory mediators. In conclusion, EGCG can be considered a potential safe natural supplement to counteract hyper-inflammation growing in COVID-19

    Archaeology and virtual acoustics. A pan flute from ancient Egypt

    This paper presents the early developments of a recently started research project, aimed at studying from a multidisciplinary perspective an exceptionally well preserved ancient pan flute. A brief discussion of the history and iconography of pan flutes is provided, with a focus on Classical Greece. Then a set of non-invasive analyses are presented, which are based on 3D scanning andmaterials chemistry, and are the starting point to inspect the geometry, construction, age and geographical origin of the instrument. Based on the available measurements, a preliminary analysis of the instrument tuning is provided, which is also informed with elements of theory of ancient Greek music. Finally, the paper presents current work aimed at realizing an interactive museum installation that recreates a virtual flute and allows intuitive access to all these research facets

    Two Naturally Occurring Terpenes, Dehydrocostuslactone and Costunolide, Decrease Intracellular GSH Content and Inhibit STAT3 Activation

    The main purpose of the present study is to envisage the molecular mechanism of inhibitory action ofdehydrocostuslactone (DCE) andcostunolide (CS), two naturally occurring sesquiterpene lactones, towards the activation of signal transducer and activator of transcription 3 (STAT3). We report that, in human THP-1 cell line, they inhibit IL-6-elicited tyrosine phosphorylation of STAT3 and its DNA binding activity with EC50 of 10 ”M with concomitantdown-regulation ofthe phosphorylation of the tyrosine Janus kinases JAK1, JAK2 and Tyk2. Furthermore, these compounds that contain an α-ÎČ-unsatured carbonyl moiety and function as potent Michael reaction acceptor, induce a rapid drop in intracellular glutathione (GSH) concentration by direct interaction with it, thereby triggering S-glutathionylation of STAT3. Dehydrocostunolide (HCS), the reduced form of CS lacking only the α-ÎČ-unsaturated carbonyl group, fails to exert any inhibitory action. Finally, the glutathione ethylene ester (GEE), the cell permeable GSH form, reverts the inhibitory action of DCE and CS on STAT3 tyrosine phosphorylation. We conclude that these two sesquiterpene lactones are able to induce redox-dependent post-translational modification of cysteine residues of STAT3 protein in order to regulate its function
