Targeting GLI factors to inhibit the Hedgehog pathway

Hedgehog (Hh) signaling has emerged in recent years as an attractive target for anticancer therapy because its aberrant activation is implicated in several cancers. Major progress has been made in the development of SMOOTHENED (SMO) antagonists, although they have shown several limitations due to downstream SMO pathway activation or the occurrence of drug-resistant SMO mutations. Recently, particular interest has been elicited by the identification of molecules able to hit glioma-associated oncogene (GLI) factors, the final effectors of the Hh pathway, which provide a valid tool to overcome anti-SMO resistance. Here, we review results achieved in developing GLI antagonists, explaining their mechanisms of action and highlighting their therapeutic potential. The retail activity of foreign banks in Italy: effects on credit supply to households and firms

This paper investigates the effects of the increasing activity of foreign banks in Italy, distinguishing between credit granted to households and firms. Foreign banks display a differentiated degree of business expansion across the provinces, we exploit this variability to measure how foreign banks affect competition and test it with reference to: i) a market share instability index ; ii) interest rates; iii) the collateral requested. Our results, over the period 1997-2006, show that an increase in foreign intermediaries’ market share leads to a less stable market share index. As for mortgage loans to households, this enhanced competitive pressure implies a reduction in the average interest rate applied and an increase in the loans granted with respect to collateral. We do not find a significant impact on the average interest rate conditions applied to firms, although, in the most recent period, a reduction in the collateral on loans with longer maturity has emerged.foreign banks, credit market competition

El mercado de estados unidos como una oportunidad de negocio para incrementar la exportación de lúcuma peruana 2017 – 2021

RESUMEN El presente informe de tesis denominado El Mercado de Estados Unidos como una oportunidad de negocio para incrementar la exportación de lúcuma peruana en el periodo 2017 – 2021, tiene como finalidad determinar en qué medida el mercado de Estados Unidos constituye una oportunidad de negocio para la pulpa de lúcuma. Esta tesis tiene como hipótesis El mercado de Estados Unidos constituye una oportunidad de negocio significativa para aumentar la exportación de pulpa lúcuma peruana, periodo 2017 – 2021 para la afirmación de esta hipótesis se ha realizado una investigación cualitativa y cuantitativa; en las cuales se analizó el mercado estadounidense y sus importaciones, sus indicadores económicos y las exportaciones de Perú a Estados Unidos, además de los hábitos y costumbres de sus consumidores. En adición se realizó una entrevista a un experto en materia de Economía y exportaciones. La conclusión a la que se ha llegado es que el mercado de Estados Unidos si constituye una oportunidad de negocio para incrementar las exportaciones de pulpa de lúcuma, esto se ve demostrado a través de la oportunidad que representa el mercado de Estados Unidos, luego de analizar la oferta exportable y un análisis económico financiero; aceptando de esta manera la hipótesis de la presente investigación.ABSTRACT This thesis report named the United States market as a business opportunity to increase Peruvian exports of lucuma in the period 2017 - 2021; it aims to determine to what extent the US market is a business opportunity for the pulp of lucuma. This thesis has as hypothesis The US market is a significant business opportunity to increase exports of Peruvian lucuma pulp, period 2017 - 2021 for the affirmation of this hypothesis has made a qualitative and quantitative research; in which the US market and its imports, its economic indicators and exports from Peru to the United States was analyzed, along with the habits and customs of consumers. In addition an interview was conducted an expert in economics and exports. The conclusion that has been reached is that the US market is a business opportunity to increase exports of pulp of lucuma, this is demonstrated through the opportunity presented by the US market, after analyzing the exportable supply and economic and financial analysis; in this way accepting the hypothesis of this research

Cybersecurity as a fundamental base in the operations of the logistics chain in international trade according to the SGCS BASC

El presente trabajo muestra la importancia que tiene la ciber-seguridad dentro de las operaciones de la cadena logística en comercio internacional. Basándose principalmente en destacar los activos de información que tiene una empresa y en muchas ocasiones ignora cómo protegerla de posibles robos o modificaciones también conocidos como ciber-ataques. Por otro lado, presenta una lista de las vulnerabilidades a la que está expuesta la información no solo de una empresa sino también personal, esta misma puede ser usada con fines ilícitos lo que puede conllevar a pérdidas económicas para las empresas y procesos judiciales para las personas que se ven involucradas. Ante estas problemáticas se proyecta al SGCS BASC como una herramienta para mitigar y prevenir las amenazas. Activities such as online purchases, communications over long distances by technological means, the connection of various places in the world, artificial intelligence as support in daily life and even the nano technology that has begun to venture into the field of medicine. All of the above is considered a great advantage of technology, but it is important not to leave aside its disadvantages, even more focusing on another global trend which is globalization and the opening of markets. In international trade, especially in supply chains, various processes are developed by technological means; from the simplest file to an entire load control program that with the smallest failure in the systems can not only generate problems in the operations, but also millionaire losses and even the total completion of operations by a company. El podcast como herramienta de comunicación del Comité Olímpico Colombiano

El Comité Olímpico Colombiano, ha experimentado en los últimos años una etapa de transición y crecimiento equivalente al aumento en importancia y número por participación del país en eventos deportivos internacionales, además del desarrollo deportivo y ascenso profesional de los atletas colombianos en competiciones de importancia mundial. Esa inesperada y rápida evolución del deporte en Colombia, y de sus federaciones que ha llevado al Comité Olímpico Colombiano a afrontar un progreso para el que no estaba preparado, hace unos años solo existían un par de personas a cargo de diversos asuntos de los que dependía la organización, éxito y posicionamiento de los atletas y federaciones en el ámbito deportivo mundial, el ejemplo más claro es que hace tres años, la organización contaba únicamente con un Jefe de Prensa, hoy existe un área de Comunicaciones y Relaciones Públicas conformada por tres personas, patrón que se ha estado repitiendo con las demás áreas. Internamente el Comité Olímpico Colombiano está compuesto por áreas que atienden todas las necesidades de federaciones, clubes y atletas para lograr su clasificación en eventos del ciclo olímpico, y la intensificación de esas necesidades ha causado que cada área se concentre en esa meta que se han olvidado de la meta unificada como organización. Se cuenta con una ventaja y es el interés de los medios en la delegación colombiana que participó en las olimpiadas de verano en Londres, pero no sirve el interés de los públicos objetivos en la organización sin un refuerzo desde el interior de ella.Comunicador (a) SocialPregrad

O Ambiente de Sala de Aula em Escolas Públicas: Voltada a um Ensino mais Refletivo

The primary goal of this paper is to examine the classroom environment in Colombian public schools based on the trainee teachers’ reflective views when undertaking practicum day-to-day experiences during the final stage of their professional studies. The most important theoretical concepts are based on critical thinking, reflective teaching, and classroom management. The method follows the qualitative research approach, catching a deeper comprehension of the classroom as a complex setting where the teaching-learning processes are carried out according to the curriculum guidelines. Through diaries, observations, narratives, interviewsand meaningful strategies, trainee teachers were able to obtain a more in-depth perception to reach a high level of critical reflections in dealing with the most common academic misconduct episodes which perturb a healthy atmosphere inside the classroom. The results are discussed in relation to the most frequent obstacles, the reasons why they emerge and the implications on the school life. Beyond the mechanical task of planning, developing, and evaluating a class, the challenge of helping the most undisciplined students requires a deep process of reflection, critical thinking, and effective solutions in the light of current international theories andnational education policies concerning teacher preparation  goals.El objetivo principal de este artículo es examinar el ambiente del aula de clase en escuelas públicas colombianas con base en las percepciones, reflexiones y experiencias directas de los educadores en proceso de formación cuando realizan la última etapa de su formación profesional. Los conceptos teóricos más importantes se basan en pensamiento crítico, enseñanza reflexiva y gestión del aula. La metodología sigue el enfoque de la investigación cualitativa, que permite comprender a profundidad el aula como un entorno complejo donde se llevan a cabo los procesosde enseñanza y aprendizaje previstos en el currículo. A través de diarios, observaciones, narrativas y estrategias significativas, los educadores en formación pudieron obtener una percepción más profunda de la realidad del aula con el fin de alcanzar un alto nivel de reflexiones críticas para aplicar estrategias efectivas en la solución de los problemas académicos más comunes que perturban un ambiente pedagógico propicio. Los resultados se discuten en relación con los obstáculos más frecuentes, las razones por las cuales emergen y las implicaciones en la vida escolar. Más allá de la tarea instrumental de planear, desarrollar y evaluar una clase, ayudar a los estudiantes indisciplinados se convierte en un desafío que exige un proceso reflexivo y crítico acompañado de soluciones efectivas a la luz de las teorías internacionales actuales y de las políticas nacionales relacionadas con la formación de educadores.O objetivo principal deste artigo foi estudar o ambiente cotidiano da sala de aula em colégios públicos colombianos baseado nas percepções, reflexões e experiências diretas dos docentes em processo de formação, especificamente quandofazem o último ciclo do seu curso profissional. A metodologia segue o modelo de pesquisa qualitativa que permite compreender a profundeza a sala de aula como um ambiente complexo, onde são feitos processos de ensino e aprendizagem programados no currículo. Através dediários, observações, narrações e estratégias significativas, os docentes em formaçãopuderam obter uma percepção mais extensa e viva da realidade da sala aula. Isto, com o intuito de atingir um alto nível de reflexões críticas para aplicar estratégias efetivas na resolução dos problemas acadêmicos mais comuns que perturbam um ambiente pedagógico favorável. Synergistic inhibition of the Hedgehog pathway by newly designed Smo and Gli antagonists bearing the isoflavone scaffold

Aberrant activation of the Hedgehog (Hh) pathway is responsible for the onset and progression of several malignancies. Small molecules able to block the pathway at the upstream receptor Smoothened (Smo) or the downstream effector Gli1 have thus emerged recently as valuable anticancer agents. Here, we have designed, synthesized, and tested new Hh inhibitors taking advantage by the highly versatile and privileged isoflavone scaffold. The introduction of specific substitutions on the isoflavone's ring B allowed the identification of molecules targeting preferentially Smo or Gli1. Biological assays coupled with molecular modeling corroborated the design strategy, and provided new insights into the mechanism of action of these molecules. The combined administration of two different isoflavones behaving as Smo and Gli antagonists, respectively, in primary medulloblastoma (MB) cells highlighted the synergistic effects of these agents, thus paving the way to further and innovative strategies for the pharmacological inhibition of Hh signaling

Inhibition of Hedgehog-dependent tumors and cancer stem cells by a newly identified naturally occurring chemotype

Hedgehog (Hh) inhibitors have emerged as valid tools in the treatment of a wide range of cancers. Indeed, aberrant activation of the Hh pathway occurring either by ligand-dependent or -independent mechanisms is a key driver in tumorigenesis. The smoothened (Smo) receptor is one of the main upstream transducers of the Hh signaling and is a validated target for the development of anticancer compounds, as underlined by the FDA-approved Smo antagonist Vismodegib (GDC-0449/Erivedge) for the treatment of basal cell carcinoma. However, Smo mutations that confer constitutive activity and drug resistance have emerged during treatment with Vismodegib. For this reason, the development of new effective Hh inhibitors represents a major challenge for cancer therapy. Natural products have always represented a unique source of lead structures in drug discovery, and in recent years have been used to modulate the Hh pathway at multiple levels. Here, starting from an in house library of natural compounds and their derivatives, we discovered novel chemotypes of Hh inhibitors by mean of virtual screening against the crystallographic structure of Smo. Hh functional based assay identified the chalcone derivative 12 as the most effective Hh inhibitor within the test set. The chalcone 12 binds the Smo receptor and promotes the displacement of Bodipy-Cyclopamine in both Smo WT and drug-resistant Smo mutant. Our molecule stands as a promising Smo antagonist able to specifically impair the growth of Hh-dependent tumor cells in vitro and in vivo and medulloblastoma stem-like cells and potentially overcome the associated drug resistance
