    A phosphorylation-induced switch in the nuclear localization sequence of the intrinsically disordered nupr1 hampers binding to importin

    Several carrier proteins are involved in protein transport from the cytoplasm to the nucleus in eukaryotic cells. One of those is importin a, of which there are several human isoforms; among them, importin a3 (Impa3) has a high flexibility. The protein NUPR1, a nuclear protein involved in the cell-stress response and cell cycle regulation, is an intrinsically disordered protein (IDP) that has a nuclear localization sequence (NLS) to allow for nuclear translocation. NUPR1 does localize through the whole cell. In this work, we studied the affinity of the isolated wild-type NLS region (residues 54–74) of NUPR1 towards Impa3 and several mutants of the NLS region by using several biophysical techniques and molecular docking approaches. The NLS region of NUPR1 interacted with Impa3, opening the way to model the nuclear translocation of disordered proteins. All the isolated NLS peptides were disordered. They bound to Impa3 with low micromolar affinity (1.7–27 µM). Binding was hampered by removal of either Lys65 or Lys69 residues, indicating that positive charges were important; furthermore, binding decreased when Thr68 was phosphorylated. The peptide phosphorylated at Thr68, as well as four phospho-mimetic peptides (all containing the Thr68Glu mutation), showed the presence of a sequential NN(i, i + 1) nuclear Overhauser effect (NOE) in the 2D-1H-NMR (two-dimensional–proton NMR) spectra, indicating the presence of turn-like conformations. Thus, the phosphorylation of Thr68 modulates the binding of NUPR1 to Impa3 by a conformational, entropy-driven switch from a random-coil conformation to a turn-like structure

    Caliope: an operational air quality forecasting system for the Iberian Peninsula, Balearic Islands and Canary Islands ? first annual evaluation and ongoing developments

    International audienceThe Caliope project funded by the Spanish Ministry of the Environment establishes an air quality forecasting system for Spain to increase the knowledge on transport and dynamics of pollutants in Spain, to assure the accomplishment of legislation and to inform the population about the levels of pollutants, topics in which the European Commission has shown a great concern. The present contribution describes the first quantitative verification study performed so far with two chemistry transport models (CMAQ and CHIMERE) for a reference year (2004) at medium spatial resolution (around 20Ă—20 km for the Iberian Peninsula). Both models perform similarly in the case of ground-level ozone. The mean normalised gross error MNGE remains below 15?20% during summertime, when ozone episodes occur, outlining the good skills of the system concerning the forecasting of air quality in Spain. Furthermore, the ongoing developments of the system towards high resolution modelling (4Ă—4 km for Spain, 12Ă—12 km for Europe, 1 h temporal resolution) and the integration with observations within the Caliope umbrella are described

    A historical vertebrate collection from the Middle Miocene of the Peruvian Amazon

    The Miocene aquatic and terrestrial fossil record from western Amazonia constitute a clear evidence of the palaeoenvironmental diversity that prevailed in the area, prior to the establishment of the Amazon River drainage. During the Miocene, the region was characterized by a freshwater megawetland basin, influenced by episodic shallow-marine incursions. A fossil vertebrate collection from the middle Miocene strata of the Pebas Formation is here studied and described. This historical collection was recovered in 1912 along the banks of the Itaya River (Iquitos, Peru), during a scientific expedition led by two scientists of the University of Zurich, Hans Bluntschli and Bernhard Peyer. Our findings include a total of 34 taxa, including stingrays, bony fishes, turtles, snakes, crocodylians, and lizards. Fishes are the most abundant group in the assemblage (~ 23 taxa), including the first fossil record of the freshwater serrasalmids Serrasalmus, and Mylossoma, and the hemiodontid Hemiodus for the Pebas system, with the latter representing the first fossil be discovered for the entire Hemiodontidae. The presence of a representative of Colubroidea in the middle Miocene of Iquitos supports the hypothesis of arrival and dispersal of these snakes into South America earlier than previously expected. This fossil assemblage sheds light on the palaeoenvironments, and the geographical/temporal range of several aquatic/terrestrial lineages inhabiting the Amazonian region

    Određivanje aglikona flavonoida iz vrsta roda Sideritis (Lamiaceae) iz Makedonije pomoću HPLC UV DAD

    Flavonoids obtained from Sideritis species (Lamiaceae), S. raeseri and S. scardica, grown in Macedonia were studied. Qualitative and quantitative analyses of the flavonoid aglycones were performed using high-performance liquid chromatography (HPLC) with a UV diode array detector. Extracts were prepared by acid hydrolysis in acetone, reextraction in ethyl acetate and evaporation to dryness; the residue dissolved in methanol was subjected to HPLC analysis. Isoscutellarein, chryseriol and apigenin were identified in the extracts. Also, a 4’-methyl ether derivative of isoscutellarein was found, together with hypolaetin and its methyl ether derivative, which were identified according to previously isolated glycosides and literature data. Quantitation was performed using calibration with apigenin. According to this screening analysis, the samples of the genus Sideritis from Macedonia are rich in polyhydroxy flavones and analogous with the previously studied Mediterranean Sideritis species from the Ibero-North African and Greek Sideritis species with respect to the presence of 8-OH flavones and their derivatives.U radu su proučavani flavonoidi dobiveni iz Sideritis vrsta (Lamiaceae), S. raeseri i S. scardica, porijeklom iz Makedonije. Ekstrakti su pripravljeni kiselom hidrolizom u acetonu, te ponovnom ekstrakcijom etil-acetatom. Ostatak nakon uparavanja je otopljen u metanolu i analiziran pomoću HPLC. Usporedbom s ranije izoliranim glikozidima i s literaturnim podacima u ekstraktima su identifirani izoskutelarein, krizeriol, apigenin, 4\u27-metil eterski derivat izoskutelareina, hipolaetin te njegov metil eter. Kvantifikacija je provedena pomoću kalibracijske krivulje za apigenin. Rezultati ukazuju da su uzorci Sideritis roda iz Makedonije bogati polihidroksiflavonima kao i ranije proučavane mediteranske Sideritis vrste iz sjeverne Afrike i Grčke Sideri

    The karyotype of Nothoscordum arenarium Herter (Gilliesioideae, Alliaceae): A populational and cytomolecular analysis

    The genus Nothoscordum Kunth comprises approximately 20 species native to South America. Karyologically, the genus is remarkable for its large chromosomes and Robertsonian translocations. Variation in chromosome number has been recorded in a few polyploid species and it is unknown among diploids. This study presents the chromosome number and morphology of 53 individuals of seven populations of N. arenarium Herter (2n = 10). In addition, karyotype analyses after C-banding, staining with CMA and DAPI, and in situ hybridization with 5S and 45S rDNA probes were performed in six individuals from one population. All individuals exhibited 2n = 10 (6M + 4A), except for one tetraploid (2n = 20, 12M + 8A) and one triploid (2n = 15, 9M + 6A) plant. C-banding revealed the presence of CMA+ /DAPI - heterochromatin in the short arm and in the proximal region of the long arm of all acrocentric chromosomes. The 45S rDNA sites co-localized with the CMA + regions of the acrocentrics short arms, while the 5S rDNA probe only hybridized with the subterminal region of a pair of metacentric chromosomes. A change in the pattern of CMA bands and rDNA sites was observed in only one individual bearing a reciprocal translocation involving the long arm of a metacentric and the long arm of an acrocentric chromosome. These data suggest that, despite isolated cases of polyploidy and translocation, the karyotype of N. arenarium is very stable and the karyotypic instability described for other species may be associated with their polyploid condition

    El análisis morfotectónico del sector central del Promontorio Balear, entre las islas de Ibiza y Mallorca, mediante batimetría multihaz y perfiles sísmicos de muy alta resolución ha permitido diferenciar varios rasgos morfológicos relacionados con la tectónica a dos escalas. Los elementos de gran escala, de dirección NE-SO, son: i) el escarpe Émile Baudot; ii) el monte submarino Émile Baudot, y iii) la alineación de montes submarinos Ausiàs March y Ses Olives. Los elementos de pequeña escala son: i) dos elevaciones longitudinales (ENE-OSO); ii) escarpes rectilíneos (NO-SE a NE-SO); iii) depresiones tectónicas (NNE SSO a NE-SO); y iv) alineaciones rectilíneas de pockmarks (NO-SE a NNE-SSO). Los elementos de mayor tamaño responden a la configuración estructural neógeno-cuaternaria. El escarpe Émile Baudot limita el Promontorio Balear y la cuenca oceánica del Mediterráneo Occidental. La alineación de montes submarinos Ausiàs March y Ses Olives está generada por una fase extensional, y el monte submarino Émile Baudot forma parte de una alineación volcánica NO-SE asociada a la Zona de Fractura de Ibiza. Los elementos de pequeña escala están relacionados con estructuras tectónicas activas que deforman las unidades sedimentarias del Cuaternario y el fondo marino. Las elevaciones longitudinales están generadas por pliegues anticlinales, mientras que el resto de estructuras están asociadas a la actividad de fallas normales de buzamiento alto.A morphotectonic analysis has been carried out between the islands of Ibiza and Majorca in the central sector of the Balearic Promontory using multibeam bathymetry and very high-resolution seismic profiles. Several morphological features related to tectonics at two different scales have been differentiated. The large-scale features have a NE-SW direction, they are: i) the Émile Baudot Escarpment; ii) the Émile Baudot seamount, and iii) the Ausiàs March and Ses Olives seamounts alignment. The differentiated small-scale elements are i) two longitudinal elevations (ENE-WSW); ii) rectilinear scarps (NW-SE to NE-SW); iii) tectonic depressions limited by one or two scarps (NNE-SSW to NE-SW), and iv) rectilinear pockmarks alignments (NW-SE to NNE-SSW). The largest elements respond to the Neogene-Quaternary structural configuration. The Émile Baudot escarpment is the limit between the Balearic Promontory and the Western Mediterranean oceanic basin. The alignment of the Ausiàs March and Ses Olives seamounts respond to the last extensional phase, and the Émile Baudot seamount forms part of a NW-SE volcanic alignment that could be associated with the Ibiza Fracture Zone. The smaller elements are related to active tectonic structures that deform Quaternary sedimentary units and the seafloor. The longitudinal elevations correspond to anticlinal folds, while the rest of the structures are associated to the activity of near vertical normal faults.En prens

    Single neutral pion production by charged-current νˉμ\bar{\nu}_\mu interactions on hydrocarbon at ⟨Eν⟩=\langle E_\nu \rangle = 3.6 GeV

    Single neutral pion production via muon antineutrino charged-current interactions in plastic scintillator (CH) is studied using the \minerva detector exposed to the NuMI low-energy, wideband antineutrino beam at Fermilab. Measurement of this process constrains models of neutral pion production in nuclei, which is important because the neutral-current analog is a background for νˉe\bar{\nu}_e appearance oscillation experiments. The differential cross sections for π0\pi^0 momentum and production angle, for events with a single observed π0\pi^0 and no charged pions, are presented and compared to model predictions.
