    Interaction of suppressor of cytokine signalling 3 with cavin-1 links SOCS3 function and cavin-1 stability

    YesEffective suppression of JAK–STAT signalling by the inducible inhibitor “suppressor of cytokine signalling 3” (SOCS3) is essential for limiting signalling from cytokine receptors. Here we show that cavin-1, a component of caveolae, is a functionally significant SOCS3- interacting protein. Biochemical and confocal imaging demonstrate that SOCS3 localisation to the plasma membrane requires cavin-1. SOCS3 is also critical for cavin-1 stabilisation, such that deletion of SOCS3 reduces the expression of cavin-1 and caveolin-1 proteins, thereby reducing caveola abundance in endothelial cells. Moreover, the interaction of cavin-1 and SOCS3 is essential for SOCS3 function, as loss of cavin-1 enhances cytokine-stimulated STAT3 phosphorylation and abolishes SOCS3-dependent inhibition of IL-6 signalling by cyclic AMP.     Specific refolding pathway of viscumin A chain in membrane-like medium reveals a possible mechanism of toxin entry into cell

    How is a water-soluble globular protein able to spontaneously cross a cellular membrane? It is commonly accepted that it undergoes significant structural rearrangements on the lipid-water interface, thus acquiring membrane binding and penetration ability. In this study molecular dynamics (MD) simulations have been used to explore large-scale conformational changes of the globular viscumin A chain in a complex environment - comprising urea and chloroform/methanol (CHCl /MeOH) mixture. Being well-packed in aqueous solution, viscumin A undergoes global structural rearrangements in both organic media. In urea, the protein is "swelling" and gradually loses its long-distance contacts, thus resembling the "molten globule" state. In CHCl /MeOH, viscumin A is in effect turned "inside out". This is accompanied with strengthening of the secondary structure and surface exposure of hydrophobic epitopes originally buried inside the globule. Resulting solvent-adapted models were further subjected to Monte Carlo simulations with an implicit hydrophobic slab membrane. In contrast to only a few point surface contacts in water and two short regions with weak protein-lipid interactions in urea, MD-derived structures in CHCl /MeOH reveal multiple determinants of membrane interaction. Consequently it is now possible to propose a specific pathway for the structural adaptation of viscumin A with respect to the cell membrane - a probable first step of its translocation into cytoplasmic targets

    Specific human leukocyte antigen DQ influence on expression of antiislet autoantibodies and progression to type 1 diabetes

    Human leukocyte antigen (HLA) DQ haplotypes have the strongest genetic association with type 1 diabetes (T1DM) risk. OBJECTIVE: The objective of the study was to analyze whether HLA DQ alleles influence the development of antiislet autoantibodies, the progression to T1DM among autoantibody-positive relatives, or both. DESIGN: The Diabetes Prevention Trial-1 screened more than 90,000 nondiabetic relatives of patients for cytoplasmic islet-cell autoantibody (ICA) expression between 1994 and 2002. SETTING: The study was conducted in the general community. PARTICIPANTS: The Diabetes Prevention Trial-1 found 2817 ICA-positive relatives who were tested for biochemical autoantibodies (GAD65, ICA512, and insulin) and HLA-DQ haplotypes, and 2796 of them were followed up for progression to diabetes for up to 8 yr (median, 3.6 yr). MAIN OUTCOME MEASURE: Progression to T1DM was measured. RESULTS: High-risk DQ haplotypes and genotypes were associated with a higher percentage of relatives expressing multiple biochemical autoantibodies and higher T1DM risk (e.g., respectively, 59 and 36% at 5 yr for carriers of the DQA1*0301-DQB1*0302/DQA1*0501-DQB1*0201 genotype). The number of autoantibodies expressed significantly increased T1DM risk and across different DQ genotypes, autoantibody positivity directly correlated with diabetes risk. However, multivariate analyses indicated that the influence of most genotypes on T1DM risk was not independent from autoantibody expression, with the possible exception of DQA1*0102-DQB1*0602. Specific genotypic combinations conferred 5-yr diabetes risks significantly lower (e.g. 7%-DQA1*0201-DQB1*0201/DQA1*0501-DQB1*0201 and 14%-DQA1*0301-DQB1*0301/DQA1*0501-DQB1*0201) than when those haplotypes were found in other combinations. CONCLUSION: HLA DQ alleles determine autoantibody expression, which is correlated with diabetes progression. Among autoantibody-positive relatives, most HLA DQ genotypes did not further influence T1DM risk

    Tomato: a crop species amenable to improvement by cellular and molecular methods

    Tomato is a crop plant with a relatively small DNA content per haploid genome and a well developed genetics. Plant regeneration from explants and protoplasts is feasable which led to the development of efficient transformation procedures. In view of the current data, the isolation of useful mutants at the cellular level probably will be of limited value in the genetic improvement of tomato. Protoplast fusion may lead to novel combinations of organelle and nuclear DNA (cybrids), whereas this technique also provides a means of introducing genetic information from alien species into tomato. Important developments have come from molecular approaches. Following the construction of an RFLP map, these RFLP markers can be used in tomato to tag quantitative traits bred in from related species. Both RFLP's and transposons are in the process of being used to clone desired genes for which no gene products are known. Cloned genes can be introduced and potentially improve specific properties of tomato especially those controlled by single genes. Recent results suggest that, in principle, phenotypic mutants can be created for cloned and characterized genes and will prove their value in further improving the cultivated tomato.

    Production and Decay of D_1(2420)^0 and D_2^*(2460)^0

    We have investigated D+πD^{+}\pi^{-} and D+πD^{*+}\pi^{-} final states and observed the two established L=1L=1 charmed mesons, the D1(2420)0D_1(2420)^0 with mass 242122+1+22421^{+1+2}_{-2-2} MeV/c2^{2} and width 2053+6+320^{+6+3}_{-5-3} MeV/c2^{2} and the D2(2460)0D_2^*(2460)^0 with mass 2465±3±32465 \pm 3 \pm 3 MeV/c2^{2} and width 2876+8+628^{+8+6}_{-7-6} MeV/c2^{2}. Properties of these final states, including their decay angular distributions and spin-parity assignments, have been studied. We identify these two mesons as the jlight=3/2j_{light}=3/2 doublet predicted by HQET. We also obtain constraints on {\footnotesize ΓS/(ΓS+ΓD)\Gamma_S/(\Gamma_S + \Gamma_D)} as a function of the cosine of the relative phase of the two amplitudes in the D1(2420)0D_1(2420)^0 decay.Comment: 15 pages in REVTEX format. hardcopies with figures can be obtained by sending mail to: [email protected]

    Measurement of the branching fraction for Υ(1S)τ+τ\Upsilon (1S) \to \tau^+ \tau^-

    We have studied the leptonic decay of the Υ(1S)\Upsilon (1S) resonance into tau pairs using the CLEO II detector. A clean sample of tau pair events is identified via events containing two charged particles where exactly one of the particles is an identified electron. We find B(Υ(1S)τ+τ)=(2.61 ± 0.12 +0.090.13)B(\Upsilon(1S) \to \tau^+ \tau^-) = (2.61~\pm~0.12~{+0.09\atop{-0.13}})%.     Study of the B^0 Semileptonic Decay Spectrum at the Upsilon(4S) Resonance

    We have made a first measurement of the lepton momentum spectrum in a sample of events enriched in neutral B's through a partial reconstruction of B0 --> D*- l+ nu. This spectrum, measured with 2.38 fb**-1 of data collected at the Upsilon(4S) resonance by the CLEO II detector, is compared directly to the inclusive lepton spectrum from all Upsilon(4S) events in the same data set. These two spectra are consistent with having the same shape above 1.5 GeV/c. From the two spectra and two other CLEO measurements, we obtain the B0 and B+ semileptonic branching fractions, b0 and b+, their ratio, and the production ratio f+-/f00 of B+ and B0 pairs at the Upsilon(4S). We report b+/b0=0.950 (+0.117-0.080) +- 0.091, b0 = (10.78 +- 0.60 +- 0.69)%, and b+ = (10.25 +- 0.57 +- 0.65)%. b+/b0 is equivalent to the ratio of charged to neutral B lifetimes, tau+/tau0.Comment: 14 page, postscript file also available at http://w4.lns.cornell.edu/public/CLN

    Measurement of the Decay Asymmetry Parameters in Λc+Λπ+\Lambda_c^+ \to \Lambda\pi^+ and Λc+Σ+π0\Lambda_c^+ \to \Sigma^+\pi^0

    We have measured the weak decay asymmetry parameters (\aLC ) for two \LC\ decay modes. Our measurements are \aLC = -0.94^{+0.21+0.12}_{-0.06-0.06} for the decay mode Λc+Λπ+\Lambda_c^+ \to \Lambda\pi^+ and \aLC = -0.45\pm 0.31 \pm 0.06 for the decay mode ΛcΣ+π0\Lambda_c \to \Sigma^+\pi^0 . By combining these measurements with the previously measured decay rates, we have extracted the parity-violating and parity-conserving amplitudes.     Observation of the Ξc+\Xi_c^+ Charmed Baryon Decays to Σ+Kπ+\Sigma^+ K^-\pi^+, Σ+Kˉ0\Sigma^+ \bar{K}^{*0}, and ΛKπ+π+\Lambda K^-\pi^+\pi^+

    We have observed two new decay modes of the charmed baryon Ξc+\Xi_c^+ into Σ+Kπ+\Sigma^+ K^-\pi^+ and Σ+Kˉ0\Sigma^+ \bar{K}^{*0} using data collected with the CLEO II detector. We also present the first measurement of the branching fraction for the previously observed decay mode Ξc+ΛKπ+π+\Xi_c^+\to\Lambda K^-\pi^+\pi^+. The branching fractions for these three modes relative to Ξc+Ξπ+π+\Xi_c^+\to\Xi^-\pi^+\pi^+ are measured to be 1.18±0.26±0.171.18 \pm 0.26 \pm 0.17, 0.92±0.27±0.140.92 \pm 0.27 \pm 0.14, and 0.58±0.16±0.070.58 \pm 0.16 \pm 0.07, respectively.Comment: 12 page uuencoded postscript file, postscript file also available through http://w4.lns.cornell.edu/public/CLN
