    Small Farm Households\u27 Vulnerabilities in South Cotabato, Philippines

    Low-productivity subsistence agriculture still prevails in many areas in the Philippines such as in the Municipality of Lake Sebu in the Province of South Cotabato in Mindanao. This study employs primary data collection methods; namely key informant interview (KII); focus group discussion (FGD) and a household survey to assess the conditions and problems constraining farming households in Lake Sebu. The study finds that constraints on sourcing and financing of farm inputs and on marketing of produce have perpetuated the dominating role of middlemen in agricultural financing and marketing that result in high input prices and low farm-gate output prices; and hence minimal net income for the farmers. The paper concludes with specific policy and program recommendations to achieve maximum and sustainable farming benefits; namely; an expansive farm-to-market infrastructure program; an institution and business support intervention program to link farmers and markets; conditional cash farming subsidies in lieu of direct provision of farm inputs; and crop diversification promotion and support programs

    Uncertainties of synchrotron microCT-based digital volume correlation bone strain measurements under simulated deformation

    Digital Volume Correlation (DVC) is used to measure internal displacements and strains in bone. Recent studies have shown that synchrotron radiation micro-computed tomography (SR-microCT) can improve the accuracy and precision of DVC. However, only zero-strain or virtually-moved test have been used to quantify the DVC uncertainties, leading to potential underestimation of the measurement errors. In this study, for the first time, the uncertainties of a global DVC approach have been evaluated on repeated SR-microCT scans of bovine cortical bone (voxel size: 1.6μm), which were virtually deformed for different magnitudes and along different directions. The results showed that systematic and random errors of the normal strain components along the deformation direction were higher than the errors along unstrained directions. The systematic percentage errors were smaller for larger virtual deformations. The random percentage error was in the order of 10% of the virtual deformation. However, higher errors were localized at the boundary of the volumes of interest, perpendicular to the deformation direction. When only the central region of the samples was considered (100 micrometers layers removed from the borders where the deformation was applied), the errors in the direction of virtual deformation were comparable to the errors in the unstrained directions. In conclusion, the method presented to estimate the uncertainties of DVC is suitable for testing anisotropic specimens as cortical bone. The good agreement between the uncertainties in measurements of strain components obtained with this approach and with the simpler zero-strain-test suggests that the latter is adequate in the tested deformation scenarios

Se aportan los datos de los pacientes incluidos en el estudio, los procedimientos clínicos a los que fueron sometidos, los efectos secundarios encontrados con el uso de tales medicamentos y la forma de evitarlos en la medida de lo posible o enfrentarse a ellos cuando se presentan.In this paper the authors review their first clinical experiences with a combination of an alpha-2 adrenergic central agonist: medetomidine and its reversal agent: atipamezole, in the sedation, premedication and anaesthesia of dogs and cats in clinical practice. The authors summarize data obtained from patients, the clinical procedures done, the undesirable side effects found with their use and which is their actual approach in arder to prevent them or how they manage those secondary effects when they re already present

    Estudio de los parámetros endoscópicos en 48 perros con colitis linfoplasmocitaria

    En el presente trabajo se realiza un estudio en cuarenta y ocho perros, sobre los hallazgos endoscópicos que caracterizan a la colitislinfoplasmocitaria. La colitis linfoplasmocitaria es una enfermedad difusa que afecta en mayor o menor grado a todas las porciones del colon. Sin embargo, los resultados del estudio demuestran que el colon descendente y el colon transverso tienen mayor predisposición a presentar úlceras y erosiones, y por tanto, también a la posterior presentación de zonas de fibrosis.In this paper the gross endoscopic appearance of lymphocytic-plasmocytic colitis was studied in forty-eight dogs. Lymphocytic-plasmocytic colitis is a díffuse disease involving all the colonic portions. Nevertheless, the results of our study showed that descending and transverse colon were particularly prone to develop ulcerative and consequent fibrotic lesions

    Top condensation model: a step towards the correct prediction of the Higgs mass

    A realization of the composite Higgs scenario in the context of the effective model with the SU(2)L×U(1)RSU(2)_L\times U(1)_R symmetric four-Fermi interactions proposed by Miransky, Tanabashi and Yamawaki is studied. The model implements Nambu's mechanism of dynamical electroweak symmetry breaking leading to the formation of tˉt\bar tt and bˉb\bar bb quark condensates. We explore the vacuum structure and spectrum of the model by using the Schwinger proper-time method. As a direct consequence of this mechanism, the Higgs acquires a mass in accord with its experimental value. The present prediction essentially differs from the known overestimated value, mχ=2mtm_\chi= 2m_t, making more favourable the top condensation scenario presented here. The mass formulas for the members of the second Higgs doublet are also obtained. The Nambu sum rule is discussed. It is shown that the anomalous U(1)AU(1)_A symmetry breaking modifies this rule at next to leading order in 1/Nc1/N_c.Comment: 14 pages, version to appear in EPJ

    Antibody Targeting Facilitates Effective Intratumoral SiRNA Nanoparticle Delivery to HER2-Overexpressing Cancer Cells

    The therapeutic potential of RNA interference (RNAi) has been limited by inefficient delivery of short interfering RNA (siRNA). Tumor-specific recognition can be effectively achieved by antibodies directed against highly expressed cancer cell surface receptors. We investigated the utility of linking an internalizing streptavidinconjugated HER2 antibody to an endosome-disruptive biotinylated polymeric nanocarrier to improve the functional cytoplasmic delivery of siRNA in breast and ovarian cancer cells in vitro and in an intraperitoneal ovarian cancer xenograft model in vivo, yielding an 80% reduction of target mRNA and protein levels with sustained repression for at least 96 hours. RNAi-mediated site specific cleavage of target mRNA was demonstrated using the 5\u27 RLM-RACE (RNA ligase mediated-rapid amplification of cDNA ends) assay. Mice bearing intraperitoneal human ovarian tumor xenografts demonstrated increased tumor accumulation of Cy5.5 fluorescently labeled siRNA and 70% target gene suppression after treatment with HER2 antibody-directed siRNA nanocarriers. Detection of the expected mRNA cleavage product by 5\u27 RLM-RACE assay confirmed that suppression occurs via the expected RNAi pathway. Delivery of siRNA via antibody-directed endosomolytic nanoparticles may be a promising strategy for cancer therapy

    Niemann-Pick disease type-B: a unique case report with compound heterozygosity and complicated lipid management

    BACKGROUND: Niemann-Pick disease (NPD) is a rare autosomal recessive hereditary disease characterized by deficient activity of acid sphingomyelinase. CASE PRESENTATION: We present a case of NPD type B with a unique compound heterozygosity for SMPD1 (NM_000543.4:c.[84delC];[96G¿>¿A]) in which both mutations that induce an early stop codon are located before the second in-frame initiation codon. The clinical presentation of the patient is compatible with NPD type B. She was initially diagnosed of Gaucher Disease, but her altered lipid profile led to a clinical suspicion of NPD. Combined high doses of atorvastatin and ezetimibe were given to treat the severe hypercholesterolemia. CONCLUSIONS: The pharmacological management of the lipid profile in these patients is important.     Biodegradable collagen matrix implant vs mitomycin-C as an adjuvant in trabeculectomy: a 24-month, randomized clinical trial

    AIM: To verify the safety and efficacy of Ologen (OLO) implant as adjuvant compared with low-dosage mitomycin-C (MMC) in trabeculectomy. METHODS: This was a prospective randomized clinical trial with a 24-month follow-up. Forty glaucoma patients (40 eyes) were assigned to trabeculectomy with MMC or OLO. Primary outcome includes target IOP at ≤21, ≤17, and ≤15 mm Hg; complete (target IOP without medications), and qualified success (target IOP regardless of medications). Secondary outcomes include bleb evaluation, according to Moorfields Bleb Grading System (MBGS); spectral domain optical coherence tomography (SD-OCT) examination; number of glaucoma medications; and frequency of postoperative adjunctive procedures and complications. RESULTS: The mean preoperative IOP was 26.5 (±5.2) in MMC and 27.3 (±6.0) in OLO eyes, without statistical significance. One-day postoperatively, the IOP dropped to 5.2 (±3.5) and 9.2 (±5.5) mm Hg, respectively (P=0.009). The IOP reduction was significant at end point in all groups (P=0.01), with a mean IOP of 16.0 (±2.9) and 16.5 (±2.1) mm Hg in MMC and OLO, respectively. The rates and Kaplan-Meier curves did not differ for both complete and qualified success at any target IOP. The bleb height in OLO group was higher than MMC one (P<0.05). SD-OCT analysis of successful/unsuccessful bleb in patients with or without complete success at IOP ≤17  mm Hg indicated a sensitivity of 83% and 73% and a specificity of 75% and 67%, respectively, for MMC and OLO groups. No adverse reaction to OLO was noted. CONCLUSIONS: Our results suggest that OLO implant could be a new, safe, and effective alternative to MMC, with similar long-term success rate

    An in vitro co-culture model of esophageal cells identifies ascorbic acid as a modulator of cell competition

    <p>Abstract</p> <p>Background</p> <p>The evolutionary dynamics between interacting heterogeneous cell types are fundamental properties of neoplastic progression but can be difficult to measure and quantify. Cancers are heterogeneous mixtures of mutant clones but the direct effect of interactions between these clones is rarely documented. The implicit goal of most preventive interventions is to bias competition in favor of normal cells over neoplastic cells. However, this is rarely explicitly tested. Here we have developed a cell culture competition model to allow for direct observation of the effect of chemopreventive or therapeutic agents on two interacting cell types. We have examined competition between normal and Barrett's esophagus cell lines, in the hopes of identifying a system that could screen for potential chemopreventive agents.</p> <p>Methods</p> <p>One fluorescently-labeled normal squamous esophageal cell line (EPC2-hTERT) was grown in competition with one of four Barrett's esophagus cell lines (CP-A, CP-B, CP-C, CP-D) under varying conditions and the outcome of competition measured over 14 days by flow cytometry.</p> <p>Results</p> <p>We demonstrate that ascorbic acid (vitamin C) can help squamous cells outcompete Barrett's cells in this system. We are also able to show that ascorbic acid's boost to the relative fitness of squamous cells was increased in most cases by mimicking the pH conditions of gastrointestinal reflux in the lower esophagus.</p> <p>Conclusions</p> <p>This model is able to integrate differential fitness effects on various cell types, allowing us to simultaneously capture effects on interacting cell types without having to perform separate experiments. This model system may be used to screen for new classes of cancer prevention agents designed to modulate the competition between normal and neoplastic cells.</p

    Reviving the Philippine Economy under a Responsible New Normal

    After the reclassification of areas under enhanced community quarantine (ECQ) to general community quarantine (GCQ), the urgent task for the Philippine government is to provide an exit plan to revive the Philippine economy. Given the significant economic damage resulting from the shutdown of roughly 75 percent of the country’s total production in the National Capital Region (NCR) and in the CALABARZON and Central Luzon areas, a gradual reopening of the economy will be necessary to prevent further economic damage that could not only be difficult to repair, but also long to overcome. Indeed, based on recent directives from the government, a substantial number of industries and services have thus been allowed to operate in both the ECQ and GCQ areas. However, as the Philippine government begins to calibrate the opening of sectors, there remain concerns as to how this process will affect jobs and livelihoods now and beyond. In this context, an economic recovery plan that talks about short-term, a transition, and full recovery phases— encompassing a revision of the current Philippine Development Plan without losing sight of the long-term goals envisioned in Ambisyon Natin 2040— is still needed. Indeed, a key component of AmBisyon 2040 has been of building resiliency over the long-term, which includes resiliency in health and economic shocks apart from natural disasters. At the same time, this recovery plan should also be accompanied by structural reforms to enhance its implementation. The Department of Finance has crafted a four-pillar socio-economic strategy aimed at: (a) supporting the more vulnerable sectors of society; (b) increasing medical resources to contain the virus and offer safety to front-liners; (c) keeping the economy afloat through financial emergency initiatives; and (d) creating jobs and sustaining the economy. Yet while enumerating the costs of these plans, the said strategy lacked details on how the country could achieve some of the goals without the availability of widespread testing and adequate health facilities. Loan guarantees, cash transfers, and other forms of subsidies can revive disrupted supply chains but cannot restore productivity in the middle of a persisting health crisis, while the uncertainty of a possible outbreak can keep workers from supplying goods and services. It is crucial to have these programs and institutions in place since a number of cities, regions and provinces have started to reopen. A modified community quarantine without the necessary health system investments, protection measures, and economic recovery plan risks amounting to an unregulated herd immunity strategy. Opting for herd immunity allows governments to blame the failure of the health and economic system on the virus, rather than on bad governance. Under current GCQ protocols, the burden on containing the virus is mostly transferred to the public. Unless the government provides mass testing, the problem of information is aggravated, probably raising the transmission risks. Moreover, unregulated herd immunity will be differentially felt by the poor. As healthy workers may recover their earnings from the modified quarantine, the poor, who have limited access to the health services and are thus more susceptible to the virus, are unlikely to benefit from this system. In effect, this will only exacerbate the inequality that prevails in the country. Moving towards a responsible new normal requires a strategy that addresses both people’s wellbeing and the socio-economic weaknesses exposed by COVID-19.
