    Bridging the Gap Between Science and Application: The Use of Cocreation Educational Workshops in Professional Youth Soccer

    We examined whether practice activities adopted by professional youth soccer coaches are modulated through the implementation of and engagement with cocreative evidence-based programs. Across two experiments, we used systematic observation to identify the practice activities of seven coaches across 134 sessions. In Experiment A, drill-based and games-based activities were recorded and quantified. To encourage behaviour change across the study, the systematic observation data were compared with skill acquisition literature to provide coaches with quantitative feedback and recommendations during workshops. Postworkshop systematic observation data indicated that practice activities used by coaches changed in accordance with the evidenced-based information (increase in games-based activities) delivered within the workshop. Interview data indicated that coaches typically stated that the workshop was a key reason for behaviour change. In a follow-up Experiment B, feedback and recommendations were delivered using an interactive video-based workshop. The systematic observation data indicated that coaches increased the use of soccer activities that contained active decision making with coaches citing the workshop as a key reason for behaviour change. These findings indicate that coaching practice activities can be supported and shaped through the implementation of cocreated workshops wherein coaches collaborate with sport scientists and researchers to bridge the gap between science and application

    Assessment of ROS Production in the Mitochondria of Live Cells

    Production of reactive oxygen species (ROS) in the mitochondria plays multiple roles in physiology, and excessive production of ROS leads to the development of various pathologies. ROS in the mitochondria are generated by various enzymes, mainly in the electron transporvt chain, and it is important to identify not only the trigger but also the source of free radical production. It is important to measure mitochondrial ROS in live, intact cells, because activation of ROS production could be initiated by changes in extramitochondrial processes which could be overseen when using isolated mitochondria. Here we describe the approaches, which allow to measure production of ROS in the matrix of mitochondria in live cells. We also demonstrate how to measure kinetic changes in lipid peroxidation in mitochondria of live cells. These methods could be used for understanding the mechanisms of pathology in a variety of disease models and also for testing neuro- or cardioprotective chemicals

    Interrogation of an Enzyme Library Reveals the Catalytic Plasticity of Naturally Evolved [4+2] Cyclases

One example is the Diels-Alder reaction, a [4+2] cycloaddition between a conjugated diene and a dienophile to form cyclohexenes. The development of biocatalysts for this reaction is paramount for unlocking sustainable routes to a plethora of important molecules. To obtain a comprehensive understanding of naturally evolved [4+2] cyclases, and to identify hitherto uncharacterised biocatalysts for this reaction, we constructed a library comprising forty-five enzymes with reported or predicted [4+2] cycloaddition activity. Thirty-one library members were successfully produced in recombinant form. In vitro assays employing a synthetic substrate incorporating a diene and a dienophile revealed broad-ranging cycloaddition activity amongst these polypeptides. The hypothetical protein Cyc15 was found to catalyse an intramolecular cycloaddition to generate a novel spirotetronate. The crystal structure of this enzyme, along with docking studies, establishes the basis for stereoselectivity in Cyc15, as compared to other spirotetronate cyclases

    Associations of Serum 25-Hydroxyvitamin D with Physical Performance and Bone Health in Overweight and Obese Older Adults

    Low vitamin D status commonly accompanies obesity, and both vitamin D deficiency and obesity have been associated with falls and fracture risk in older adults. We aimed to determine the associations of serum 25-hydroxyvitamin D (25(OH)D) concentrations with physical performance and bone health in community-dwelling, overweight and obese older men and women. Serum 25(OH)D concentrations were measured in 84 participants with body mass index ≥25 kg/m² (mean ± SD age 62.4 ± 7.9 years; 55% women). Physical function was determined by short physical performance battery, hand grip and quadriceps strength, and stair climb power tests. Body composition and bone structure were assessed by dual-energy X-ray absorptiometry and peripheral quantitative computed tomography, respectively. Mean ± SD 25(OH)D was 49.6 ± 17.7 nmol/L, and 50% of participants had low 25(OH)D ( 0.05). Lower 25(OH)D concentrations are associated with poorer quadricep strength and muscle power in overweight and obese older women but not men

    Effects of a brief mindfulness-based intervention on emotional regulation and levels of mindfulness in senior students

    Mindfulness-based interventions have been applied in diverse populations and achieved mental health benefits. This study examined the effects of a brief mindfulness program for emotional regulation and levels of mindfulness on senior students in Brazil. The intervention consisted of six weekly meetings attended by 30 participants. It is a pre-experimental research, with pre- and post-test comparative and correlation measurements. The preliminary results, which relied on parametrical and non-parametrical tests, revealed a reduction in total emotional regulation difficulties (p = 0.0001; r = − 0.55). Also, there was an increase in the levels of mindfulness in the subtests for both dimensions under evaluation: “Awareness” (p = 0.0001; d = 0.77) and “Acceptance” (p = 0.048; d = 0.37). By associating the amount of meditative practices performed by students with the variables, a significant positive correlation was found with the mindfulness dimension “Awareness” (rP = 0.422; p = 0.020), and there was a significant negative correlation with Difficulties in emotion regulation (rS = − 0.478; p = 0.008) and with its respective subscales “Non-acceptance” (rS = − 0.654; p = 0.0001) and “Clarity” (rS = − 0.463; p = 0.010). In conclusion, the application of a brief mindfulness-based intervention is promising in Brazilian university contexts; moreover, it can bring benefits to students, e.g., an increase in emotion regulation as well as in levels of mindfulness.     The multiscale hierarchical structure of Heloderma suspectum osteoderms and their mechanical properties

    Osteoderms are hard tissues embedded in the dermis of vertebrates and have been suggested to be formed from several different mineralized regions. However, their nano architecture and micro mechanical properties had not been fully characterized. Here, using electron microscopy, µ-CT, atomic force microscopy and finite element simulation, an in-depth characterization of osteoderms from the lizard Heloderma suspectum, is presented. Results show that osteoderms are made of three different mineralized regions: a dense apex, a fibre-enforced region comprising the majority of the osteoderm, and a bone-like region surrounding the vasculature. The dense apex is stiff, the fibre-enforced region is flexible and the mechanical properties of the bone-like region fall somewhere between the other two regions. Our finite element analyses suggest that when combined into the osteoderm structure, the distinct tissue regions are able to shield the body of the animal by dampening the external forces. These findings reveal the structure-function relationship of the Heloderma suspectum osteoderm in unprecedented detail

    A multiple spacecraft detection of the 2 April 2022 M-class flare and filament eruption during the first close Solar Orbiter perihelion

    CONTEXT: The Solar Orbiter mission completed its first remote-sensing observation windows in the spring of 2022. On 2 April 2022, an M-class flare followed by a filament eruption was seen both by the instruments on board the mission and from several observatories in Earth’s orbit, providing an unprecedented view of a flaring region with a large range of observations. AIMS: We aim to understand the nature of the flaring and filament eruption events via the analysis of the available dataset. The complexity of the observed features is compared with the predictions given by the standard flare model in 3D. METHOD: In this paper, we use the observations from a multi-view dataset, which includes extreme ultraviolet (EUV) imaging to spectroscopy and magnetic field measurements. These data come from the Interface Region Imaging Spectrograph, the Solar Dynamics Observatory, Hinode, as well as several instruments on Solar Orbiter. RESULTS: The large temporal coverage of the region allows us to analyse the whole sequence of the filament eruption starting with its pre-eruptive state. Information given by spectropolarimetry from SDO/HMI and Solar Orbiter PHI/HRT shows that a parasitic polarity emerging underneath the filament is responsible for bringing the flux rope to an unstable state. As the flux rope erupts, Hinode EIS captures blue-shifted emission in the transition region and coronal lines in the northern leg of the flux rope prior to the flare peak. This may be revealing the unwinding of one of the flux rope legs. At the same time, Solar Orbiter SPICE captures the whole region, complementing the Doppler diagnostics of the filament eruption. Analyses of the formation and evolution of a complex set of flare ribbons and loops, of the hard and soft X-ray emissions with STIX, show that the parasitic emerging bipole plays an important role in the evolution of the flaring region. CONCLUSIONS: The extensive dataset covering this M-class flare event demonstrates how important multiple viewpoints and varied observations are in order to understand the complexity of flaring regions. While the analysed data are overall consistent with the standard flare model, the present particular magnetic configuration shows that surrounding magnetic activity such as nearby emergence needs to be taken into account to fully understand the processes at work. This filament eruption is the first to be covered from different angles by spectroscopic instruments, and provides an unprecedented diagnostic of the multi-thermal structures present before and during the flare.
