Local three-nucleon interaction from chiral effective field theory
The three-nucleon (NNN) interaction derived within the chiral effective field
theory at the next-to-next-to-leading order (N2LO) is regulated with a function
depending on the magnitude of the momentum transfer. The regulated NNN
interaction is then local in the coordinate space, which is advantages for some
many-body techniques. Matrix elements of the local chiral NNN interaction are
evaluated in a three-nucleon basis. Using the ab initio no-core shell model
(NCSM) the NNN matrix elements are employed in 3H and 4He bound-state
calculations.Comment: 17 pages, 9 figure
Incorporation of doxorubicin in different polymer nanoparticles and their anticancer activity
Background: Nanoparticles are under investigation as carrier systems for anticancer drugs. The expression of efflux transporters such as the ATP-binding cassette (ABC) transporter ABCB1 is an important resistance mechanism in therapy-refractory cancer cells. Drug encapsulation into nanoparticles has been shown to bypass efflux-mediated drug resistance, but there are also conflicting results. To investigate whether easy-to-prepare nanoparticles made of well-tolerated polymers may circumvent transporter-mediated drug efflux, we prepared poly(lactic-co-glycolic acid) (PLGA), polylactic acid (PLA), and PEGylated PLGA (PLGA-PEG) nanoparticles loaded with the ABCB1 substrate doxorubicin by solvent displacement and emulsion diffusion approaches and assessed their anticancer efficiency in neuroblastoma cells, including ABCB1-expressing cell lines, in comparison to doxorubicin solution.
Results: The resulting nanoparticles covered a size range between 73 and 246 nm. PLGA-PEG nanoparticle preparation by solvent displacement led to the smallest nanoparticles. In PLGA nanoparticles, the drug load could be optimised using solvent displacement at pH 7 reaching 53 µg doxorubicin/mg nanoparticle. These PLGA nanoparticles displayed sustained doxorubicin release kinetics compared to the more burst-like kinetics of the other preparations. In neuroblastoma cells, doxorubicin-loaded PLGA-PEG nanoparticles (presumably due to their small size) and PLGA nanoparticles prepared by solvent displacement at pH 7 (presumably due to their high drug load and superior drug release kinetics) exerted the strongest anticancer effects. However, nanoparticle-encapsulated doxorubicin did not display increased efficacy in ABCB1-expressing cells relative to doxorubicin solution.
Conclusion: Doxorubicin-loaded nanoparticles made by different methods from different materials displayed substantial discrepancies in their anticancer activity at the cellular level. Optimised preparation methods resulted in PLGA nanoparticles characterised by increased drug load, controlled drug release, and high anticancer efficacy. The design of drug-loaded nanoparticles with optimised anticancer activity at the cellular level is an important step in the development of improved nanoparticle preparations for anticancer therapy. Further research is required to understand under which circumstances nanoparticles can be used to overcome efflux-mediated resistance in cancer cells
Spin-orbit induced backflow in neutron matter with auxiliary field diffusion Monte Carlo
The energy per particle of zero-temperature neutron matter is investigated,
with particular emphasis on the role of the interaction. An
analysis of the importance of explicit spin--orbit correlations in the
description of the system is carried out by the auxiliary field diffusion Monte
Carlo method. The improved nodal structure of the guiding function, constructed
by explicitly considering these correlations, lowers the energy. The proposed
spin--backflow orbitals can conveniently be used also in Green's Function Monte
Carlo calculations of light nuclei.Comment: 4 pages, 1 figur

Neutron matter at zero temperature with auxiliary field diffusion Monte Carlo
The recently developed auxiliary field diffusion Monte Carlo method is
applied to compute the equation of state and the compressibility of neutron
matter. By combining diffusion Monte Carlo for the spatial degrees of freedom
and auxiliary field Monte Carlo to separate the spin-isospin operators, quantum
Monte Carlo can be used to simulate the ground state of many nucleon systems
(A\alt 100). We use a path constraint to control the fermion sign problem. We
have made simulations for realistic interactions, which include tensor and
spin--orbit two--body potentials as well as three-nucleon forces. The Argonne
and two nucleon potentials plus the Urbana or Illinois
three-nucleon potentials have been used in our calculations. We compare with
fermion hypernetted chain results. We report results of a Periodic Box--FHNC
calculation, which is also used to estimate the finite size corrections to our
quantum Monte Carlo simulations. Our AFDMC results for models of pure
neutron matter are in reasonably good agreement with equivalent Correlated
Basis Function (CBF) calculations, providing energies per particle which are
slightly lower than the CBF ones. However, the inclusion of the spin--orbit
force leads to quite different results particularly at relatively high
densities. The resulting equation of state from AFDMC calculations is harder
than the one from previous Fermi hypernetted chain studies commonly used to
determine the neutron star structure.Comment: 15 pages, 15 tables and 5 figure
Doxorubicin-Loaded Human Serum Albumin Nanoparticles Overcome Transporter-Mediated Drug Resistance in Drug-Adapted Cancer Cells
Resistance to systemic drug therapy is a major reason for the failure of anticancer therapies. Here, we tested doxorubicin-loaded human serum albumin (HSA) nanoparticles in the neuroblastoma cell line UKF-NB-3 and its ABCB1-expressing sublines adapted to vincristine (UKF-NB-3rVCR1) and doxorubicin (UKF-NB-3rDOX20). Doxorubicin-loaded nanoparticles displayed increased anticancer activity in UKF-NB-3rVCR1 and UKF-NB-3rDOX20 cells relative to doxorubicin solution, but not in UKF-NB-3 cells. UKF-NB-3rVCR1 cells were re-sensitised by nanoparticle-encapsulated doxorubicin to the level of UKF-NB-3 cells. UKF-NB-3rDOX20 cells displayed a more pronounced resistance phenotype than UKF-NB-3rVCR1 cells and were not re-sensitised by doxorubicin-loaded nanoparticles to the level of parental cells. ABCB1 inhibition using zosuquidar resulted in similar effects like nanoparticle incorporation, indicating that doxorubicin-loaded nanoparticles successfully circumvent ABCB1-mediated drug efflux. The limited re-sensitisation of UKF-NB-3rDOX20 cells to doxorubicin by circumvention of ABCB1-mediated efflux is probably due to the presence of multiple doxorubicin resistance mechanisms. So far, ABCB1 inhibitors have failed in clinical trials probably because systemic ABCB1 inhibition results in a modified body distribution of its many substrates including drugs, xenobiotics, and other molecules. HSA nanoparticles may provide an alternative, more specific way to overcome transporter-mediated resistance
Ground-state correlations and final state interactions in the process 3He(e,e'pp)n
The two-proton emission process is theoretically investigated
using realistic three-nucleon wave functions and taking the final state
interaction into account by an approach which can be used when the value of the
three-nucleon invariant mass is either below or above the pion emission
threshold. Various kinematical conditions which enhance or minimize the effects
of the final state interaction are thoroughly analyzed.Comment: 26 pages, 12 eps-figures. Introduction and abstract updated, few

Subcutaneous rupture of the Achilles tendon and ipsilateral fracture of the medial malleolus
BACKGROUND: Although ankle fractures and an Achilles tendon rupture are relatively frequent in isolation, their association in the same injury is uncommon. CASE PRESENTATION: A 38 year old male tree surgeon fell six meters from a tree, sustaining a subcutaneous rupture of the Achilles tendon and an ipsilateral closed fracture of the medial malleolus. The injuries were diagnosed following clinical examination and imaging. CONCLUSION: This injury combination is infrequent, and management of the Achilles tendon rupture should take into account the necessity not to secondarily displace the fracture of the medial malleollus
Femtometer Toroidal Structures in Nuclei
The two-nucleon density distributions in states with isospin , spin
=1 and projection =0 and 1 are studied in H, He,
Li and O. The equidensity surfaces for =0 distributions are
found to be toroidal in shape, while those of =1 have dumbbell shapes
at large density. The dumbbell shapes are generated by rotating tori. The
toroidal shapes indicate that the tensor correlations have near maximal
strength at fm in all these nuclei. They provide new insights and simple
explanations of the structure and electromagnetic form factors of the deuteron,
the quasi-deuteron model, and the , and =2 (-wave)
components in He, He and Li. The toroidal distribution has a
maximum-density diameter of 1 fm and a half-maximum density thickness of
0.9 fm. Many realistic models of nuclear forces predict these values,
which are supported by the observed electromagnetic form factors of the
deuteron, and also predicted by classical Skyrme effective Lagrangians, related
to QCD in the limit of infinite colors. Due to the rather small size of this
structure, it could have a revealing relation to certain aspects of QCD.Comment: 35 pages in REVTeX, 25 PostScript figure
