Pre-implantation mouse embryos cultured In vitro under different oxygen concentrations show altered ultrastructures
Abstract
Assisted Reproductive Technologies routinely utilize different culture media and oxygen (O2) concentrations to culture human embryos. Overall, embryos cultured under physiological O2 tension (5%) have improved development compared to embryos cultured under atmospheric O2 conditions (20%). The mechanisms responsible for this remain unclear. This study aimed to evaluate the effect of physiologic (5%) or atmospheric O2 (20%) tension on the microscopic ultrastructure of pre-implantation mouse embryos using Transmission Electron Microscopy (TEM). Embryos flushed out of the uterus after natural mating were used as the control. For use as the control, 2-cells, 4-cells, morulae, and blastocysts were flushed out of the uterus after natural fertilization. In vitro fertilization (IVF) was performed using potassium simplex optimized medium (KSOM) under different O2 tensions (5% and 20%) until the blastocyst stage. After collection, embryos were subjected to the standard preparative for light microscopy (LM) and TEM. We found that culture in vitro under 5% and 20% O2 results in an increase of vacuolated shaped mitochondria, cytoplasmic vacuolization and presence of multi-vesicular bodies at every embryonic stage. In addition, blastocysts generated by IVF under 5% and 20% O2 showed a lower content of heterochromatin, an interruption of the trophectodermal and inner cell mass cell membranes, an increased density of residual bodies, and high levels of glycogen granules in the cytoplasm. In conclusion, this study suggests that in vitro culture, particularly under atmospheric O2 tension, causes stage-specific changes in preimplantation embryo ultrastructure. In addition, atmospheric (20%) O2 is associated with increased alterations in embryonic ultrastructure; these changes may explain the reduced embryonic development of embryos cultured with 20% O2
Hydro-socio-economic implications for water management strategies: the case of Roussillon coastal aquifer
In many Mediterranean coastal areas, agriculture, drinking water supply, tourism and industry strongly depend on the available groundwater resources. As a result of the significant economic development during the last three decades along the coast, abstractions from coastal aquifers have increased tremendously, frequently leading to overexploitation and saltwater intrusion. Geological, hydrogeological and socio-economical studies as a multidisciplinary approach on a coastal Mediterranean aquifer- the Roussillon - have been carried out in order to design effective water management strategies on areas sensitive to seawater intrusion risk. Geology provides geometry and architecture of the different aquifers, hydrogeology assess the seawater intrusion risk while socio-economic study includes consulting the stakeholders with the aim of suggesting water management and policy option acceptable to the majority of population. This paper then highlights the economic interests at stake, diversity of viewpoints expressed by stakeholders and political dimension of the issue, which are likely to be encountered for all similar situations on both sides of the Mediterranean Sea.GESTION DE L'EAU;STRATEGIE;HYDROGEOLOGIE;SOCIOLOGIE;ECONOMIE;MER MEDITERRANEE;ROUSSILLON
EVALUATION OF PHOTOGRAMMETRIC BLOCK ORIENTATION USING QUALITY DESCRIPTORS FROM STATISTICALLY FILTERED TIE POINTS
Due to the increasing number of low-cost sensors, widely accessible on the market, and because of the supposed granted correctness of the semi-automatic workflow for 3D reconstruction, highly implemented in the recent commercial software, more and more users operate nowadays without following the rigorousness of classical photogrammetric methods. This behaviour often naively leads to 3D products that lacks metric quality assessment. This paper proposes and analyses an approach that gives the users the possibility to preserve the trustworthiness of the metric information inherent in the 3D model, without sacrificing the automation offered by modern photogrammetry software. At the beginning, the importance of Data Quality Assessment is outlined, together with some recall of photogrammetry best practices. With the purpose of guiding the user through a correct pipeline for a certified 3D model reconstruction, an operative workflow is proposed, focusing on the first part of the object reconstruction steps (tie-points extraction, camera calibration, and relative orientation). A new GUI (Graphical User Interface) developed for the open source MicMac suite is then presented, and a sample dataset is used for the evaluation of the photogrammetric block orientation using statistically obtained quality descriptors. The results and the future directions are then presented and discussed
The effect of follicular fluid hormones on oocyte recovery after ovarian stimulation: FSH level predicts oocyte recovery.
BackgroundOvarian stimulation for assisted reproductive technology (ART) overcomes the physiologic process to develop a single dominant follicle. However, following stimulation, egg recovery rates are not 100%. The objective of this study is to determine if the follicular fluid hormonal environment is associated with oocyte recovery.MethodsThis is a prospective study involving patients undergoing ART by standard ovarian stimulation protocols at an urban academic medical center. A total of 143 follicular fluid aspirates were collected from 80 patients. Concentrations of FSH, hCG, estradiol, progesterone, testosterone and prolactin were determined. A multivariable regression analysis was used to investigate the relationship between the follicular fluid hormones and oocyte recovery.ResultsIntrafollicular FSH was significantly associated with oocyte recovery after adjustment for hCG (Adjusted odds ratio (AOR) = 1.21, 95%CI 1.03-1.42). The hCG concentration alone, in the range tested, did not impact the odds of oocyte recovery (AOR = 0.99, 95%CI 0.93-1.07). Estradiol was significantly associated with oocyte recovery (AOR = 0.98, 95% CI 0.96-0.99). After adjustment for progesterone, the strength of association between FSH and oocyte recovery increased (AOR = 1.84, 95%CI 1.45-2.34).ConclusionThe relationship between FSH and oocyte recovery is significant and appears to work through mechanisms independent of the sex hormones. FSH may be important for the physiologic event of separation of the cumulus-oocyte complex from the follicle wall, thereby influencing oocyte recovery. Current methods for inducing the final stages of oocyte maturation, with hCG administration alone, may not be optimal. Modifications of treatment protocols utilizing additional FSH may enhance oocyte recovery

The idea of this joint article involving digital humanists and heritage researchers is to examine communities, concepts and research applications as well as shared challenges. Beyond a collection of problem-centred essays this is intended to initiate a fruitful discussion about commonalities and differences between both scholarly fields as well as to assess to which extent they are two sides of the same medal
Participatory analysis for adaptation to climate change in Mediterranean agricultural systems: possible choices in process design (versĂŁo Pre Print)
There is an increasing call for local measures to adapt to climate change, based on foresight analyses in collaboration with actors. However, such analyses involve many challenges, particularly because the actors concerned may not consider climate change to be an urgent concern. This paper examines the methodological choices made by three research teams in the design and implementation of participatory foresight analyses to explore agricultural and water management options for adaptation to climate change. Case studies were conducted in coastal areas of France, Morocco, and Portugal where the groundwater is intensively used for irrigation, the aquifers are at risk or are currently overexploited, and a serious agricultural crisis is underway. When designing the participatory processes, the researchers had to address four main issues: whether to avoid or prepare dialogue between actors whose relations may be limited or tense; how to select participants and get them involved; how to facilitate discussion of issues that the actors may not initially consider to be of great concern; and finally, how to design and use scenarios. In each case, most of the invited actors responded and met to discuss and evaluate a series of scenarios. Strategies were discussed at different levels, from farming practices to aquifer management. It was shown that such participatory analyses can be implemented in situations which may initially appear to be unfavourable. This was made possible by the flexibility in the methodological choices, in particular the possibility of framing the climate change issue in a broader agenda for discussion with the actors
Efecto de la producción láctea en el desarrollo de endometritis en vacas lecheras de Santa Fe (Argentina)
Se planteĂł como objetivo del presente trabajo evaluar la incidencia de la producciĂłn de leche individual en el desarrollo de endometritis subclĂnica en el posparto de vacas lecheras Holstein de la provincia de Santa Fe, Argentina. Se utilizaron 274 vacas de las cuales 222 fueron diagnosticadas como sanas y 52 padeciendo endometritis subclĂnica. Dicha enfermedad se diagnosticĂł cuando una vaca manifestaba en su mucosa uterina un porcentaje de polimorfonucleares neutrĂłfilos ≥ 5%, obtenido con la tĂ©cnica complementaria conocida como cytobrush. Las variables productivas analizadas fueron la condiciĂłn corporal, el nĂşmero de partos y los litros totales de leche producida. No se obtuvieron diferencias estadĂsticamente significativas en las variables entre las vacas sanas y las vacas con endometritis subclĂnica. Se concluye que las variables productivas como el estado corporal, el nĂşmero de partos y la producciĂłn láctea, no influencian el desarrollo de endometritis subclĂnica
