    Accelerated Epigenetic Aging in Older Adults With HIV Disease: Associations With Serostatus, HIV Clinical Factors, and Health Literacy

    The prevalence of older persons with HIV (PWH) disease has increased considerably in the last 20 years, but our understanding of biological factors of aging and their clinical correlates among PWH remains limited. Study participants were 149 persons aged 50 and older, including 107 PWH and 42 seronegatives. All participants completed a blood draw, research medical evaluation, structured psychiatric interview, neurocognitive assessment, questionnaires, and measures of health literacy. Four epigenetic clocks were generated from stored blood samples using standardized laboratory methods. In regression models adjusting for sex and smoking status, PWH had significantly higher epigenetic aging acceleration values than seronegatives on all four indicators. Within the PWH sample, higher levels of epigenetic aging acceleration were moderately associated with lower current CD4 count, AIDS diagnoses, higher scores on the Veterans Aging Cohort Study Index, and lower telomere values. Higher epigenetic aging acceleration indices were also associated with lower health literacy among PWH. PWH experience accelerated aging as measured by a series of epigenetic clocks, which may be linked to immune compromise and risk of all-cause mortality. Health literacy may be a modifiable target for mitigating the risk of accelerated aging among older PWH

    Averaged Dynamic Modeling and Control of a Quasi-Z-Source Inverter for Wind Power Applications

    Typically, permanent magnet synchronous generator (PMSG)-driven wind turbines (WTs) present a two-stage power converter topology based on a DC/DC boost converter and voltage source inverter. In this study, this configuration is substituted by a quasi-Z-source inverter (qZSI), which is an attractive solution for boosting and converting the voltage from DC to AC in a single stage. A 2 MW PMSG WT with qZSI was studied herein. A switched dynamic model (SDM) of the qZSI (including the modeling of all switches and firing pulses) is not recommended for steady-state stability studies, long-term simulations, or large electric power systems. For such studies, two averaged dynamic models are proposed in this work. Both models present the same control system as the SDM, except for the generation of firing pulses, which is not necessary in the averaged models.     An Outbreak of Hepatic Veno-Occlusive Disease in Western Afghanistan Associated with Exposure to Wheat Flour Contaminated with Pyrrolizidine Alkaloids

    Pyrrolizidine alakloids (PAs) are known to cause hepatic veno-occlusive disease (VOD). Outbreaks have occurred in Western Afghanistan since 1974, the latest in February 2008. We conducted an outbreak investigation using a case-control design. Sixty-seven cases of VOD were compared with 199 community controls. Consumption of bread was strongly associated with disease (adjusted odds ratio: 35.8 [95%CI: 7.6–168.2]). Toxic doses of PA were found in plant extracts and in samples of wheat flour taken from the study area. Compared to wheat flour there was 1000 times less PA in milk and whey and in water samples the PA content was zero. Although direct analysis was not possible, contaminated wheat flour used to make bread was the likely source of PA causing the outbreak. Eating a more varied diet including meat and fruit may be protective. Prevention and control measures will rely on community awareness and agricultural interventions to ensure safety of the food supply

    Optimal energy management system using biogeography based optimization for grid-connected MVDC microgrid with photovoltaic, hydrogen system, electric vehicles and Z-source converters

    Currently, the technology associated with charging stations for electric vehicles (EV) needs to be studied and improved to further encourage its implementation. This paper presents a new energy management system (EMS) based on a Biogeography-Based Optimization (BBO) algorithm for a hybrid EV charging station with a configuration that integrates Z-source converters (ZSC) into medium voltage direct current (MVDC) grids. The EMS uses the evolutionary BBO algorithm to optimize a fitness function defining the equivalent hydrogen consumption/generation. The charging station consists of a photovoltaic (PV) system, a local grid connection, two fast charging units and two energy storage systems (ESS), a battery energy storage (BES) and a complete hydrogen system with fuel cell (FC), electrolyzer (LZ) and hydrogen tank. Through the use of the BBO algorithm, the EMS manages the energy flow among the components to keep the power balance in the system, reducing the equivalent hydrogen consumption and optimizing the equivalent hydrogen generation. The EMS and the configuration of the charging station based on ZSCs are the main contributions of the paper. The behaviour of the EMS is demonstrated with three EV connected to the charging station under different conditions of sun irradiance. In addition, the proposed EMS is compared with a simpler EMS for the optimal management of ESS in hybrid configurations. The simulation results show that the proposed EMS achieves a notable improvement in the equivalent hydrogen consumption/generation with respect to the simpler EMS.     Anxiolytic and anti-stress effects of acute administration of acetyl-L-carnitine in zebrafish

    Studies have suggested that oxidative stress may contribute to the pathogenesis of mental disorders. In this context, molecules with antioxidant activity may be promising agents in the treatment of these deleterious conditions. Acetyl-L-carnitine (ALC) is a multi-target molecule that modulates the uptake of acetyl-CoA into the mitochondria during fatty acid oxidation, acetylcholine production, protein, and membrane phospholipid synthesis, capable of promoting neurogenesis in case of neuronal death. Moreover, neurochemical effects of ALC include modulation of brain energy and synaptic transmission of multiple neurotransmitters, including expression of type 2 metabotropic glutamate (mGlu2) receptors. The aim of this study was to investigate the effects of ALC in zebrafish by examining behavioral and biochemical parameters relevant to anxiety and mood disorders in zebrafish. ALC presented anxiolytic effects in both novel tank and light/dark tests and prevented the anxiety-like behavior induced by an acute stressor (net chasing). Furthermore, ALC was able to prevent the lipid peroxidation induced by acute stress in the zebrafish brain. The data presented here warrant further investigation of ALC as a potential agent in the treatment of neuropsychiatric disorders. Its good tolerability also subsidizes the additional studies necessary to assess its therapeutic potential in clinical settings

    Genotipagem de Toxoplasma gondii em galinhas domésticas em uma área rural do Rio Grande do Sul, Brasil

    Free-range chickens may ingest oocysts of T. gondii present in the environment and consequently harbor virulent strains of this parasite in different tissues, without any clinical signs. Isolation of T. gondii through bioassays on mice and cats from naturally infected chicken tissues has been described in several countries, demonstrating the importance of free-range chickens in the transmission of this parasite. The aim of this study was the genotypic characterization of T. gondii isolates obtained from naturally infected free-range chickens in a rural area of the state of Rio Grande do Sul, Brazil. Brain and heart tissue from 12 chickens seropositive for T. gondii were processed using peptic digestion technique for parasite isolation. From 12 samples subjected to mouse bioassay, nine isolates were obtained. RFLP-PCR genotypic characterization was performed using 11 genetic markers: SAG1, 5'-3'SAG2, alt.SAG2, SAG3, BTUB, GRA6, c22-8, c29- 2, L358, PK1 and Apico. Genetic characterization of the isolates revealed the presence of five atypical genotypes according to ToxoDB (# 11, # 55, # 64, # 140 and # 163). Our results showed a wide genetic diversity of T. gondii in free-range chickens in this region.Galinhas criadas ao ar livre podem ingerir oocistos de T. gondii presentes no ambiente e, com isso, albergar cepas virulentas desse parasita em diferentes tecidos, sem sinais clínicos. O isolamento de T. gondii por meio de bioensaios em camundongos e gatos, a partir de tecidos de galinhas naturalmente infectadas, tem sido descrito em vários países. Isso demonstra a importância das galinhas caipiras na epidemiologia desse parasita. O objetivo deste trabalho foi caracterizar genotipicamente isolados de T. gondii obtidos de galinhas caipiras naturalmente infectadas em uma área rural do município de Santa Maria, estado do Rio Grande do Sul, Brasil. Fragmentos de cérebro e de coração, de 12 galinhas soropositivas para T. gondii, foram processados pela técnica de digestão péptica para isolamento do parasita. Das 12 amostras submetidas a bioensaio com camundongos, nove isolados foram obtidos. A caracterização genotípica por RFLP-PCR foi realizada utilizando-se 11 marcadores genéticos: SAG1, 5'- 3'SAG2, alt.SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1 e Apico e revelou a presença de cinco genótipos atípicos de acordo com o ToxoDB (# 11, # 55, # 64, # 140 e # 163).     A multilevel analysis of the association between social networks and support on leisure time physical activity: evidence from 40 disadvantaged areas in London

    There has been extensive conceptual and empirical work on the associations between social relations and health in recent years. However, the specific pathways through which social interactions impact on health have not been fully elucidated. The aim of this paper is to estimate associations between leisure time physical activity (LTPA) and social networks and support. Using data from a cross-sectional household survey in 40 disadvantaged Lower Super Output Areas in London, we applied a multilevel model to investigate psychosocial and environmental determinants of physical activity in these populations. Our findings present a strong case for the influence of individual-level social networks on the level of LTPA, although the associations between the types of social support and LTPA were insignificant. We also found that crime rate was an important area-level correlates of LTPA
