    Preservice Science Teachers\u27 Experiences with Repeated, Guided Inquiry

    The purpose of this study was to examine preservice science teachers’ experiences with repeated scientific inquiry (SI) activities. The National Science Education Standards (National Research Council, 1996) stress students should understand and possess the abilities to do SI. For students to meet these standards, science teachers must understand and be able to perform SI; however, previous research demonstrated that many teachers have naïve understandings in this area. Teacher preparation programs provide an opportunity to facilitate the development of inquiry understandings and abilities. In this study, preservice science teachers had experiences with two inquiry activities that were repeated three times each. The research questions for this study were (a) How do preservice science teachers’ describe their experiences with repeated, guided inquiry activities? (b) What are preservice science teachers’ understandings and abilities of SI? This study was conducted at a large, urban university in the southeastern United States. The 5 participants had bachelor’s degrees in science and were enrolled in a graduate science education methods course. The researcher was one of the course instructors but did not lead the activities. Case study methodology was used. Data was collected from a demographic survey, an open-ended questionnaire with follow-up interviews, the researcher’s observations, participants’ lab notes, personal interviews, and participants’ journals. Data were coded and analyzed through chronological data matrices to identify patterns in participants’ experiences. The five domains identified in this study were understandings of SI, abilities to conduct SI, personal feelings about the experience, science content knowledge, and classroom implications. Through analysis of themes identified within each domain, the four conclusions made about these preservice teachers’ experiences with SI were that the experience increased their abilities to conduct inquiry, increased their understanding of how they might use SI in their classroom, increased their understanding of why variables are used in experiments, and did not increase their physics content knowledge. These conclusions suggest that preservice science teachers having repeated, guided experiences with inquiry increase their abilities to conduct SI and consider how inquiry could be used in their future science classrooms

    Risk and the School-to-Work Transition in East Germany and the United States.

    This study investigates how young adults in vocational education perceive risk in regard to their professional futures in East Germany and the United States. It analyzes students’ career aspirations and life plans in both countries and how they cope with uncertainties at the labor market. It further outlines underlying values, beliefs, and attitudes that guide young Americans and East Germans in their planning. Several theoretical frameworks ground this study and comprise the majority of the relevant literature. This cross-cultural comparative case study takes a mixed method approach using a concurrent triangulation design. The inquiry is framed by theories in the field of risk and cultural risk perception, school-to-work transition, vocational education and training, and welfare studies. In both countries, 129 students filled out a questionnaire. Additionally, narratives from nine focus groups and 29 biographical interviews were conducted. A three level analysis of the data was compiled that outlines the themes and categorizes them according to an individual, institutional, and macro-structural level of influence on risk perception in each country. Emerging premises on an individual level were choice, family and career planning concerns, geographical and occupational flexibility, further education and training, and agency. On the institutional level the influence of public career advisement institutions, teachers, parents, peers and friends was outlined. On the macro-structural level unemployment, political welfare reforms, the vocational education and training system were themes that have been of concern among the East German population. The dissertation also offers a comparative analysis of the data. This study reveals that young adults in East Germany are highly concerned about their occupational futures and tend to be pessimistic about current welfare reforms. They hold on to the idea of a standard biography and try to make strategic career plans. Their counterparts in the United States are highly optimistic about their futures, expressed little concern about labor market policies, but also appeared to be short term oriented in their life planning in order to remain flexible and mobile

    Factors influencing carbon stocks and accumulation rates in eelgrass meadows across New England, USA

    Increasing the protection of coastal vegetated ecosystems has been suggested as one strategy to compensate for increasing carbon dioxide (CO2) in the atmosphere as the capacity of these habitats to sequester and store carbon exceeds that of terrestrial habitats. Seagrasses are a group of foundation species that grow in shallow coastal and estuarine systems and have an exceptional ability to sequester and store large quantities of carbon in biomass and, particularly, in sediments. However, carbon stocks (Corg stocks) and carbon accumulation rates (Corg accumulation) in seagrass meadows are highly variable both spatially and temporally, making it difficult to extrapolate this strategy to areas where information is lacking. In this study, Corg stocks and Corg accumulation were determined at 11 eelgrass meadows across New England, representing a range of eutrophication and exposure conditions. In addition, the environmental factors and structural characteristics of meadows related to variation in Corg stocks were identified. The objectives were accomplished by assessing stable isotopes of δ13C and δ15N as well as %C and %N in plant tissues and sediments, measuring grain size and 210Pb of sediment cores, and through assessing site exposure. Variability in Corg stocks in seagrass meadows is well predicted using commonly measured environmental variables such as grain size distribution. This study allows incorporation of data and insights for the northwest Atlantic, where few studies on carbon sequestration by seagrasses have been conducted

    AIM2 Inflammasome Activation Leads to IL-1α and TGF-β Release From Exacerbated Chronic Obstructive Pulmonary Disease-Derived Peripheral Blood Mononuclear Cells

    Chronic obstructive pulmonary disease (COPD) is now the fourth-leading cause of death worldwide and its prevalence is increasing. The progressive decline of lung function and airway remodelling are a consequence of chronic inflammatory responses. It was recently postulated the involvement of the inflammasome in COPD, although the underlying mechanism/s still need to be elucidated. Therefore, we isolated peripheral blood mononuclear cells (PBMCs) from exacerbated/unstable COPD patients. The stimulation of PBMCs with an AIM2 inflammasome activator, Poly dA:dT, led to IL-1α, but not IL-1β, release. The release of this cytokine was caspase-1- and caspase-4-dependent and correlated to higher levels of 8-OH-dG in COPD compared to non-smoker and smoker-derived PBMCs. Interestingly, AIM2-depedent IL-1α release was responsible for higher TGF-β levels, crucial mediator during pro-fibrotic processes associated to COPD progression. In conclusion, our data highlight the involvement of AIM2/caspase-1/caspase-4 in IL-1α-induced TGF-β release in unstable COPD-derived PBMCs, opening new therapeutic perspectives for unstable COPD patients

    Modelling hCDKL5 heterologous expression in bacteria

    hCDKL5 refers to the human cyclin-dependent kinase like 5 that is primarily expressed in the brain. Mutations in its coding sequence are often causative of hCDKL5 deficiency disorder, a devastating neurodevelopmental disorder currently lacking a cure. The large-scale recombinant production of hCDKL5 is desirable to boost the translation of preclinical therapeutic approaches into the clinic. However, this is hampered by the intrinsically disordered nature of almost two-thirds of the hCDKL5 sequence, making this region more susceptible to proteolytic attack, and the observed toxicity when the enzyme is accumulated in the cytoplasm of eukaryotic host cells. The bacterium Pseudoalteromonas haloplanktis TAC125 (PhTAC125) is the only prokaryotic host in which the full-length production of hCDKL5 has been demonstrated. To date, a system-level understanding of the metabolic burden imposed by hCDKL5 production is missing, although it would be crucial for upscaling of the production process. Here, we combined experimental data on protein production and nutrients assimilation with metabolic modelling to infer the global consequences of hCDKL5 production in PhTAC125 and to identify potential overproduction targets. Our analyses showed a remarkable accuracy of the model in simulating the recombinant strain phenotype and also identified priority targets for optimised protein production

    Species traits and geomorphic setting as drivers of global soil carbon stocks in seagrass meadows

    Our knowledge of the factors that can influence the stock of organic carbon (OC) that is stored in the soil of seagrass meadows is evolving, and several causal effects have been used to explain the variation of stocks observed at local to national scales. To gain a global-scale appreciation of the drivers that cause variation in soil OC stocks, we compiled data on published species-specific traits and OC stocks from monospecific and mixed meadows at multiple geomorphological settings. Species identity was recognized as an influential driver of soil OC stocks, despite their large intraspecific variation. The most important seagrass species traits associated with OC stocks were the number of leaves per seagrass shoot, belowground biomass, leaf lifespan, aboveground biomass, leaf lignin, leaf breaking force and leaf OC plus the coastal geomorphology of the area, particularly for lagoon environments. A revised estimate of the global average soil OC stock to 20 cm depth of 15.4 Mg C ha−1 is lower than previously reported. The largest stocks were still recorded in Mediterranean seagrass meadows. Our results specifically identify Posidonia oceanica from the Mediterranean and, more generally, large and persistent species as key in providing climate regulation services, and as priority species for conservation for this specific ecosystem service
