    Anastomosis grouping and genetic diversity analysis of Rhizoctonia solani isolates causing wet root rot in chickpea

    Rhizoctonia solani is considered as one of the most destructive soil-and-seed borne plant pathogens infecting various agricultural crops including chickpea. The 50 chickpea isolates of R. solani representing 10 different states of India were variable in hyphal anastomosis reactions and they were grouped into seven AGs as AG1, AG2-2, AG2-2LP, AG2-3, AG3, AG4 and AG5. Genetic diversity of the pathogen was determined by using molecular markers namely, simple sequence repeats (SSR) and rDNA internal transcribed spacer (ITS). A neighbor-joining tree constructed based on the profiles generated by SSR markers grouped the isolates into eight categories. This revealed 90% of genetic similarity among the isolates and partial correlation with reference to their geographical origin and AGs. The isolates were amplified with a set of primers ITS 1 and ITS 4 and they produced a specific band ≈650 bp. Low level of (7%) variability was observed in the nucleotide sequences of the ITS regions of these isolates. The phylogenic tree generated from bootstrap neighboring joint analysis grouped the Indian populations of R. solani into two categories.Keywords: Chickpea, wet root rot, genetic diversity, simple sequence repeats (SSR), internal transcribed spacer (ITS)

    Sustainable food production for a rapidly growing global population is a major challenge of this century. In order to meet the demand for food production, an additional land area of 2.7 to 4.9 Mha year -1 will be required for agriculture. However, one third of arable lands are already contaminated, therefore the use of polluted lands will have to feature highly in modern agriculture. The use of such lands comes however with additional challenges and suitable agrotechnological interventions are essential for ensuring the safety and sustainability of relevant production system. There are also other issues to consider such as, cost benefit analysis, the possible entry of pollutants into to the phytoproducts, certification and marketing of such products, in order to achieve a the large scale exploitation of polluted land

    Polyethylene Maleate Copolyesters as Coating Materials for Piezoelectric Quartz Crystal-based Chemical Sensors

    Polyethylene maleate (PEM) was synthesised by direct polycondensation of maleic anhydride and ethylene glycol in toluene under reflux usingp-toluene sulphonic acid as a catalyst. Structure of PEM was further modified by varying nature of diols and acid components, chain length of glycols, incorporation of aromatic and fluorine groups in the chain. Formation of PEM was monitored by gas chromatography. The parameters like degree of polymerisation, number average molecular weight (Mn), and weight average molecular weight (Mw) were calculated from the extent of reaction and stoichiometric ratio of the reactants. The number average molecular weight was also determined using Knauer vapour pressure osmometer. Cohesive energy, volume, and solubility parameters of PEM-based copolyesters were calculated by group contribution method. These PEM-based copolyesters’ have been evaluated as sorbent-coating materials for the detection of organ0 phosphorus(OP)compo using dimethylmethylphosphonate (DMMP) as model compound and piezoelectric crystal detector. PEMbisphenol A is found to be the most sensitive and potential coating material for the detection of OP compounds using piezoelectric crystal detector.     Characterization of Toxoplasma gondii isolates in free-range chickens from Chile, South America

    The prevalence of Toxoplasma gondii in free-ranging chickens is a good indicator of the prevalence of T. gondii oocysts in the soil because chickens feed from the ground. The prevalence of T. gondii in 85 free-range chickens (Gallus domesticus) from Chile was determined. Antibodies to T. gondii were assayed by the modified agglutination test (MAT), and found in 47 of 85 (55.3.9%) chickens with titers of 1:5 in six, 1:10 in four, 1:20 in four 1: 40 in three, 1: 80 in nine, 1: 160 in four 1:320 in nine, and 1: 640 or higher in eight. Hearts and brains of 47 chickens with titers of 1:5 or higher were pooled for each chicken and bioassayed in mice. Tissues from 16 seronegative (MAT < 1:5) chickens were pooled and fed to one T. gondii-free cat. Feces of the cat were examined for oocysts but none was found based on bioassay of fecal floats in mice. Hearts and brains from seven seronegative (<1:5) were pooled and bioassayed in mice; T. gondii was not isolated. T. gondii was isolated by bioassay in mice from 22 chickens with MAT titers of 1:20 or higher. Genotyping of these 22 isolates using polymorphisms at the loci SAG1, SAG2, SAG3, BTUB and GRA6 revealed three genotypes. Seventeen isolates had type II alleles and four isolates had type III alleles at all loci. One isolate contained the combination of type I and III alleles. This is the first report of genetic characterization of T. gondii isolates from Chile, South America

    Imposing a Lagrangian Particle Framework on an Eulerian Hydrodynamics Infrastructure in Flash

    In many astrophysical simulations, both Eulerian and Lagrangian quantities are of interest. For example, in a galaxy cluster merger simulation, the intracluster gas can have Eulerian discretization, while dark matter can be modeled using particles. FLASH, a component-based scientific simulation code, superimposes a Lagrangian framework atop an adaptive mesh refinement Eulerian framework to enable such simulations. The discretization of the field variables is Eulerian, while the Lagrangian entities occur in many different forms including tracer particles, massive particles, charged particles in particle-in-cell mode, and Lagrangian markers to model fluid structure interactions. These widely varying roles for Lagrangian entities are possible because of the highly modular, flexible, and extensible architecture of the Lagrangian framework. In this paper, we describe the Lagrangian framework in FLASH in the context of two very different applications, Type Ia supernovae and galaxy cluster mergers, which use the Lagrangian entities in fundamentally different ways

    Reproductive endocrine effects of intranasal administration of norethisterone to adult female rhesus monkeys (Macaca mulatta)

    Intranasal administration of norethisterone at a daily dose of 9 μg between Days 5 and 14 of the menstrual cycles blocked ovulation in 10 out of 17 adult female monkeys. Serum concentrations of hormones indicated that ovulation was blocked due to a suppression of the mid-cycle, oestradiol-induced LH surge. Ovarian follicular activity in the treated menstrual cycles was not affected by norethisterone but there was a marked delay in the onset of the mid-cycle oestradiol surge in most of the treated animals. The duration of the menstrual cycle length after the oestradiol peak was significantly reduced in all the treated monkeys, indicative of a shortened luteal phase

    Hepatitis Associated with a Sarcocystis canis–like Protozoan in a Hawaiian Monk Seal (Monachus schauinslandi)

    A Hawaiian monk seal (Monachus schauinslandi) died in captivity at the National Marine Fisheries Service, Kewalo Basin Facility in Honolulu, Hawaii. The animal was icteric, and the liver was friable. Microscopic lesions were detected in the colon and liver. Colonic lesions included multifocal, necrohemorrhagic colitis associated with gram-negative bacilli. The liver lesions included random hepatic necrosis and cholestasis. Asexual stages of a Sarcocystis canis–like apicomplexan were detected in hepatocytes. The parasite divided by endopolygeny. Merozoites occasionally formed rosettes around a central residual body. Ultrastructurally, merozoites lacked rhoptries. This is the first report of S. canis infection in M. schauinslandi, which is an endangered pinniped in U.S. waters
