    Analysis of β-globin chromatin micro-environment using a novel 3C variant, 4Cv

    Copyright: © 2010 Pink et al.Higher order chromatin folding is critical to a number of developmental processes, including the regulation of gene expression. Recently developed biochemical techniques such as RNA TRAP and chromosome conformation capture (3C) have provided us with the tools to probe chromosomal structures. These techniques have been applied to the β-globin locus, revealing a complex pattern of interactions with regions along the chromosome that the gene resides on. However, biochemical and microscopy data on the nature of β-globin interactions with other chromosomes is contradictory. Therefore we developed a novel 4C variant, Complete-genome 3C by vectorette amplification (4Cv), which allows an unbiased and quantitative method to examine chromosomal structure. We have used 4Cv to study the microenvironment of the β-globin locus in mice and show that a significant proportion of the interactions of β-globin are inter-chromosomal. Furthermore, our data show that in the liver, where the gene is active, β-globin is more likely to interact with other chromosomes, compared to the brain where the gene is silent and is more likely to interact with other regions along the same chromosome.     Pengaruh Jumlah Tawas terhadap Hasil Pewarnaan Dylon pada Bulu Entok sebagai Aksesoris Headpiece

      Tawas adalah garam rangkap aluminium sulfat, yang dipakai untuk menjernihkan air atau campuran bahan celup. Tawas memiliki ciri kristal putih gelap, tembus cahaya, bersifat menguatkan warna. Zat tersebut digunakan sebagai mordan dalam penggunaan zat warna sintetis dylon. Tujuan dari penelitian ini adalah untuk mengetahui hasil jadi pewarnaan dylon dan adanya pengaruh jumlah tawas terhadap hasil pewarnaan pada bulu entok sebagai aksesoris headpiece. Jenis penelitian ini adalah eksperimen. Variabel bebas pada penelitian ini adalah jumlah tawas seberat 5 gram/liter, 10 gram/liter dan 15 gram/liter. Variabel terikat pada penelitian ini adalah hasil pewarnaan. Teknik pengumpulan data menggunakan observasi, yang diolah dengan metode analisis statistik anava tunggal dengan bantuan SPSS 21 dengan taraf signifikan α ≤ 0,05. Hasil analisis data menyatakan bahwa hasil pewarnaan dylon pada aspek kerataan warna  dan ketajaman warna dengan jumlah tawas 5 gram/liter dalam kategori tidak baik, jumlah tawas 10 gram/liter dalam kategori baik dan jumlah tawas 15 gram/liter dalam kategori sangat baik. Ada pengaruh jumlah tawas 5 gram/liter, 10 gram/liter, 15 gram/liter terhadap hasil pewarnaan dylon ditinjau dari aspek kerataan warna dengan signifikan α= 0,00 dan aspek ketajaman warna dengan signifikan α= 0,00. Berdasarkan dari hasil analisis data dan pembahasan dapat disimpulkan bahwa hasil jadi pewarnaan dylon pada bulu entok sebagai aksesoris headpiece dengan jumlah tawas 15 gram/liter sangat baik dan terdapat pengaruh jumlah tawas terhadap hasil pewarnaan dylon pada bulu entok sebagai aksesoris headpiece ditinjau dari aspek kerataan warna dan ketajaman warna.   Kata Kunci : jumlah tawas, pewarnaan, dylon, bulu entok, aksesoris headpiece. &nbsp

    The human myometrium differentially expresses mTOR signalling components before and during pregnancy: Evidence for regulation by progesterone

    Emerging studies implicate the signalling of the mammalian target of rapamycin (mTOR) in a number of reproductive functions. To this date, there are no data regarding the expression of mTOR signalling components in the human myometrium during pregnancy. We hypothesized that mTOR-related genes might be differentially expressed in term or preterm labour as well as in labour or non-labour myometria during pregnancy. Using quantitative RT-PCR we demonstrate for first time that there is a significant downregulation of mTOR, DEPTOR, and Raptor in preterm labouring myometria when compared to non-pregnant tissues taken from the same area (lower segment). We used an immortalized myometrial cell line (ULTR) as an in vitro model to dissect further mTOR signalling. In ULTR cells DEPTOR and Rictor had a cytoplasmic distribution, whereas mTOR and Raptor were detected in the cytoplasm and the nucleus, indicative of mTORC1 shuttling. Treatment with inflammatory cytokines caused only minor changes in gene expression of these components, whereas progesterone caused significant down-regulation. We performed a non-biased gene expression analysis of ULTR cells using Nimblegen human gene expression microarray (n = 3), and selected genes were validated by quantitative RT-PCR in progesterone treated myometrial cells. Progesterone significantly down-regulated key components of the mTOR pathway. We conclude that the human myometrium differentially expresses mTOR signalling components and they can be regulated by progesterone.     Analysis of Brassica oleracea early stage abiotic stress responses reveals tolerance in multiple crop types and for multiple sources of stress

    BACKGROUND Brassica oleracea includes a number of important crop types such as cabbage, cauliflower, broccoli and kale. Current climate conditions and weather patterns are causing significant losses in these crops, meaning that new cultivars with improved tolerance of one or more abiotic stress types must be sought. In this study, genetically fixed B. oleracea lines belonging to a Diversity Fixed Foundation Set (DFFS) were assayed for their response to seedling stage-imposed drought, flood, salinity, heat and cold stress. RESULTS Significant (P ≤ 0.05) variation in stress tolerance response was found for each stress, for each of four measured variables (relative fresh weight, relative dry weight, relative leaf number and relative plant height). Lines tolerant to multiple stresses were found to belong to several different crop types. There was no overall correlation between the responses to the different stresses. CONCLUSION Abiotic stress tolerance was identified in multiple B. oleracea crop types, with some lines exhibiting resistance to multiple stresses. For each stress, no one crop type appeared significantly more or less tolerant than others. The results are promising for the development of more environmentally robust lines of different B. oleracea crops by identifying tolerant material and highlighting the relationship between responses to different stresses. © 2017 Society of Chemical Industr

    Overcoming a Communication Barrier on the Way Towards a Global Sensor Network

    In a global sensor network different sensor platforms will be deployed. A grave obstacle on the way of building sensor networks out of different sensor nodes are incompatible implementations of network protocol stacks used with different sensor node platforms. We describe our efforts to overcome this obstacle in a heterogeneous sensor network consisting out of MICAz Motes and Sun SPOTs, both using an IEEE 802.15.4 radio chip. We explain the major differences in the respective network stacks and our approach to bridge them. A network stack that bridges the gap between different platforms allows for more flexible and robust networks

    ‘It stays with you’: multiple evocative representations of dance and future possibilities for studies in sport and physical cultures

    This article considers the integration of arts-based representations via poetic narratives together with artistic representation on dancing embodiment so as to continue an engagement with debates regarding multiple forms/representations. Like poetry, visual images are unique and can evoke particular kinds of emotional and visceral responses, meaning that alternative representational forms can resonate in different and powerful ways. In the article, we draw on grandparent-grandchild interactions, narrative poetry, and artistic representations of dance in order to illustrate how arts-based methods might synergise to offer new ways of ‘knowing’ and ‘seeing’. The expansion of the visual arts into interdisciplinary methodological innovations is a relatively new, and sometimes contentious approach, in studies of sport and exercise. We raise concerns regarding the future for more arts-based research in the light of an ever-changing landscape of a neoliberal university culture that demands high productivity in reductionist terms of what counts as ‘output’, often within very restricted time-frames. Heeding feminist calls for ‘slow academies’ that attempt to ‘change’ time collectively, and challenge the demands of a fast-paced audit culture, we consider why it is worth enabling creative and arts-based methods to continue to develop and flourish in studies of sport, exercise and health, despite the mounting pressures to ‘perform’

    Correspondence in the December issue by Jonathan Gressel not only states that gender issues in rural settings have not been adequately addressed with respect to weed control biotech but also asserts that such technology can increase the quality of life of rural women in developing countries. Improved weed control is a labor-saving technology that can result in less employment in a labor surplus rural economy. Often in rural areas, wage income is the main source of income and an important determinant of the quality of life, particularly where employment opportunities are generally limited. Apart from soil preparation, planting and weeding, harvesting is also 'femanual' work that can generate more employment if yields are higher. Biotech can enhance the quality of life of women but only if the technology is associated with overall generation of rural employment

    Comparing interferon-gamma release assays with tuberculin skin test for identifying latent tuberculosis infection that progresses to active tuberculosis : systematic review and meta-analysis

    Background Timely and accurate identification of people with latent tuberculosis infection (LTBI) is important for controlling Mycobacterium tuberculosis (TB). There is no gold standard for diagnosis of LTBI. Screening tests such as interferon gamma release assays (IGRAs) and tuberculin skin test (TST) provide indirect and imperfect information. This systematic review compared two types of IGRAs QuantiFERON®-TB Gold In-Tube test (QFT-GIT) and T-SPOT.TB with TST for identification of LTBI by predicting progression to a diagnosis of active TB in three subgroups: children, immunocompromised people, and those recently arrived from countries with high TB burden. Methods Cohort studies were eligible for inclusion. We searched MEDLINE, EMBASE, the Cochrane Library and other databases from December 2009 to June 2015. One reviewer screened studies, extracted data, and assessed risk of bias with cross checking by a second reviewer. Strength of association between test results and incidence of TB was summarised using cumulative incidence ratios (CIRs with 95% CIs). Summary effect measures: the ratio of CIRs (R-CIR) with 95% CIs. R-CIRs, were pooled using a random-effects model. Heterogeneity was assessed using Chi-squared and I2 statistics. Results Seventeen studies, mostly of moderate or high risk of bias (five in children, 10 in immunocompromised people, and two in those recently arrived) were included. In children, while in two studies, there was no significant difference between QFT-GIT and TST (≥5 mm) (pooled R-CIR = 1.11, 95% CI: 0.71, 1.74), two other studies showed QFT-GIT to outperform TST (≥10 mm) in identifying LTBI. In immunocompromised people, IGRA (T-SPOT.TB) was not significant different from TST (≥10 mm) for identifying LTBI, (pooled R-CIR = 1.01, 95% CI: 0.65, 1.58). The forest plot of two studies in recently arrived people from countries with high TB burden demonstrated inconsistent findings (high heterogeneity; I2 = 92%). Conclusions Prospective studies comparing IGRA testing against TST on the progression from LTBI to TB were sparse, and these results should be interpreted with caution due to uncertainty, risk of bias, and unexplained heterogeneity. Population-based studies with adequate sample size and follow-up are required to adequately compare the performance of IGRA with TST in people at high risk of TB

    Scaling Behaviour and Complexity of the Portevin-Le Chatelier Effect

    The plastic deformation of dilute alloys is often accompanied by plastic instabilities due to dynamic strain aging and dislocation interaction. The repeated breakaway of dislocations from and their recapture by solute atoms leads to stress serrations and localized strain in the strain controlled tensile tests, known as the Portevin-Le Chatelier (PLC) effect. In this present work, we analyse the stress time series data of the observed PLC effect in the constant strain rate tensile tests on Al-2.5%Mg alloy for a wide range of strain rates at room temperature. The scaling behaviour of the PLC effect was studied using two complementary scaling analysis methods: the finite variance scaling method and the diffusion entropy analysis. From these analyses we could establish that in the entire span of strain rates, PLC effect showed Levy walk property. Moreover, the multiscale entropy analysis is carried out on the stress time series data observed during the PLC effect to quantify the complexity of the distinct spatiotemporal dynamical regimes. It is shown that for the static type C band, the entropy is very low for all the scales compared to the hopping type B and the propagating type A bands. The results are interpreted considering the time and length scales relevant to the effect.Comment: 35 pages, 6 figure
