Hal ini dapat diwujudkan melalui kerjasama internal dengan membuka kelas kerjasama atau kelas industri pada SMK. SMKN 10 Malang adalah salah satu sekolah yang menerapkan kelas kerjasama dengan pihak Honda di jurusan teknik kendaraan ringan. Kelas tersebut bernama kelas HTEC (Honda Technician Education Class) yang dirilis sejak tanggal 10 Desember 2014. Tujuan dengan adanya kelas HTEC ini mampu meningkatkan kompetensi produktif siswa pada kelas XII agar  mempunyai kompetensi yang baik. Penelitian ini bertujuan untuk mengetahui perbedaan kompetensi produktif antara siswa kelas industri dengan kelas reguler. Desain penelitian ini termasuk dalam desain eksperimen dengan post-test only control design. Variabel dalam penelitian ini terdiri dari variabel bebas dan variabel terikat. Variabel bebas dalam penelitian ini adalah pengelompokkan kelas yang terdiri dari kelas industri dan kelas reguler sedangkan kompetensi produktif merupakan variabel terikat. Subyek penelitian ini adalah siswa kelas XII Teknik Kendaraan Ringan pada kelas industri dan kelas reguler menggunakancluster sampling dan proportional sample sebagai teknik pengambilan sampel penelitian. Teknik analisis data yang digunakan adalah teknik uji t (Independent Sample t-Test). Rata-rata nilai post-test kompetensi produktif siswa kelas industri adalah 83.8 dan rata-rata nilai post-test kompetensi produktif siswa kelas reguler adalah 76.94. Hasil uji t diperoleh nilai sig.     Science Hybrid Orbiter and Lunar Relay (SCHOLR) Architecture and Design

    Considered both a stepping-stone to deep space and a key to unlocking the mysteries of planetary formation, the Moon offers a unique opportunity for scientific study. Robotic precursor missions are being developed to improve technology and enable new approaches to exploration. Robots, lunar landers, and satellites play significant roles in advancing science and technologies, offering close range and in-situ observations. Science and exploration data gathered from these nodes and a lunar science satellite is intended to support future human expeditions and facilitate future utilization of lunar resources. To attain a global view of lunar science, the nodes will be distributed over the lunar surface, including locations on the far side of the Moon. Given that nodes on the lunar far side do not have direct line-of-sight for Earth communications, the planned presence of such nodes creates the need for a lunar communications relay satellite. Since the communications relay capability would only be required for a small portion of the satellite s orbit, it may be possible to include communication relay components on a science spacecraft. Furthermore, an integrated satellite has the potential to reduce lunar surface mission costs. A SCience Hybrid Orbiter and Lunar Relay (SCHOLR) is proposed to accomplish scientific goals while also supporting the communications needs of landers on the far side of the Moon. User needs and design drivers for the system were derived from the anticipated needs of future robotic and lander missions. Based on these drivers and user requirements, accommodations for communications payload aboard a science spacecraft were developed. A team of interns identified and compared possible SCHOLR architectures. The final SCHOLR architecture was analyzed in terms of orbiter lifetime, lunar surface coverage, size, mass, power, and communications data rates. This paper presents the driving requirements, operational concept, and architecture views for SCHOLR within a lunar surface nodal network.     Oxaliplatin induces drug resistance more rapidly than cisplatin in H69 small cell lung cancer cells

    Cisplatin produces good responses in solid tumours including small cell lung cancer (SCLC) but this is limited by the development of resistance. Oxaliplatin is reported to show activity against some cisplatin-resistant cancers but there is little known about oxaliplatin in SCLC and there are no reports of oxaliplatin resistant SCLC cell lines. Studies of drug resistance mainly focus on the cellular resistance mechanisms rather than how the cells develop resistance. This study examines the development of cisplatin and oxaliplatin resistance in H69 human SCLC cells in response to repeated treatment with clinically relevant doses of cisplatin or oxaliplatin for either 4 days or 2h. Treatments with 200ng/ml cisplatin or 400ng/ml oxaliplatin for 4 days produced sublines (H69CIS200 and H69OX400 respectively) that showed low level (approximately 2-fold) resistance after 8 treatments. Treatments with 1000ng/ml cisplatin or 2000ng/ml oxaliplatin for 2h also produced sublines, however these were not stably resistant suggesting shorter treatment pulses of drug may be more effective. Cells survived the first five treatments without any increase in resistance, by arresting their growth for a period and then regrowing. The period of growth arrest was reduced after the sixth treatment and the H69CIS200 and H69OX400 sublines showed a reduced growth arrest in response to cisplatin and oxaliplatin treatment suggesting that "regrowth resistance" initially protected against drug treatment and this was further upregulated and became part of the resistance phenotype of these sublines. Oxaliplatin dose escalation produced more surviving sublines than cisplatin dose escalation but neither set of sublines were associated with increased resistance as determined by 5-day cytotoxicity assays, also suggesting the involvement of regrowth resistance. The resistant sublines showed no change in platinum accumulation or glutathione levels even though the H69OX400 subline was more sensitive to buthionine sulfoximine treatment. The H69CIS200 cells were cross-resistant to oxaliplatin demonstrating that oxaliplatin does not have activity against low level cisplatin resistance. Relative to the H69 cells, the H69CIS200 and H69OX400 sublines were more sensitive to paclitaxel and taxotere suggests the taxanes may be useful in the treatment of platinum resistant SCLC. These novel cellular models of cisplatin and oxaliplatin resistant SCLC will be useful in developing strategies to treat platinum-resistant SCLC

    Development of core entrustable professional activities linked to a competency-based veterinary education framework

    Purpose: Despite the adoption of competency-based education in some veterinary schools over the past 15 years, only recently has a concerted effort been directed toward this in veterinary education internationally. Methods: In 2015, educational leaders from the Association of American Veterinary Medical Colleges (AAVMC) member schools came together with a strong call to action to create shared tools for clinical competency assessment. Results: This resulted in the formation of the AAVMC Competency-Based Veterinary Education (CBVE) Working Group, which then embarked on the creation of a shared competency framework and the development of eight core entrustable professional activities (EPAs) linked to this framework. Conclusions: This paper will report on the development of these EPAs and their integration with the concurrently-developed CBVE Framework

    Accurate and exact CNV identification from targeted high-throughput sequence data

    <p>Abstract</p> <p>Background</p> <p>Massively parallel sequencing of barcoded DNA samples significantly increases screening efficiency for clinically important genes. Short read aligners are well suited to single nucleotide and indel detection. However, methods for CNV detection from targeted enrichment are lacking. We present a method combining coverage with map information for the identification of deletions and duplications in targeted sequence data.</p> <p>Results</p> <p>Sequencing data is first scanned for gains and losses using a comparison of normalized coverage data between samples. CNV calls are confirmed by testing for a signature of sequences that span the CNV breakpoint. With our method, CNVs can be identified regardless of whether breakpoints are within regions targeted for sequencing. For CNVs where at least one breakpoint is within targeted sequence, exact CNV breakpoints can be identified. In a test data set of 96 subjects sequenced across ~1 Mb genomic sequence using multiplexing technology, our method detected mutations as small as 31 bp, predicted quantitative copy count, and had a low false-positive rate.</p> <p>Conclusions</p> <p>Application of this method allows for identification of gains and losses in targeted sequence data, providing comprehensive mutation screening when combined with a short read aligner.</p

    Structure and mechanism of human DNA polymerase Ρ

    The variant form of the human syndrome xeroderma pigmentosum (XPV) is caused by a deficiency in DNA polymerase eta (Pol eta), a DNA polymerase that enables replication through ultraviolet-induced pyrimidine dimers. Here we report high-resolution crystal structures of human Pol eta at four consecutive steps during DNA synthesis through cis-syn cyclobutane thymine dimers. Pol eta acts like a 'molecular splint' to stabilize damaged DNA in a normal B-form conformation. An enlarged active site accommodates the thymine dimer with excellent stereochemistry for two-metal ion catalysis. Two residues conserved among Pol eta orthologues form specific hydrogen bonds with the lesion and the incoming nucleotide to assist translesion synthesis. On the basis of the structures, eight Pol eta missense mutations causing XPV can be rationalized as undermining the molecular splint or perturbing the active-site alignment. The structures also provide an insight into the role of Pol eta in replicating through D loop and DNA fragile sites

    How User‐centric Innovation is Affecting Stakeholder Marketing Strategies: Exploratory Findings from the Music Industry

    This paper empirically explores how user‐centric innovation (UCI) in the music industry is affecting how key stakeholder groups are approaching and developing their marketing (and associated management) strategies. The three‐stage interview‐based research methodology consisted of 52 semi‐structured in‐depth interviews with UCI experts and artist managers, as well as representatives from major record labels. The findings make four substantial contributions to theory and practice in the interrelated fields of UCI, marketing and the music industry. First, they provide practical and pragmatic insights for industry practitioners on how different UCI marketing approaches are affecting their management strategies. Second, they take steps towards answering many of the identified gaps in research and knowledge relating to the concept of UCI. Third, they present theoretical models as a foundation for which new UCI marketing theory can be built upon. Last, they offer directions for future research to advance our empirical findings.</jats:p

    Using screen video capture software to aide and inform cognitive interviewing

    Web-based surveys are a salient tool in the repertoire of social and behavioral scientists. The increase in web-based surveys is understandable considering the distinct advantages offered, including: (a) lower costs and reduced labor time, (b) ability to directly transfer data into statistical packages (reducing coding errors), (c) customization options enabling more attractive presentation, (d) ability to reduce respondent burden by embedding skip patterns, and (e) access to larger sample sizes in different geographic regions. It is important to note, however, that administering web-based surveys also introduces distinct sources of error (e.g., coverage, sampling and non-response). Regardless of format (e.g., paper-and-pencil or web-based), specific, prescribed steps must be followed when constructing an instrument in order to reduce survey error and lend credence to the data collected before subsequent analysis is performed. One of those crucial stages integral to the pretesting process is cognitive interviewing. Cognitive interviewing is a qualitative process, encompassing two main techniques: think aloud interviewing and verbal probing. Collectively, these two methods seek to (a) produce information on what the respondent is thinking while answering the questions, (b) the cognitive processes used to answer the questions, and (c) how the respondent answers the questions.
