    A monoclonal antibody against Echinococcus multilocularis Em2 antigen

    A monoclonal antibody (MAb G11) species-specific to the Em2 antigen of Echinococcus multilocularis was generated for (i) further biological characterization of the Em2 antigen, (ii) easy affinity-purification of Em2 antigen for immunodiagnostic and immunological investigations and (iii) development of a sandwich-ELISA for the detection of Em2 antigen in diagnostic samples and thus species-specific identification of E. multilocularis metacestode material. The MAb G11 was used in an antibody sandwich-ELISA to detect soluble Em2 antigen with a methodical sensitivity of 80 ng E. multilocularis antigen/ml of solution. MAb G11 specifically detected Em2 antigen in all of 15 E. multilocularis-isolates originating from various geographical areas and in none of other helminth isolates (e.g. Echinococcus granulosus, E. vogeli, and others). Further biological analysis by FITC-labelled MAb G11 demonstrated unique binding activity to the laminated layer of the metacestode. Also, oncospheres were binding FITC-labelled MAb G11 on an outer layer synthesized during cultivation in vitro for 13 days after hatching. Application of the MAb G11 antibody sandwich-ELISA for investigation of solubilized oncospheres confirmed the in vitro synthesis of Em2 antigen by oncospheres on day 13 p.i. Adult stages (somatic antigens) and freshly hatched oncospheres were always MAb G11 negative. Solid-phase MAb G11 was used for purification of the corresponding Em2 antigen by affinity chromatography. A preliminary serological evaluation of the Em2(G11) antigen by ELISA revealed identical immunodiagnostic characteristics, compared to Em2 obtained by classical means, thus suggesting the presented method for future isolation of large-scale Em2 antige

    Mapping the increasing risk of human alveolar echinococcosis in Limburg, The Netherlands

    The parasite Echinococcus multilocularis was first detected in The Netherlands in 1996 and repeated studies have shown that the parasite subsequently spread in the local population of foxes in the province of Limburg. It was not possible to quantify the human risk of alveolar echinococcosis because no relationship between the amount of parasite eggs in the environment and the probability of infection in humans was known. Here, we used the spread of the parasite in The Netherlands as a predictor, together with recently published historical records of the epidemiology of alveolar echinococcosis in Switzerland, to achieve a relative quantification of the risk. Based on these analyses, the human risk in Limburg was simulated and up to three human cases are predicted by 2018. We conclude that the epidemiology of alveolar echinococcosis in The Netherlands might have changed from a period of negligible risk in the past to a period of increasing risk in the forthcoming year

    An enzyme-linked immunosorbent assay for diagnostic detection of Taenia saginata copro-antigens in humans

    An immunodiagnostic sandwich enzyme-linked immunosorbent assay (ELISA) was developed for the detection of soluble Taenia saginata antigens in stool samples (copro-antigens) of infected humans, using affinity-purified polyclonal antibodies obtained from rabbits hyperimmunized with excretory/secretory antigens derived from T. saginata maintained in vitro. Investigation of operating characteristics showed very low cross-reactivity with crude antigens from helminths other than Taenia, including Dipylidium caninum and Diphyllobothrium latum. The specificity of the assay was 95% when testing stool samples from 100 persons who were either infected with Ascaris lumbricoides, Trichuris trichiura, hookworms, Enterobius vermicularis or Hymenolepis nana, or who had no intestinal helminthosis detected. Analysis of diagnostic sensitivity demonstrated that in 85% of 34 samples from 23 untreated persons with intestinal T. saginata infection (selected by previous proglottid and/or egg detection) copro-antigens were detected by the T. saginata ELISA. In the same samples, Taenia eggs were detected in 62%. Only 41% of the samples reacted positively in a heterologous T. hydatigena ELISA. Post-treatment control revealed a high concentration of T. saginata copro-antigens for 1-4 d after administration of niclosamide or praziquantel, and negative values 9-17 d after treatment.     Coprological study on intestinal helminths in Swiss dogs: temporal aspects of anthelminthic treatment

    Coproscopic examination of 505 dogs originating from the western or central part of Switzerland revealed the presence (prevalence data) of the following helminthes: Toxocara canis (7.1%), hookworms (6.9%), Trichuris vulpis (5.5%), Toxascaris leonina (1.3%), Taeniidae (1.3%), Capillaria spp. (0.8%), and Diphyllobothrium latum (0.4%). Potential risk factors for infection were identified by a questionnaire: dogs from rural areas significantly more often had hookworms and taeniid eggs in their feces when compared to urban family dogs. Access to small rodents, offal, and carrion was identified as risk factor for hookworm and Taeniidae, while feeding of fresh and uncooked meat did not result in higher prevalences for these helminths. A group of 111 dogs was treated every 3months with a combined medication of pyrantel embonate, praziquantel, and febantel, and fecal samples were collected for coproscopy in monthly intervals. Despite treatment, the yearly incidence of T. canis was 32%, while hookworms, T. vulpis, Capillaria spp., and Taeniidae reached incidences ranging from 11 to 22%. Fifty-seven percent of the 111 dogs had helminth eggs in their feces at least once during the 1-year study period.     Implementing Quantum Gates using the Ferromagnetic Spin-J XXZ Chain with Kink Boundary Conditions

    We demonstrate an implementation scheme for constructing quantum gates using unitary evolutions of the one-dimensional spin-J ferromagnetic XXZ chain. We present numerical results based on simulations of the chain using the time-dependent DMRG method and techniques from optimal control theory. Using only a few control parameters, we find that it is possible to implement one- and two-qubit gates on a system of spin-3/2 XXZ chains, such as Not, Hadamard, Pi-8, Phase, and C-Not, with fidelity levels exceeding 99%.Comment: Updated Acknowledgement

    Is high prevalence of Echinococcus multilocularis in wild and domestic animals associated with disease incidence in humans?

    We investigated a focus of highly endemic Echinococcus multilocularis infection to assess persistence of high endemicity in rural rodents, explore potential for parasite transmission to domestic carnivores, and assess (serologically) putative exposure versus infection frequency in inhabitants of the region. From spring 1993 to spring 1998, the prevalence of E. multilocularis in rodents was 9% to 39% for Arvicola terrestris and 10% to 21% for Microtus arvalis. From June 1996 to October 1997, 6 (7%) of 86 feral dogs and 1 of 33 cats living close to the region tested positive for intestinal E. multilocularis infection. Testing included egg detection by coproscopy, antigen detection by enzyme-linked immunosorbent assay (ELISA), and specific parasite DNA amplification by polymerase chain reaction. Thus, the presence of infected domestic carnivores can increase E. multilocularis exposure risk in humans. A seroepidemiologic survey of 2,943 blood donors in the area used specific Em2-ELISA. Comparative statistical analyses of seroprevalence and clinical incidence showed an increase in Em2-seroprevalence from 1986 and 1996-97 but no increase in clinical incidence of alveolar hydatid disease
