    Photosynthetic activity is a fundamental process in the physiology of plants, and its regulation plays an important role in determining the effect of abiotic factors. Quinoa is a plant species of agronomic and nutritional interest that has been recognized for its adaptability to extreme environmental conditions, however, climate change may result in unfavorable conditions capable of affecting the natural development of this species, which is of great interest culture and research in South America. To evaluate the response of quinoa to stress, techniques could be used that quantify the loss of light energy through its dissipation in the form of heat. However, the measurement of chlorophyll fluorescence is the most widely used and accessible technique for field research, which allows to recognize the relationships between the plant and agroclimatic factors.     Gastric perforation secondary to gastric balloon

    Introduction: The use of the gastric balloon is an alternative and minimally invasive treatment of morbid obesity, with specific indications. Although the insertion procedure is easy and well tolerated by most patients, complications can occur. A serious and uncommon complication is gastric perforation. Objective:To present a patient with gastric perforation secondary to an intragastric balloon who underwent endoscopic treatment in two stages. Clinical case: A 25-year-old patient with gastric perforation treated with a gastric balloon 6 months before going to the emergency department with severe abdominal pain is presented. Chest X-ray showed little right subdiaphragmatic pneumoperitoneum, so contrasted computed axial tomography of abdomen was performed, which showed signs suggestive of gastric perforation. Based on the tomographic findings, the patient underwent diagnostic laparoscopy. A plastron was observed in the epigastrium with a perforation of 0.5 mm. The purulent peritonitis was drained laparoscopically, with gastrorrhaphy and epiploplasty. An inflated balloon was palpated could not be accessed through the perforation orifice. The patient was then scheduled for an endoscope-guided gastric balloon removal, which was performed without complications. Conclusions: Gastric perforation by intragastric balloon is an infrequent complication, but should be suspected in the event of severe abdominal pain. Contrasted computed tomography should be performed for diagnosis prior to treatment

    Métodos de conservación para actinobacterias con actividad solubilizadora de fósforo

    El objetivo de esta investigación fue evaluar diferentes métodos de conservación para actinobacterias solubilizadoras de fósforo debido a la poca información de métodos específicos reportados para estos microorganismos. Los métodos de conservación se evaluaron a 3 diferentes periodos de tiempo; largo, mediano y corto plazo, usando métodos de congelación y liofilización; arcilla, sílica, arena  y transferencia periódica, respectivamente. Para ello se usaron 15 aislamientos de 3 localidades diferentes (La Vega, Maní y Tota) y un banco de referencia de la Unidad de Investigaciones Agropecuarias de la Pontificia Universidad Javeriana. Se prepararon todos los inóculos en solución salina al 0,85% (p/v) y se ajustaron a una concentración de 108 cel/mL, seguido a ello se inocularon los viales de cada método de conservación con sus respectivos crioprotectantes, glicerol 20% (v/v), 30% (v/v) para congelación y skim milk 18% (p/v) para liofilización. Los métodos a mediano plazo se ejecutaron de igual manera, el inóculo se agregó a 10 perlas de arcilla, 10 g de arena y 5 g de sílica, posteriormente se almacenaron a 4ºC. El método de corto plazo se evaluó en agar avena (15 g/L). La evaluación se realizó mediante recuento directo en cámara de Neubauer por la técnica de azul de tripán, además de la caracterización macroscópica y microscópica de cada aislamiento en transferencia periódica. Se estableció que la actividad solubilizadora de fósforo se mantuvo más estable en los métodos de glicerol 30 % (p/v) y liofilización según el análisis estadístico.  Palabras clave: viabilidad, crioprotectantes, banco, liofilización, crioconservación.The aim of this research was to evaluate different methods of preservation for actinobacteria with phosphate solubilization activity due to there are a few specific methods reported for these organisms. The methods were evaluated at three different time periods; long, medium and short-term and employing methods as cryopreservation and dry-freezing; clay, silica and sand; and periodically plating, respectively. Therefore 15 isolates from 3 different locations (La Vega, Maní and Tota) and a bank reference from Livestock Research Unit of the Pontificia Universidad Javeriana were used. All inocula were prepared in 0.85% saline solution (w/v) which were adjusted to a concentration of 108 cells/mL, followed each vial was inoculated with their respective storage cryoprotectants , glycerol 20% (v/v), 30% (v/v) to freeze and 18% skim milk (w/v) for dry-freezing. Medium-term methods were performed similarly; the inoculum was added to 10 clay beads, 10 g of sand and 5 g of silica and then stored at 4 °C. The short-term method was evaluated in oatmeal agar (15 g/L). The evaluation was performed by direct counting in a Neubauer chamberusing trypan blue staining technique, in addition to macroscopic and microscopic characterization of each isolate in periodic plating. It was established that the phosphorus solubilizing activity was more stable in glycerol 30% (w/v) and lyophilization for statistical analysis.Keywords: Viability; cryoprotectants; bank; dry-freezing; cryopreservation

    Histologic, metabolomic, and transcriptomic differences in fir trees from a peri-urban forest under chronic ozone exposure

    Abstract Urbanization modifies ecosystem conditions and evolutionary processes. This includes air pollution, mostly as tropospheric ozone (O3), which contributes to the decline of urban and peri-urban forests. A notable case are fir (Abies religiosa) forests in the peripheral mountains southwest of Mexico City, which have been severely affected by O3 pollution since the 1970s. Interestingly, some young individuals exhibiting minimal O3?related damage have been observed within a zone of significant O3 exposure. Using this setting as a natural experiment, we compared asymptomatic and symptomatic individuals of similar age (≤15?years old; n?=?10) using histologic, metabolomic, and transcriptomic approaches. Plants were sampled during days of high (170?ppb) and moderate (87?ppb) O3 concentration. Given that there have been reforestation efforts in the region, with plants from different source populations, we first confirmed that all analyzed individuals clustered within the local genetic group when compared to a species-wide panel (Admixture analysis with ~1.5K SNPs). We observed thicker epidermis and more collapsed cells in the palisade parenchyma of needles from symptomatic individuals than from their asymptomatic counterparts, with differences increasing with needle age. Furthermore, symptomatic individuals exhibited lower concentrations of various terpenes (ß-pinene, ß-caryophylene oxide, α-caryophylene, and ß-α-cubebene) than asymptomatic trees, as evidenced through GC?MS. Finally, transcriptomic analyses revealed differential expression for 13 genes related to carbohydrate metabolism, plant defense, and gene regulation. Our results indicate a rapid and contrasting phenotypic response among trees, likely influenced by standing genetic variation and/or plastic mechanisms. They open the door to future evolutionary studies for understanding how O3 tolerance develops in urban environments, and how this knowledge could contribute to forest restoration

    Differential effects of the second SARS-CoV-2 mRNA vaccine dose on T cell immunity in naive and COVID-19 recovered individuals

    The rapid development of mRNA-based vaccines against the severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) led to the design of accelerated vaccination schedules that have been extremely effective in naive individuals. While a two-dose immunization regimen with the BNT162b2 vaccine has been demonstrated to provide a 95% efficacy in naive individuals, the effects of the second vaccine dose in individuals who have previously recovered from natural SARS-CoV-2 infection has not been investigated in detail. In this study, we characterize SARS-CoV-2 spike-specific humoral and cellular immunity in naive and previously infected individuals during and after two doses of BNT162b2 vaccination. Our results demonstrate that, while the second dose increases both the humoral and cellular immunity in naive individuals, COVID-19 recovered individuals reach their peak of immunity after the first dose.
