    Dextran Sulphate Crowding and Sodium Deoxycholate Lysis of Primary Breast Fibroblast Cells Achieve Extracellular Matrix Deposition and Decellularization for Breast Cancer Stem Cell Culture

    Background: The microenvironment including stromal cells and extracellular matrix (ECM) is now consideredan active contributor to tumor progression. Certain studies have developed ECM which supports a suitable cellulargrowth in vitro. The ECM is a complex supramolecular assembly of a variety of glycoproteins and proteoglycans.Extracellular matrix provides tissue integrity, acts as a native scaffold for cell attachment and interaction and alsoserves as reservoir for growth factors. The aim of this experiment w asto achieve the deposition and decellularizationof ECM.Methods: Cells crowder have been developed to increase ECM deposit in the primary breast fibroblast cells layerobtained from isolation of single cell from breast mammoplasty specimen. Five hundred kDa dextran was addedinto DMEM medium containing 0.5% fetal bovine serum (FBS) and 100 μm of L-ascorbic acid 2-phosphate. Afterseven days, cells were lysed by sodium deoxycholate (DOC). Results were observed in a fluorescence microscope.Results: Extracellular matrix deposition and decellularization of primary breast fibroblast cells were detected byusing extracellular matrix protein, fibronectin via rabbit anti human fibronectin and anti-rabbit IgG conjugated withAlexa Fluor 488.Conclusion: Dextran sulphate increased extracellular matrix deposit in primary breast fibroblast cell layer and thetreatment with sodium deoxycolate lysed cells resulted in extracellular matrix decellularization.     Characterization of brick masonry: Study towards retrofitting URM walls with timber-panels

    The overall purpose of this study is to explore the possibility of using timber-panels to retrofit URM walls. However, this paper only present the overall proposed experimental program together with the experimental characterization of mechanical properties of masonry components: units and mortar. The present work developed finite element numerical model that is able to predict the strength of the masonry cube, on the safe side. The numerical model was validated with an experimental test on masonry cube showing 9% difference in the maximum compressive strength of masonry. The modelling technique adopted is the detailed micro modelling where the unit and mortar were represented by their respective mechanical properties using ABAQUS. Because, the numerical results compliment what was observed during the experimental test, then the developed model can be used to predict the general behaviour of masonry wall in the subsequent study.- (undefined

    Psychosocial health and psychological adjustment in adolescents and young adults with congenital melanocytic nevi: Analysis of self-reports

    This study assessed self-reported health-related quality of life and psychological adjustment in 43 adolescents and young adults (ages in years: 14–24, M = 17.6, SD = 2.2) with congenital melanocytic nevi (CMN) and examined associations with sociodemographic variables, characteristics of the CMN, perceived social reactions, and cognitive emotion regulation strategies. Outcome measures included the Pediatric Quality of Life Inventory^{™} 4.0 and the Strengths and Difficulties Questionnaire. Findings suggest impaired psychosocial health and psychological adjustment in youth with CMN compared to community norms. Impairments were associated with higher age of participants, lower socioeconomic status, visibility of the skin lesion, perceived stigmatization, poorer perceived social support, and maladaptive cognitive emotion regulation strategies (self-blame, rumination, and catastrophizing), but not with sex of participants, extent of the skin lesion, and surgical removal of the nevus. Implications for clinical practice and future research are discussed

    Heterogeneity of resting and hyperemic myocardial blood flow in healthy humans

    Objective: Absolute myocardial blood flow (MBF) is not well-defined in large normal populations, and appears to be heterogeneous in both humans and animals. These factors contribute to the difficulties in defining resting MBF to hibernating myocardium. We therefore assessed absolute baseline and hyperemic MBF in a large population of normal humans. Methods: MBF was quantified by positron emission tomography with oxygen-15-labeled water at baseline and during hyperemia induced by either adenosine or dipyridamole in 131 men and 38 women, aged 21-86 (mean 46±12) years. MBF was corrected for workload using the rate-pressure product (RPP). Results: Uncorrected baseline MBF ranged from 0.590 to 2.050 (mean 0.985±0.230) ml/min/g (coefficient of variation=27%), and corrected MBF from 0.736 to 2.428 (mean 1.330±0.316) ml/min/g (coefficient of variation=24%). MBF in the inferior region was significantly (P<0.0001) lower than either the anterior or lateral regions. Baseline MBF in females was significantly (P<0.001) higher than in males. Conclusions: These results confirm the heterogeneity of MBF in normals and highlight the difficulty in establishing the lower limit of normal MB

Para isso, busca-se o desenvolvimento de novas tecnologias, materiais e métodos que ofereçam as melhorias e a qualidade de vida buscada. Frente aos novos parâmetros impostos pelo tempo, o desejo e a necessidade de mudança, trazem consigo o homem que não planeja, não pesquisa e extrapola os seus recursos. A muito tempo, ações que inicialmente deveriam trazer melhorias, vem sabotando a qualidade da vida humana e do planeta. Altos índices de poluição e desmatamento, causados pelo uso de recursos em excesso, geram índices alarmantes. Os objetivos para o desenvolvimento sustentável (ODS) propostos pela ONU (Organização das Nações Unidas em 2016, indicam a necessidade urgente de novos parâmetros para o desenvolvimento humano, correlacionados a preocupação com o meio ambiente e a disponibilidade limitada de matéria prima. Neste âmbito, a indústria da construção civil é quase uma inimiga. Suas atividades demandam a utilização exacerbada de recursos naturais que em contra partida, voltam para natureza em forma de resíduo, depósitos irregulares e altos índices de poluição (ar, água e terra). Porém, ao mesmo tempo que esta classe polui, também oferece meios para incorporação de atitudes favoráveis ao meio ambiente, como por exemplo, a utilização de seus resíduos dentro da própria obra. Atualmente, grande parte dos resíduos sólidos são formados por resíduos de construção e demolição (RCD). Este projeto visa atrair os olhares da sociedade para o potencial da aplicabilidade e reutilização do RCD, através da sua valorização em utilitários produzidos com concreto, onde os agregados são substituídos por diferentes granulometrias do material reciclado.
