Das Ziel dieser Arbeit ist die Ergründung von Leben und Werk einer historischen Person anhand von biografischer Forschung und Deduktion. Diese Person ist Mimi Grossberg. Im Vordergrund dieser Forschungsarbeit stehen im Speziellen ihre Tätigkeiten als Organisatorin sowie Exilforscherin im Exil. Sie war Lyrikerin, Autorin und Herausgeberin mehrerer Anthologien. Ausgangspunkt und wichtigster Bestandteil der Recherche stellt der gesamte Nachlass Grossbergs, welcher sich im Literaturhaus in Wien befindet, dar. Die im Nachlass befindlichen Materialien bzw. Dokumente wurden zum Inhalt der biografischen Forschung. Außerdem stark gewichtet wurde die Operationalisierung der Anforderungsprofile einer Kommunikatorin, um in weiterer Folge deduktive Schlüsse von diesem allgemeinen Profil auf Mimi Großberg ziehen zu können. Im Rahmen der vorliegenden, biografischen Forschung wurden sämtliche Archive und Bibliotheken nach verwertbarem Material über die relevante Person durchsucht. Die gewonnenen Daten, welche vorwiegend aus dem ergiebigen Nachlass Grossbergs stammen, wurden in weiterer Folge geordnet und genutzt, um einerseits die Biografie über Mimi Grossberg zu vervollständigen beziehungsweise anzureichern und andererseits, um sie in das Anforderungsprofil einer Kommunikatorin einordnen zu können. Ihre Persönlichkeit und in erster Linie ihre Tätigkeiten im Exil für die deutschsprachige Literaturszene werden an dieser Stelle zum Gegenstand einer eingehenden Untersuchung. Ihre detaillierten Aufzeichnungen, gesammelten Dokumente und die Korrespondenz mit der Familie und Freunden lieferten wichtige Hinweise. Ihr Schaffen wurde, insbesondere vor dem historisch-sozialen Hintergrund der erzwungenen Emigration betrachtet. Aus dieser Diplomarbeit ergibt sich somit ein stimmungsreiches Bild zur Person Mimi Grossberg und ihrer Tätigkeiten im Exil.The aim of this work is the exploration of the life and work of a historical person using biographical research and the method of deduction. Mimi Grossberg, an Austrian Jewish publicist, is the subject of the research which focuses on her activities as a communicator in exile. The materials and documents, which are part of her legacy that Grossberg donated to the „Literaturhaus Wien“ in Vienna, became the content of the biographical research. Furthermore, the creation of a general profile of a communicator was another important part within this study. Based on this general profile, it was possible to draw a deductive conclusion of Mimi Grossberg´s role as a communicator in exile. For the purpose of the biographical research, it was necessary to search through many different archives and libraries to find applicable information about the relevant person. Still, most of the data originates from Grossberg´s legacy and was used to author her biography as well as to classify her through a general profile of a communicator. Her work in exile for the German-speaking literature scene is a further subject of extensive analyses. Therefore her detailed records, collected documents and her correspondence with her family were important indicators. Finally, this thesis reflects an interesting depiction of Mimi Grossberg´s life, her activities and works in New York City

BART-Seq: cost-effective massively parallelized targeted sequencing for genomics, transcriptomics, and single-cell analysis.

We describe a highly sensitive, quantitative, and inexpensive technique for targeted sequencing of transcript cohorts or genomic regions from thousands of bulk samples or single cells in parallel. Multiplexing is based on a simple method that produces extensive matrices of diverse DNA barcodes attached to invariant primer sets, which are all pre-selected and optimized in silico. By applying the matrices in a novel workflow named Barcode Assembly foR Targeted Sequencing (BART-Seq), we analyze developmental states of thousands of single human pluripotent stem cells, either in different maintenance media or upon Wnt/beta-catenin pathway activation, which identifies the mechanisms of differentiation induction. Moreover, we apply BART-Seq to the genetic screening of breast cancer patients and identify BRCA mutations with very high precision. The processing of thousands of samples and dynamic range measurements that outperform global transcriptomics techniques makes BART-Seq first targeted sequencing technique suitable for numerous research applications
