Accurate prediction of core-level spectra of radicals at density functional theory cost via square gradient minimization and recoupling of mixed configurations
State-specific orbital optimized approaches are more accurate at predicting
core-level spectra than traditional linear-response protocols, but their
utility had been restricted on account of the risk of `variational collapse'
down to the ground state. We employ the recently developed square gradient
minimization (SGM, J. Chem. Theory Comput. 16, 1699-1710, 2020) algorithm to
reliably avoid variational collapse and study the effectiveness of orbital
optimized density functional theory (DFT) at predicting second period element
1s core-level spectra of open-shell systems. Several density functionals
(including SCAN, B3LYP and B97X-D3) are found to predict excitation
energies from the core to singly occupied levels to high accuracy ( eV
RMS error), against available experimental data. Higher excited states are
however more challenging by virtue of being intrinsically multiconfigurational.
We thus present a CI inspired route to self-consistently recouple single
determinant mixed configurations obtained from DFT, in order to obtain
approximate doublet states. This recoupling scheme is used to predict the C
K-edge spectra of the allyl radical, the O K-edge spectra of CO and the N
K-edge of NO to high accuracy relative to experiment, indicating
substantial promise in using this approach for computation of core-level
spectra for doublet species (vs more traditional time dependent DFT, EOM-CCSD
or using unrecoupled mixed configurations). We also present general guidelines
for computing core-excited states from orbital optimized DFT.Comment: Added more dat
The Phoenix Deep Survey: The 1.4 GHz microJansky catalogue
The initial Phoenix Deep Survey (PDS) observations with the Australia
Telescope Compact Array have been supplemented by additional 1.4 GHz
observations over the past few years. Here we present details of the
construction of a new mosaic image covering an area of 4.56 square degrees, an
investigation of the reliability of the source measurements, and the 1.4 GHz
source counts for the compiled radio catalogue. The mosaic achieves a 1-sigma
rms noise of 12 microJy at its most sensitive, and a homogeneous radio-selected
catalogue of over 2000 sources reaching flux densities as faint as 60 microJy
has been compiled. The source parameter measurements are found to be consistent
with the expected uncertainties from the image noise levels and the Gaussian
source fitting procedure. A radio-selected sample avoids the complications of
obscuration associated with optically-selected samples, and by utilising
complementary PDS observations including multicolour optical, near-infrared and
spectroscopic data, this radio catalogue will be used in a detailed
investigation of the evolution in star-formation spanning the redshift range 0
< z < 1. The homogeneity of the catalogue ensures a consistent picture of
galaxy evolution can be developed over the full cosmologically significant
redshift range of interest. The 1.4 GHz mosaic image and the source catalogue
are available on the web at http://www.atnf.csiro.au/~ahopkins/phoenix/ or from
the authors by request.Comment: 16 pages, 11 figures, 4 tables. Accepted for publication by A
Untangling the stranglehold through mathematical modelling of Streptococcus equi subspecies equi transmission
Strangles, a disease caused by infection with Streptococccus equi subspecies equi (S. equi), is endemic worldwide and one of the most frequently diagnosed infectious diseases of horses. Recent work has improved our knowledge of key parameters of transmission dynamics, but important knowledge gaps remain. Our aim was to apply mathematical modelling of S. equi transmission dynamics to prioritise future research areas, and add precision to estimates of transmission parameters thereby improving understanding of S. equi epidemiology and quantifying the control effort required. A compartmental deterministic model was constructed. Parameter values were estimated from current literature wherever possible. We assessed the sensitivity of estimates for the basic reproduction number on the population scale to varying assumptions for the unknown or uncertain parameters of: (mean) duration of carriership (1∕γ C), relative infectiousness of carriers (f), proportion of infections that result in carriership (p), and (mean) duration of immunity after natural infection (1∕γ R). Available incidence and (sero-)prevalence data were compared to model outputs to improve point estimates and ranges for these currently unknown or uncertain transmission-related parameters. The required vaccination coverage of an ideal vaccine to prevent major outbreaks under a range of control scenarios was estimated, and compared available data on existing vaccines. The relative infectiousness of carriers (as compared to acutely ill horses) and the duration of carriership were identified as key knowledge gaps. Deterministic compartmental simulations, combined with seroprevalence data, suggest that 0.05<fˆ<0.5 and that the duration of protective immunity after infection is likely 4-6 years. The presence of carriers alone may suffice to keep S. equi endemic in a population, implying that carriers cannot be ignored in control efforts. Weekly screening of herds for signs of strangles could be sufficient to ensure R < 1, provided all horses are screened for carriership post-infection. In some of worst-case scenarios, vaccination alone would not suffice to prevent major outbreaks from occurring. A stochastic agent-based model was also constructed and validated, and used to simulate a remount depot, to evaluate whether historical incidence data of recurrence of strangles within individuals could be explained without the assumption that one in four horses fail to mount a lasting immune response. These simulations demonstrated that the observed data could have occurred without that assumption

Design of a multicentered randomized controlled trial on the clinical and cost effectiveness of schema therapy for personality disorders

Abstract

Background

Despite international guidelines describing psychotherapy as first choice for people with personality disorders (PDs), well-designed research on the effectiveness and cost-effectiveness of psychotherapy for PD is scarce. Schema therapy (ST) is a specific form of psychological treatment that proved to be effective for borderline PD. Randomized controlled studies on the effectiveness of ST for other PDs are lacking. Another not yet tested new specialized treatment is Clarification Oriented Psychotherapy (COP). The aim of this project is to perform an effectiveness study as well as an economic evaluation study (cost effectiveness as well as cost-utility) comparing ST versus COP versus treatment as usual (TAU). In this study, we focus on avoidant, dependent, obsessive-compulsive, paranoid, histrionic and narcissistic PD.

Methods/Design

In a multicentered randomized controlled trial, ST, and COP as an extra experimental condition, are compared to TAU. Minimal 300 patients are recruited in 12 mental health institutes throughout the Netherlands, and receive an extensive screening prior to enrolment in the study. When eligible, they are randomly assigned to one of the intervention groups. An economic evaluation and a qualitative research study on patient and therapist perspectives on ST are embedded in this trial. Outcome assessments (both for clinical effectiveness and economic evaluation) take place at 6,12,18,24 and 36 months after start of treatment. Primary outcome is recovery from PD; secondary measures include general psychopathological complaints, social functioning and quality of life. Data for the cost-effectiveness and cost-utility analyses are collected by using a retrospective cost interview. Information on patient and therapist perspectives is gathered using in-depth interviews and focus groups, and focuses on possible helpful and impeding aspects of ST.

Discussion

This trial is the first to compare ST and COP head-to-head with TAU for people with a cluster C, paranoid, histrionic and/or narcissistic PD. By combining clinical effectiveness data with an economic evaluation and with direct information from primary stakeholders, this trial offers a complete and thorough view on ST as a contribution to the improvement of treatment for this PD patient group.

Trial registration

Netherlands Trial Register (NTR): NTR566</p
