    Function and Dysfunction in Distinct Facets of Empathy

    Empathy is crucial for successful social interactions and it is impaired in many devastating disorders. Empathy deficits are highly burdensome for affected individuals, caregivers, and significant others, and costly for society as a whole. However, empathy is thought to be a multifaceted construct, including cognitive empathy, affective sharing, and empathic concern components. These constituents may be linked to different behavioural outcomes and neurocognitive substrates, and presentation varies depending on the facets affected. Thus, there is a critical need to determine the behavioural and neurocognitive substrates of different components of empathic responding, and how these are affected in particular disorders. The present work aimed to elucidate the nature of different components of empathy and how they vary as a function of clinical diagnoses and individual differences in subclinical traits, as well as their underlying functional neural mechanisms. Study I used the Multifaceted Empathy Test, a performance-based task tapping cognitive empathy, affective sharing, and empathic concern elicited by realistic emotional images, in patients with behavioural variant frontotemporal dementia (bvFTD). This revealed a global cognitive empathy deficit, deficient affective sharing for negative experiences, and a generalized processing impairment for negative stimuli in bvFTD. In Study II, healthy adults completed the Multifaceted Empathy Test and questionnaire measures of autistic traits, coldhearted psychopathic traits, and trait anxiety. Coldhearted traits were found to disrupt affective sharing and empathic concern, whereas trait anxiety appeared to influence subjective affective experience via generalized arousal. Study III investigated the involvement of action-perception matching, simulation mechanisms in cognitive versus emotional empathy, using fMRI during cognitive empathy, emotional empathy, and simulation network localizer tasks in healthy adults. Increased activation was observed in identified simulation regions during emotional versus cognitive empathy, providing evidence for greater involvement of simulation mechanisms in emotional empathy. Taken together, this work suggests that cognitive empathy, and emotional empathy, including affective sharing and empathic concern, represent aspects of empathy that are distinguishable and differentially linked with certain patient populations, subclinical traits, and neurocognitive mechanisms. These findings are discussed with respect to the nature and conceptualization of empathy and its components, as well as implications for disorders featuring empathy dysfunction

    Mitigating uncertainties in mineral exploration targeting: Majority voting and confidence index approaches in the context of an exploration information system (EIS)

    Various mineral prospectivity modelling (MPM) approaches are available for targeting mineral deposits, each method capable of predicting areas of high prospectivity. Given the diversity of MPM approaches, the modelled areas of high prospectivity can differ across different MPMs. However, rather than a negative, different MPM outputs can benefit mineral exploration targeting because each method has its advantages. Rather, the problem lies in the lack of consensus over how to best select and delimit mineral exploration targets from different MPM results. Here we aim to address the challenges outlined above whilst quantifying and mitigating the effects of inherent uncertainties. We first generate eleven different prospectivity models utilising deep learning, machine learning, fuzzy logic, and geometric average integration methods. Then, we adopt a majority voting ensemble technique to incorporate and combine the predictions of each prospectivity model. Next, we propose a confidence index designed to mitigate uncertainty associated with our multi-technique approach to MPM. The confidence index quantifies variation in prospectivity values for each cell of the MPM target area. The conjunction of a confidence index and majority voting model facilitates consistent and robust algorithm-driven extraction of exploration targets based on an ensemble of prospectivity models

    Rind from purple mangosteen (Garcinia mangostana) attenuates diet-induced physiological and metabolic changes in obese rats

    The pulp of the purple mangosteen, Garcinia mangostana, is a popular tropical fruit but the rind containing xanthones such as α-mangostin together with procyanidins and anthocyanidins is usually discarded as waste. However, this rind has been used in South-East Asia for diarrhoea, dysentery, skin infections and wounds. As xanthones have reported anti-inflammatory and antioxidant responses, this study has determined the bioactive compounds and evaluated the effects of G. mangostana rind on physiological, metabolic, liver and cardiovascular parameters in rats with diet-induced metabolic syndrome. Rats fed a diet with increased simple sugars and saturated fats developed obesity, hypertension, increased left ventricular stiffness, dyslipidaemia and fatty liver. Administration of G. mangostana rind as 5% of the food to rats with diet-induced metabolic syndrome gave a dose of 168 mg/kg/day α-mangostin, 355 mg/kg/day procyanidins, 3.9 mg/kg/day anthocyanins and 11.8 mg/kg/day hydroxycitric acid for 8 weeks which reduced body weight and attenuated physiological and metabolic changes in rats including decreased abdominal fat deposition, decreased abdominal circumference and whole-body fat mass, improved liver structure and function and improved cardiovascular parameters such as systolic blood pressure, left ventricular stiffness and endothelial function. These responses were associated with decreased infiltration of inflammatory cells, decreased deposition of collagen in both heart and liver and decreased mean adipocyte size in retroperitoneal adipose tissues. We conclude that, in rats with diet-induced metabolic syndrome, chronic intake of G. mangostana rind decreased infiltration of inflammatory cells which decreased physiological, metabolic, liver and cardiovascular symptoms

    Anthocyanins in chronic diseases : the power of purple

    Anthocyanins are mainly purple-coloured phenolic compounds of plant origin that as secondary metabolites are important in plant survival. Understanding their health benefits in humans requires sourcing these unstable compounds in sufficient quantities at a reasonable cost, which has led to improved methods of extraction. Dark-coloured fruits, cereals and vegetables are current sources of these compounds. The range of potential sustainable sources is much larger and includes non-commercialised native plants from around the world and agri-waste containing anthocyanins. In the last 5 years, there have been significant advances in developing the therapeutic potential of anthocyanins in chronic human diseases. Anthocyanins exert their beneficial effects through improvements in gut microbiota, oxidative stress and inflammation, and modulation of neuropeptides such as insulin-like growth factor-1. Their health benefits in humans include reduced cognitive decline; protection of organs such as the liver, as well as the cardiovascular system, gastrointestinal tract and kidneys; improvements in bone health and obesity; and regulation of glucose and lipid metabolism. This review summarises some of the sources of anthocyanins and their mechanisms and benefits in the treatment of chronic human diseases

    Physiological and metabolic effects of yellow mangosteen (Garcinia dulcis) rind in rats with food-induced metabolic syndrome

    Metabolic syndrome is a cluster of disorders that increase the risk of cardiovascular disease and diabetes. This study has investigated the responses to rind of yellow mangosteen (Garcinia dulcis), usually discarded as waste, in a rat model of human metabolic syndrome. The rind contains higher concentrations of phytochemicals (such as garcinol, morelloflavone and citric acid) than the pulp. Male Wistar rats aged 8–9 weeks were fed either corn starch diet or high-carbohydrate, high-fat diet for 16 weeks, which were supplemented with 5% freeze-dried G. dulcis fruit rind powder during the last 8 weeks. We characterised metabolic, cardiovascular, liver and gut microbiota parameters. High-carbohydrate, high-fat diet-fed rats developed abdominal obesity, hypertension, increased left ventricular diastolic stiffness, decreased glucose tolerance, fatty liver and reduced Bacteroidia with increased Clostridia in the colonic microbiota. G. dulcis fruit rind powder attenuated these changes, improved cardiovascular and liver structure and function, and attenuated changes in colonic microbiota. G. dulcis fruit rind powder may be effective in metabolic syndrome by appetite suppression, inhibition of inflammatory processes and increased fat metabolism, possibly related to changes in the colonic microbiota. Hence, we propose the use of G. dulcis fruit rind as a functional food to ameliorate symptoms of metabolic syndrome

    Neural effects of oxytocin and mimicry in frontotemporal dementia: A randomized crossover study

    OBJECTIVE: To determine whether intranasal oxytocin, alone or in combination with instructed mimicry of facial expressions, would augment neural activity in patients with frontotemporal dementia (FTD) in brain regions associated with empathy, emotion processing, and the simulation network, as indexed by blood oxygen-level dependent (BOLD) signal during fMRI. METHODS: In a placebo-controlled, randomized crossover design, 28 patients with FTD received 72 IU intranasal oxytocin or placebo and then completed an fMRI facial expression mimicry task. RESULTS: Oxytocin alone and in combination with instructed mimicry increased activity in regions of the simulation network and in limbic regions associated with emotional expression processing. CONCLUSIONS: The findings demonstrate latent capacity to augment neural activity in affected limbic and other frontal and temporal regions during social cognition in patients with FTD, and support the promise and need for further investigation of these interventions as therapeutics in FTD.     Single Eye mRNA-Seq Reveals Normalisation of the Retinal Microglial Transcriptome Following Acute Inflammation

    Background: Whether retinal microglia can maintain or restore immune homeostasis during and after inflammation is unclear. We performed single-eye mRNA-sequencing on microglia at different timepoints following a single inflammatory stimulus to characterise their transcriptome during and after resolution of endotoxin-induced uveitis (EIU). / Experimental Approach: Cx3cr1CreER:R26-tdTomato (C57BL/6) male heterozygotes were administered tamoxifen via different regimes at 4–5 weeks of age. Four weeks post-tamoxifen, mice were injected intravitreally with 10 ng lipopolysaccharide (endotoxin induced uveitis, EIU). Six-hundred retinal microglia were obtained by FACS from individual naïve retinas and at 4 h, 18 h, and 2 weeks following EIU induction. Samples were sequenced to a depth of up to 16.7 million reads using the SMART-Seq v4 Ultra Low Input RNA kit. The data was analysed using Partek software and Ingenuity Pathway Analysis. Genes were considered differentially-expressed (DEG) if the FDR step-up p-value was ≤0.05 and the fold-change was ≥±2. / Results: Flow cytometric analysis indicates that the Cx3cr1CreER:R26-tdTomato strain is both sensitive (>95% tagging) and specific (>95% specificity) for microglia when tamoxifen is administered topically to the eye for 3 days. During “early” activation, 613 DEGs were identified. In contrast, 537 DEGs were observed during peak cellular infiltrate and none at 2 weeks, compared to baseline controls (1,069 total unique DEGs). Key marker changes were validated by qPCR, flow cytometry, and fluorescence microscopy. C5AR1 was identified and validated as a robust marker of differentiating microglial subsets during an LPS response. / Conclusion: Using EIU to provide a single defined inflammatory stimulus, mRNA-Seq identified acute transcriptional changes in retinal microglia which returned to their original transcriptome after 2 weeks. Yolk-sac derived microglia are capable of restoring their homeostatic state after acute inflammation

    Genetic Factors and Orofacial Motor Learning Selectively Influence Variability in Central Sulcus Morphology in Chimpanzees (Pan troglodytes)

    International audienceCaptive chimpanzees (Pan troglodytes) have been shown to learn the use of novel attention-getting (AG) sounds to capture the attention of humans as a means of requesting or drawing their attention to a desired object or food. There are significant individual differences in the use of AG sounds by chimpanzees and, here, we examined whether changes in cortical organization of the central sulcus (CS) were associated with AG sound production. MRI scans were collected from 240 chimpanzees, including 122 that reliably produced AG sounds and 118 that did not. For each subject, the depth of CS was quantified along the superior-inferior plane with specific interest in the inferior portion corresponding to the region of the motor cortex where the mouth and orofacial movements are controlled. Results indicated that CS depth in the inferior, but not superior, portion was significantly greater in chimpanzees that reliably produced AG sounds compared with those who did not. Quantitative genetic analyses indicated that overall CS surface area and depth were significantly heritable, particularly in the superior regions, but less so in the inferior and central portions. Further, heritability in CS depth was altered as a function of acquisition of AG sounds. The collective results suggest that learning to produce AG sounds resulted in region-specific cortical reorganization within the inferior portion of the CS, a finding previously undocumented in chimpanzees or any nonhuman primate
