    Biotransformation of digitoxigenin by Cochliobolus lunatus

    The biotransformation of digitoxigenin (1) by Cochliobolus lunatus was investigated. The biotransformation reaction was carried out in a 4-day process, resulting in the isolation of four products, whose structures were elucidated as 1beta-hydroxydigitoxigenin (2), 7beta-hydroxydigitoxigenin (3), 8beta-hydroxydigitoxigenin (4) and digitoxigenone (5). The production of these derivatives under the employed conditions has never been described so far. This is also the first report on the production of compound 4 by a biotransformation reaction

    Effect of cerium-containing hydroxyapatite in bone repair in female rats with osteoporosis induced by ovariectomy

    Osteoporosis is a public health problem, with bone loss being the main consequence. Hydroxyapatite (HA) has been largely used as a bioceramic to stimulate bone growth. In our work, a cerium-containing HA (Ce-HA) has been proposed and its effects on the antimicrobial and bone-inducing properties were investigated. The synthesis of the materials occurred by the suspension–precipitation method (SPM). The XRD (X-ray Diffraction) confirmed the crystalline phase, and the Rietveld refinement confirmed the crystallization of HA and Ce-HA in a hexagonal crystal structure in agreement with ICSD n◦ 26205. Characterizations by FT-IR (Fourier Transform Infrared Spectroscopy), XPS (X-ray Photoemission Spectroscopy), and FESEM-EDS (Field Emission Scanning Electron Microscope-Energy Dispersive X-ray Spectroscopy) confirmed the presence of cerium (Ce3+ and Ce4+ ). The antibacterial activity of Has was evaluated against Staphylococcus aureus 25,923 and Escherichia coli 25,922 strains, which revealed that the material has antimicrobial properties and the cytotoxicity assay indicated that Ce-containing HA was classified as non-toxic. The effects of Ce-HA on bone repair, after application in bone defects in the tibia of female rats with osteoporosis induced by ovariectomy (OVX), were evaluated. After 15 and 30 days of implantation, the samples were analyzed by Raman, histology and X-ray microtomography.     O morangueiro requer mão-de-obra qualificada e bastantes cuidados em seu manejo. O cultivo dessa fruta no Brasil ocorre, principalmente nos estados do Sul e Sudeste do país. Na região Nordeste do país pouco se planta, principalmente em Pernambuco, devido principalmente ao clima. Esta fruta que apresenta grande retorno financeiro gerando empregos e divisas para estados e municípios. A descoberta de locais propícios ao cultivo poderá gerar maior desenvolvimento local. Visando expansão da fronteira agrícola do morango, neste trabalho objetivou-se avaliar a cultivar mais adaptada a Região Metropolitana do Recife. Foram testadas 8 cultivares de morangueiro com 8 repetições. Avaliou-se a emissão de estolhos e produtividade para a seleção da cultivar mais adaptada ao clima quente e úmido. As estimativas de produtividade de todos os tratamentos estiveram abaixo da média nacional para o cultivo do morangueiro, fator que se deve ao plantio em época quente

    Grasslands are the major source of feed for ruminants (Zanine 2005). Seasonality of production, however, is a constraint in forage-based systems. Silvopastoril systems combine different components (animals, trees, and forages) into one integrated system and may improve forage distribution across seasons. Resource use is usually more efficient both spatially and temporally, increasing land use efficiency (Nair 1993). Tree legumes present potential for silvopastoril systems because they can fix N from the atmosphere, improve cattle diet, and lead to a faster N cycle. In addition, trees provide shade and may reduce heat stress for grazing animals in warm-climate grasslands. Legume trees are commonly found in warm-season climates and present potential for use in silvopastoril systems. Polymeric nanoparticles have increasing demands in pharmaceutical applications and require high reproducibility, homogeneity, and control over their properties. Work explores the use of cashew phthalate gum (PCG) as a particle-forming polymer. PCG exhibited a pH-sensitive behavior due to the of acid groups on its chains, and control drug release. We report the development of nanoparticles carrying benznidazole. Formulations were characterized by DLS, encapsulation efficiency, drug loading, FTIR, pH-responsive behavior, release, and in vitro kinetics. Interaction between polymer and drug was an evaluated by molecular dynamics. Morphology was observed by SEM, and in vitro cytotoxicity by MTT assay. Trypanocidal effect for epimastigote and trypomastigote forms was also evaluated. NPs responded to the slightly basic pH, triggering the release of BNZ. In acidic medium, they presented small size, spherical shape, and good stability. It was indicated NP with enhanced biological activity, reduced cytotoxicity, high anti T. cruzi performance, and pH-sensitive release. This work investigated properties related to the development and enhancement of nanoparticles. PCG has specific physicochemical properties that make it a promising alternative to drug delivery, however, there are still challenges to be overcome.publishersversionpublishe

    Os efeitos dos estrogênios conjugados equinos e do tamoxifeno na histomorfologia da glândula tireóide de ratas

    OBJECTIVE: The aim of this study was to evaluate the action of the conjugated equine estrogens and tamoxifen on the morphology of thyroid gland in ovariectomized (OVx) rats. METHODS: Conjugated equine estrogens (CEE), clinically used as estrogen therapy, is a complex formulation containing multiple estrogens that decrease menopausal symptoms. Thirty ovariectomized rats were randomly divided into 3 treatment groups: GI, vehicle (propylene glycol); GII, CEE 200 &micro;g/kg per day; and GIII, tamoxifen 1 mg/kg per day. Another group of 10 rats with intact ovaries (GIV) was included, treated with the vehicle, and sacrificed during estrous. All animals were treated by gavage for 50 days, after which they were sacrificed. Blood samples were collected, and the thyroid was removed for morphological analysis and PCNA evaluation through immunohistochemical study. RESULTS: The thyroid follicular cell height was increased in animals treated with CEE (14.90 &plusmn; 0.20 µm), with TAM (14.90 &plusmn; 0.10 µm), and in rats with intact ovaries (15.10 &plusmn; 0.50 µm) in comparison to that of the vehicle group (9.90 &plusmn; 0.20 µm) (P < 0.001). The follicular area was larger in the CEE (2,225 &plusmn; 51 µm²) and TAM (2,127 &plusmn; 67 µm²) groups compared to that of the vehicle group (5,016 &plusmn; 53 &micro;m²). The levels of T4 and T3 in rats treated with CEE, with Tamoxifen and in rats with intact ovaries, were higher than those those in the vehicle group (P < 0.001). The PCNA index in the vehicle group was lower than in other groups. CONCLUSION: Our data suggest that estrogen and tamoxifen administration has a proliferative effect on the thyroid.OBJETIVO: Avaliar a ação dos estrogênios conjugados eqüinos e do tamoxifeno na histomorfologia da tireóide de ratas. MÉTODO: Estrogênios conjugados eqüinos são ministrados clinicamente como terapia estrogênica e contêm formulação complexa com muitos tipos de estrogênios que diminuem os sintomas da pós-menopausa. Trinta ratas adultas ooforectomizadas foram divididas aleatoriamente em três grupos: GI - veículo (propilenoglicol); GII - ECE 200 &micro;g/Kg por dia; e GIII - TAM 1 mg/Kg por dia. Acrescentou-se ainda um grupo de 10 animais com os ovários intactos e tratados com veículo (GIV). Todos os animais foram tratados por gavagem durante 50 dias consecutivos, ao final foram coletadas amostras do sangue e a tireóide removida e processada para análise morfológica e imunohistoquímico para avaliar o PCNA. RESULTADOS: A maior altura das células foliculares foi observada nos animais tratados com ECE (14,90 &plusmn; 0,20 µm), TAM (14,90 &plusmn; 0,10 µm) e no grupo com ovários intactos (15,10 &plusmn; 0,50 µm), comparando-se aos controles ovariectomizados (GI) (9,90 &plusmn; 0,20 µm) (
