Reconstructing the Density of States by History-Dependent Metadynamics
We present a novel method for the calculation of the energy density of states
D(E) for systems described by classical statistical mechanics. The method
builds on an extension of a recently proposed strategy that allows the free
energy profile of a canonical system to be recovered within a pre-assigned
accuracy,[A. Laio and M. Parrinello, PNAS 2002]. The method allows a good
control over the error on the recovered system entropy. This fact is exploited
to obtain D(E) more efficiently by combining measurements at different
temperatures. The accuracy and efficiency of the method are tested for the
two-dimensional Ising model (up to size 50x50) by comparison with both exact
results and previous studies. This method is a general one and should be
applicable to more realistic model systems
Poredbene hematološke i biokemijske analize divovske amazonske kornjače uzgajane pod lošim i normalnim hranidbenim uvjetima
Besides indicating the baseline values of the species, blood parameter assessments of chelonian may also be used as quick tools for diagnosing health status. An investigation was carried out to assess and compare red blood cells parameters, white blood cell (WBC) and total thrombocyte counts and certain blood biochemical parameters for freshwater turtles, Podocnemis expansa Schweigger, 1812 (Pelomedusidae) reared on normal (control group) and poor nutrition (malnourished group). For the malnourished turtles, a signififi cant decrease (P<0.05) in red blood cell counts was found, including hematocrit, plasma glucose, plasma total protein, cholesterol and urea levels, as well as WBC counts, azurophils and heterophils. Malnutrition did not alter the hemostasis, but caused severe normocytic-hypocromic anemia and marked immune depression, which were diagnosed here. This study was the fifi rst to characterize the physiological and immune status of giant turtles from
the Amazon under adverse nutritional conditions.
Relationship between muscle strength and motor function in Duchenne muscular dystrophy
Measuring muscle strength and motor function is part of Duchenne muscular dystrophy (DMD) assessment. However, the relationship between these variables is controversial. Objective: To investigate the relationship between muscle strength and motor function and between these variables and age. Method: Muscle strength was measured by Medical Research Council (MRC) scale and motor function, by Motor Function Measure (MFM), in 40 non-ambulatory patients. Spearman tests investigated the relationships between muscle strength, motor function and age. Results: Total MRC and MFM scores were strongly related to each other (r = 0.94p 0.05). Strong and moderate relationships between partial muscle strength and motor function scores were found. Higher correlation coefficients were found between total scores and Dimensions 2 (axial/ proximal control) and 3 (distal control) of MFM. Conclusion: Muscle strength and motor function are strongly correlated and seem to decrease proportionally in DMD.Univ Sao Paulo, Fac Med, Dept Fisioterapia Fonoaudiol & Terapia Ocupac, Sao Paulo, SP, BrazilUniv Fed Sao Paulo, Fac Med, Dept Neurol, Sao Paulo, SP, BrazilUniv Fed Sao Paulo, Fac Med, Dept Neurol, Sao Paulo, SP, BrazilWeb of Scienc
Electrospun nanofibers of polyCD/PMAA polymers and their potential application as drug delivery system
Herein, we used an electrospinning process to develop highly efficacious and
hydrophobic coaxial nanofibers based on poly-cyclodextrin (polyCD) associated
with poly(methacrylic acid) (PMAA) that combines polymeric and supramolecular
features for modulating the release of the hydrophilic drug, propranolol
hydrochloride (PROP). For this purpose, polyCD was synthesized and
characterized, and its biocompatibility was assessed using fibroblast
cytotoxicity tests. Moreover, the interactions between the guest PROP molecule
and both polyCD and CD were found to be spontaneous. Subsequently, PROP
was encapsulated in uniaxial and coaxial polyCD/ PMAA nanofibers. A lower PROP
burst effect (reduction of approximately 50%) and higher modulation were
observed from the coaxial than from the uniaxial fibers. Thus, the coaxial
nanofibers could potentially be a useful strategy for developing a controlled
release system for hydrophilic molecules.Comment: 10 pages, 8 figures + Supplementary Data (4 pages, 6 figures
THEORY OF THE SOCIAL REPRESENTATIONS IN THE SCOPE OF THE AGING: ANINTEGRATIVE REVIEW
Ordering dynamics of the multi-state voter model
The voter model is a paradigm of ordering dynamics. At each time step, a
random node is selected and copies the state of one of its neighbors.
Traditionally, this state has been considered as a binary variable. Here, we
relax this assumption and address the case in which the number of states is a
parameter that can assume any value, from 2 to \infty, in the thermodynamic
limit. We derive mean-field analytical expressions for the exit probability and
the consensus time for the case of an arbitrary number of states. We then
perform a numerical study of the model in low dimensional lattices, comparing
the case of multiple states with the usual binary voter model. Our work
generalizes the well-known results for the voter model, and sheds light on the
role of the so far almost neglected parameter accounting for the number of
states
Methylene tetrahydrofolate reductase (MTHFR) and vascular endothelial growth factor (VEGF) polymorphisms in Brazilian patients with Hepatitis C virus (HCV)-related hepatocellular carcinoma (HCC)
OBJECTIVE: The folate pathway is involved in hepatic carcinogenesis and angiogenesis. Polymorphisms in genes related to such processes, including methylene tetrahydrofolate reductase (MTHFR) and vascular endothelial growth factor (VEGF)] may play an important role in the development of hepatocellular carcinoma (HCC). The objective of this study was to evaluate MTHFR and VEGF polymorphisms in Brazilian patients with hepatitis C virus (HCV)-related HCC.
METHODS: A total of 119 patients diagnosed with confirmed HCC and HCV were included in the study. SNP genotyping assays were performed using real-time PCR. VEGFA (rs2010963, rs3025039, and rs833061) and MTHFRC677T (rs1801133, rs1801131) polymorphisms were evaluated.
RESULTS: The C alleles of MTHFR (rs1801131) and VEGF (rs2010963) were associated with protection against the development of multinodular HCC, while the T allele of MTHFR (rs1801133) was associated with a higher risk of multinodular presentation [p=0.04 OR 1.835 CI (1.022-3.297)]. Multivariate analysis revealed that the GG/GC genotypes of VEGF rs2010963 were independently associated with multinodular tumors at diagnosis (p=0.013; OR 4.78 CI (1.38-16.67)].
CONCLUSION: Our results suggest that these polymorphisms may increase the risk of rapid tumor progression in patients with HCV infection. This subgroup of patients with HCC and who present polymorphism is more likely to be diagnosed with multinodular disease and not be amenable to receiving curative treatments. These data must be validated in larger cohorts, and the screening intervals can be customized based on genetic history
Co-infecção pelo vírus dengue 3 e 4 em pacientes da Amazônia brasileira
The natural co-infection with dengue virus can occur in highly endemic areas where different serotypes have been observed for many years. We report here four cases of DENV-3/DENV-4 co-infection detected by serological and molecular tests among 674 patients with acute undifferentiated fever from the tropical medicine reference center of Manaus City, Brazil, between 2005 and 2010. Analysis of the sequences obtained indicated the presence of genotype 3 and 1 for DENV-3 and DENV-4 respectively.A co-infecção natural com os vírus dengue pode ocorre em áreas altamente endêmicas onde diferentes sorotipos têm sido transmitidos por muitos anos. Hepatitis E virus infection increases the risk of diabetes and severity of liver disease in patients with chronic hepatitis C virus infection
OBJECTIVES: Co-infection with hepatitis A or B viruses may aggravate liver injury in patients infected with hepatitis C virus (HCV). However, few studies have assessed co-infection with hepatitis E virus (HEV) and HCV. Therefore, this study aimed to assess the prevalence and impact of HEV infection among Brazilian patients with chronic HCV infection.
METHODS: This observational study included adult patients with chronic HCV infection who were naive to antiviral therapy from January 2013 to March 2016. A total of 181 patients were enrolled, and HEV serology and PCR were performed for all patients.
RESULTS: Seropositivity for anti-HEV IgG was detected in 22 (12.0%) patients and anti-HEV immunoglobulin M in 3 (1.6%). HEV RNA showed inconclusive results in nine (4.9%) patients and was undetectable in the remaining patients. HEV serology positive patients had more severe liver disease, characterized by liver fibrosis ≥3 versus ≤2 (p<0.001), Aspartate Aminotransferase-to-Platelet Ratio Index of ≥1.45 (p=0.003), and Fibrosis-4 score of ≥3.25 (p=0.001). Additionally, the odds of HEV-positive patients developing diabetes mellitus were 3.65 (95% CI 1.40-9.52) times the corresponding odds of HEV-negative patients. A case-control-based histological analysis (n=11 HEV-HCV-positive patients and n=22 HCV-positive patients) showed no significant differences between the groups.
CONCLUSIONS: This prevalence is higher than that reported in previous studies of the general population in Brazil. Thus, HEV infection may influence the severity of liver disease and may represent an additional risk of developing diabetes mellitus in patients with HCV infection
