    Get PDF
The paper deals with the issues in program management used for engineering innovative products. The existing project management tools were analyzed. The aim is to develop a decision support system that takes into account the features of program management used for high-tech products: research intensity, a high level of technical risks, unpredictable results due to the impact of various external factors, availability of several implementing agencies. The need for involving experts and using intelligent techniques for information processing is demonstrated. A conceptual model of common information space to support communication between members of the collaboration on high-tech programs has been developed. The structure and objectives of the information analysis system "Geokhod" were formulated with the purpose to implement the conceptual model of common information space in the program "Development and production of new class mining equipment - "Geokhod"

AbstractThe technique of fluorescence recovery after photobleaching was used to measure the lateral mobility of membrane integral proteins in reticulocyte plasma membranes which were treated to modify the ‘fluid’ lipid or immobilized protein fractions, hence increasing the relative prevalence of obstacles to protein lateral motion. This was achieved by either: (1) treating the plasma membranes with phospholipase A2 followed by extraction of the hydrolysis products using fatty-acid-free bovine serum albumin, resulting in a decrease in the membrane ‘fluid’ lipid portion; or (2) preincubating the plasma membranes with polylysines, resulting in plasma membrane protein aggregation and immobilization. As the prevalence of obstacles to lateral motion increased in plasma membranes through the treatments described above, the mobility of the membrane integral proteins diminished. Experimental results for the dependence of protein mobility on the prevalence of obstacles to lateral motion were compared to theoretical data in order to verify the applicability of the percolation theory to reticulocyte plasma membranes. The influence of a decrease in the ‘fluid’ lipid and an increase in the immobilized membrane protein fractions upon the hormone-stimulated adenylate cyclase activity has been studied as well. As the ‘solid’ lipid and immobilized membrane protein fractions decreased, both the hormone-stimulated adenylate cyclase activity and the fraction of β-adrenergic receptors with high affinity to hormone diminished. It was shown that this correlation can be caused by a decrease in membrane fraction accessible to the movement of the interacting proteins of the adenylate cyclase complex. Hormonal stimulation of adenylate cyclase is discussed in terms of the percolation theory

Background. The development of psychological practices has emerged such promising method for evaluating and transforming the value semantic sphere of the individual as film therapy. However, the wide dissemination of the method is held up by insufficient knowledge of the mechanisms that make up its psychological impact. The urgent task is to elaborate on the projective method for diagnosing the value semantic sphere of the individual by means of artistic cinematography, and also to develop the technologies for quasi-forming and learning technique. The Objective is to analyze the possibilities of using film therapy for solving psychotherapeutic, diagnostic, correctional, and developmental problems. Design. The theoretical bases of film therapy as a kind of art therapy are considered, similarities and differences with bibliotherapy are emphasized. The paper discusses the development of film therapy as a method with a wide range of tasks from coping with emotional disturbances and restoring the balance of psychodynamic forces to a wide array of issues including optimizing personal development and education. Psychological impact mechanisms in the process of cinema therapy, i.e. projection, identification with the hero, reflection, catharsis, social learning are analyzed. Basic methods of personal transformation used in film therapy are singled out. Basic topics for group and individual discussion after watching a feature film are identified. The results of a pilot study that proved the high efficiency of the film therapy method in pregnant women are presented. Research results. Based on the analysis of the psychological practice using artistic cinematography as a method of psychological influence the following tips should be laid emphasis on. Although the method of film therapy has successfully proved itself in solving psychotherapeutic, developmental, and educational problems, the psychological impact mechanisms have not received enough focus, and the possibilities of film therapy in the development of personal potential are far from being fully employed. Conclusion. Prospects for the development of this method are associated with the development of projective methods for investigating the unconscious implicit attitudes of the individual, which will increase the effectiveness of diagnostic work, and also the development of quasi-forming techniques of psychological impact on the human value sphere enhancing their personal potential and taking into account the actual development challenges of each age group

A criterion of colorectal cancer diagnosis using exosome fluorescence-lifetime imaging

This study was aimed to investigate the applicability of the exosome fluorescence-lifetime imaging microscopy (FLIM) for colorectal cancer (CRC) diagnosis. Differential ultra-centrifugation was used to extract exosomes from the blood plasma of 11 patients with colon polyps (CPs) and 13 patients with CRC at the T2-4, N0-3, and M0-1 stages. Analysis was performed using a two-photon FLIM device. In total, 165 and 195 FLIM images were recorded for the CP and CCR patient groups, respectively. Two classes of exosomes differentiated by autofluorescence average lifetime tm were discovered in the samples. The first class of exosomes with tm = (0.21 0.06) ns was mostly found in samples from CRC patients. The second class with tm = (0.43 0.19) ns was mostly found in samples from CP patients. The relative number of “CRC-associated” exosomes Nch in the FLIM dataset was shown to be very small for the CP patient group and large for the CRC patient group. This difference was statistically significant. Therefore, the suggested CRS diagnostics criterion can be as follows. If Nch > 0.5, the probability of CRC is high. If Nch < 0.3, the probability of CRC is low

In this work chaotic dynamics contact interaction of two flexible Tymoshenko beams, under the action of a transversal alternating load is investigated. The contact interaction of the beams is taken into account by the Kantor model. The geometric nonlinearity is taken into account by the model of T. von Karman. The system of partial differential equations of the twelfth order reduces to the system of ordinary differential equations by the method of finite differences of the second order. The resulting system by methods of Runge-Kutta type of the second, fourth and eighth orders was solved. Our theoretical/numerical analysis is supported by methods of nonlinear dynamics and the qualitative theory of differential equations. Chaotic vibrations of two flexible beams of Timoshenko were investigated and the optimal step values over the spatial coordinate and the time steps for the numerical experiment were found. Convergence for all applicable numerical methods have been achieved and shown that chaotic signals are true

Amphiphilic Polypeptides for VEGF siRNA Delivery into Retinal Epithelial Cells

Polyethyleneimine, poly-L-lysine, chitosan and some others cationic polymers have been thoroughly studied as nucleic acid delivery systems in gene therapy. However, the drug release from these systems proceeds at a very low rate due to extremely high binding between a carrier and gene material. To reduce these interactions and to enhance drug release, we developed a set of amphiphilic polypeptides containing positively and negatively charged amino acids as well as a hydrophobic one. The copolymers obtained were characterized by size-exclusion chromatography, static light scattering, HPLC amino acid analysis and (HNMR)-H-1 spectroscopy. All copolymers formed particles due to a self-assembly in aqueous media. Depending on polypeptide composition, the formation of particles with hydrodynamic diameters from 180 to 900 nm was observed. Stability of polymer particles, loading and release efficiency were carefully studied. Cellular uptake of the particles was efficient and their cytotoxicity was negligible. The application of polymer carriers, containing siRNA, to vascular endothelial growth factor (VEGF-A165) silencing of ARPE-19 cells was successful. The gene silencing was confirmed by suppression of both messenger RNA and protein expression.Peer reviewe
