    Antioxidant Activity and Oxidation Products of 1,2,3,4- Tetrahydroquinoxalines in Peroxyl Radical Scavenging Reactions, Part I

    This paper studies the antioxidant activity of 1,2,3,4-tetrahydroquinolines, 3,4-dihydro-2H-benzo[1,4]thiazines and 1,2,3,4-tetrahydroquinoxalines in the inhibition of the peroxidation of tetralin induced by an azo initiator. Neither 1,2,3,4- tetrahydroquinoline nor 3,4-dihydro-2H-benzo[1,4]thiazine alone acted as an antioxidant, but when they have an electron-donating group at the para position to the NH group, they act as potent antioxidants. On the other hand, 1,2,3,4- tetrahydroquinoxaline on its own showed good antioxidant activity. However, 1,2,3,4-tetrahydroquinoxalines with methyl and methoxy groups in the phenyl ring have reactivities similar to or less than that of unsubstituted 1,2,3,4-tetrahydroquinoxaline. The induction periods of 1,2,3,4-tetrahydroquinoxalines with an alkyl group or phenyl group at the 2-position were all longer than the value for the unsubstituted 1,2,3,4-tetrahydroquinoxaline, except for a compound with a t-butyl group.     ASTE CO(3-2) Mapping toward the Whole Optical Disk of M 83: Properties of Inter-arm GMAs

    We present a new on-the-fly (OTF) mapping of CO(J=3-2) line emission with the Atacama Submillimeter Telescope Experiment (ASTE) toward the 8' x 8' (or 10.5 x 10.5 kpc at the distance of 4.5 Mpc) region of the nearby barred spiral galaxy M 83 at an effective resolution of 25''. Due to its very high sensitivity, our CO(J=3-2) map can depict not only spiral arm structures but also spur-like substructures extended in inter-arm regions. This spur-like substructures in CO(J=3-2) emission are well coincident with the distribution of massive star forming regions traced by Halpha luminosity and Spitzer/IRAC 8 um emission. We have identified 54 CO(J=3-2) clumps as Giant Molecular-cloud Associations (GMAs) employing the CLUMPFIND algorithm, and have obtained their sizes, velocity dispersions, virial masses, and CO luminosity masses. We found that the virial parameter alpha, which is defined as the ratio of the virial mass to the CO luminosity mass, is almost unity for GMAs in spiral arms, whereas there exist some GMAs whose alpha are 3 -- 10 in the inter-arm region. We found that GMAs with higher α\alpha tend not to be associated with massive star forming regions, while other virialized GMAs are. Since alpha mainly depends on velocity dispersion of the GMA, we suppose the onset of star formation in these unvirialized GMAs with higher alpha are suppressed by an increase in internal velocity dispersions of Giant Molecular Clouds within these GMAs due to shear motion.Comment: 42 pages, 16 figures, ApJ in press, version with high resolution figures is available via http://www.nro.nao.ac.jp/~kmuraoka/m83paper/m83aste-otf.pd

    Serum Antibody Against NY-ESO-1 and XAGE1 Antigens Potentially Predicts Clinical Responses to Anti–Programmed Cell Death-1 Therapy in NSCLC

    Introduction: Programmed cell death-1 (PD-1) inhibitors effectively treat NSCLC and prolong survival. Robust biomarkers for predicting clinical benefits of good response and long survival with anti-PD-1 therapy have yet to be identified; therefore, predictive biomarkers are needed to select patients with benefits. Methods: We conducted a prospective study to explore whether serum antibody against NY-ESO-1 and/or XAGE1 cancer-testis antigens predicted primarily good clinical response and secondarily long survival with anti-PD-1 therapy for NSCLC. The serum antibody was detected by enzyme-linked immunosorbent assay, and tumor immune microenvironment and mutation burden were analyzed by immunohistochemistry and next-generation sequencing. Results: In the discovery cohort (n = 13), six antibody-positive NSCLC cases responded to anti-PD-1 therapy (two complete and four partial responses), whereas seven antibody-negative NSCLC cases did not. Antibody positivity was associated with good response and survival, regardless of tumor programmed death ligand 1 (PD-L1) expression, mutation burden, and CD8+ T-cell infiltration. In the validation cohort (n = 75), 17 antibody-positive NSCLC cases responded well to anti-PD-1 therapy as compared with 58 negative NSCLC cases (objective response rate 65% versus 19%, p = 0.0006) and showed significantly prolonged progression-free survival and overall survival. Antibody titers highly correlated with tumor reduction rates. In the multivariate analysis, response biomarkers were tumor programmed death ligand 1 expression and antibody positivity, and only antibody positivity was a significantly better predictive biomarker of progression-free survival (hazard ratio = 0.4, p = 0.01) and overall survival (hazard ratio = 0.2, p = 0.004). Conclusions: Our results suggest that NY-ESO-1 and/or XAGE1 serum antibodies are useful biomarkers for predicting clinical benefits in anti-PD-1 therapy for NSCLC and probably for other cancers

    Serum NY-ESO-1 antibody as a predictive biomarker for postoperative recurrence of gastric cancer: a multicenter prospective observational study

We designed a clinical trial to investigate the utility of serum NY-ESO-1 antibody responses as a predictive marker for postoperative recurrence in gastric cancer. Methods: A multicenter prospective study was conducted between 2012 and 2021. Patients with resectable cT3-4 gastric cancer were included. Postoperative NY-ESO-1 and p53 antibody responses were serially evaluated every 3 months for 1 year in patients with positive preoperative antibody responses. The recurrence rate was assessed by the positivity of antibody responses at 3 and 12 months postoperatively. Results: Among 1001 patients, preoperative NY-ESO-1 and p53 antibody responses were positive in 12.6% and 18.1% of patients, respectively. NY-ESO-1 antibody responses became negative postoperatively in non-recurrent patients (negativity rates; 45% and 78% at 3 and 12 months, respectively), but remained positive in recurrent patients (negativity rates; 9% and 8%, respectively). p53 antibody responses remained positive in non-recurrent patients. In multivariate analysis, NY-ESO-1 antibody positivity at 3 months (P < 0.03) and 12 months (P < 0.001) were independent prognostic factors for a shorter recurrence-free interval. Conclusions: Serum NY-ESO-1 antibodies may be a useful predictive marker for postoperative recurrence in gastric cancer. Clinical trial registration: UMIN000007925

    Unveiling the physical properties and kinematics of molecular gas in the Antennae Galaxies (NGC 4038/9) through high resolution CO (J = 3-2) observations

    We present a ~ 1" (100 pc) resolution 12CO (3-2) map of the nearby intermediate stage interacting galaxy pair NGC 4038/9 (the Antennae galaxies) obtained with the Submillimeter Array. We find that half the CO (3-2) emission originates in the overlap region where most of the tidally induced star formation had been previously found in shorter wavelength images, with the rest being centered on each of the nuclei. The gross distribution is consistent with lower resolution single dish images, but we show for the first time the detailed distribution of the warm and dense molecular gas across this galaxy pair at resolutions comparable to the size of a typical giant molecular complex. While we find that 58% (33/57) of the spatially resolved Giant Molecular Associations (GMAs; a few x 100 pc) are located in the overlap region, only \leqq 30% spatially coincides with the optically detected star clusters, suggesting that the bulk of the CO (3-2) emission traces the regions with very recent or near future star formation activity. The spatial distribution of the CO (3-2)/CO (1-0) integrated brightness temperature ratios mainly range between 0.3 and 0.8, which suggests that on average the CO (3-2) line in the Antennae is not completely thermalized and similar to the average values of nearby spirals. A higher ratio is seen in both nuclei and the southern complexes in the overlap region. Higher radiation field associated with intense star formation can account for the nucleus of NGC 4038 and the overlap region, but the nuclear region of NGC 4039 show relatively little star formation or AGN activities and cannot be easily explained. We show kinematical evidence that the high line ratio in NGC 4039 is possibly caused by gas inflow into the counter-rotating central disk.Comment: 20 pages, 12 figures, 4 tables, accepted for publication in Astrophysical Journa
