    Yellow-cedar blue intensity tree ring chronologies as records of climate, Juneau, Alaska, USA

Don; Oerst. ex D.P. Little). The latewood BI chronology shows a much stronger temperature sensitivity than ring-widths (RW), and thus can provide information on past climate. The well-replicated BI chronology exhibits a positive January-August average maximum temperature signal for 1900-1975, after which it loses temperature sensitivity following the 1976/77 shift in northeast Pacific climate. The positive temperature response appears to recover and remains strong for the most recent decades although the coming years will continue to test this observation. This temporary loss of temperature sensitivity from about 1976 to 1999 is not evident in RW or in a change in forest health, but is consistent with prior work linking cedar decline to warming. A confounding factor is the uncertain influence of a shift in color variation from the heartwood/sapwood boundary. Future expansion of the yellow-cedar BI network and further investigation of the influence of the heartwood/sapwood transitions in the BI signal will lead to a better understanding of the utility of this species as a climate proxy.PostprintPeer reviewe

    The association of homeobox gene expression with stem cell formation and morphogenesis in cultured Medicago truncatula

    Somatic embryogenesis (SE) is induced in vitro in Medicago truncatula 2HA by auxin and cytokinin but rarely in wild type Jemalong. The putative WUSCHEL (MtWUS), CLAVATA3 (MtCLV3) and the WUSCHEL-related homeobox gene WOX5 (MtWOX5) were investigated in M. truncatula (Mt) and identified by the similarity to Arabidopsis WUS, CLV3 and WOX5 in amino acid sequence, phylogeny and in planta and in vitro expression patterns. MtWUS was induced throughout embryogenic cultures by cytokinin after 24–48 h and maximum expression occurred after 1 week, which coincides with the induction of totipotent stem cells. During this period there was no MtCLV3 expression to suppress MtWUS. MtWUS expression, as illustrated by promoter-GUS studies, subsequently localised to the embryo, and there was then the onset of MtCLV3 expression. This suggests that the expression of the putative MtCLV3 coincides with the WUS-CLAVATA feedback loop becoming operational. RNAi studies showed that MtWUS expression is essential for callus and somatic embryo production. Based on the presence of MtWUS promoter binding sites, MtWUS may be required for the induction of MtSERF1, postulated to have a key role in the signalling required for SE induced in 2HA. MtWOX5 expressed in auxin-induced root primordia and root meristems and appears to be involved in pluripotent stem cell induction. The evidence is discussed that the homeobox genes MtWUS and MtWOX5 are “hijacked” for stem cell induction, which is key to somatic embryo and de novo root induction. In relation to SE, a role for WUS in the signalling involved in induction is discussed

    Item does not contain fulltextCONTEXT: The annual death rate of Prader-Willi syndrome (PWS) patients is very high (3%). Many of these deaths are sudden and unexplained. OBJECTIVE: Because most deaths occur during moderate infections and PWS patients suffer from various hypothalamic insufficiencies, we investigated whether PWS patients suffer from central adrenal insufficiency (CAI) during stressful conditions. DESIGN: Overnight single-dose metyrapone tests were performed. Metyrapone (30 mg/kg) was administered at 2330 h. At 0400, 0600, and 0730 h, ACTH, 11-deoxycortisol, cortisol, and glucose levels were measured. Diurnal salivary cortisol profiles were assessed on a different day at wake-up, 30 min after wake-up, at 1400 h, and at 2000 h. SETTING: The study was conducted in a pediatric intensive care unit. PATIENTS: Patients included 25 randomly selected PWS patients. MAIN OUTCOME MEASURE: Patients were considered as having CAI when ACTH levels remained below 33 pmol/liter at 0730 h. RESULTS: Median (interquartile range) age was 9.7 (6.8-13.6) yr. Fifteen patients (60%) had an insufficient ACTH response (CAI, P < 0.001). There was no significant difference in age, gender, genotype, and body mass index SD score between patients with CAI and those without. Morning salivary cortisol levels and diurnal profiles were normal in all children, suggesting that CAI becomes apparent only during stressful conditions. CONCLUSIONS: Strikingly, 60% of our PWS patients had CAI. The high percentage of CAI in PWS patients might explain the high rate of sudden death in these patients, particularly during infection-related stress. Based on our data, one should consider treatment with hydrocortisone during acute illness in PWS patients unless CAI has recently been ruled out with a metyrapone test
