The mitochondrial uncoupling as a promising pharmacological target against cancer
Context: Mitochondria represent a key intracellular signalling hub that is emerging as important determinants of numerous aspects of cancer development and progression. In this sense, the organelle constitutes a promising target for the development of novel anticancer agents. Despite the negative history, mitochondrial uncoupling has recently proposed as a pharmacological target against cancer, but little is known about the mechanisms involved.
Aims: To demonstrate the role of mitochondria in tumor formation and to describe how targeting of mitochondria uncoupling can be beneficial in the therapy of these diseases, which affect a large human population.
Methods: The data for this systematic review were collected from three popular databases including PubMed, Google Scholar, and Scopus and included in the search terms the words “mitochondrial uncoupling” and “cancer”. The influence of mitochondrial uncoupling on cells physiology that could lead to cancer cells death, invasion and metastasis was critically appraised from relevant researches. The anticancer effects of small molecules mitochondrial uncouplers and their anticancer mechanisms were also discussed. The present dataset finally included 201 published articles.
Results: It was found that the mitochondrial uncoupling-mediated responses possibly involved in the anti-cancer/anti-metastatic effects include Ca2+ homeostasis and bioenergetics disruption, mitochondrial membrane potential dissipation, reactive oxygen species generation, mitochondrial dynamics alteration, and gene expression modulation.
Conclusions: Overall, this critical review suggests that mitochondrial uncoupling could be an interesting pharmacological target to be considered in the design and synthesis of novel anti-cancer compounds with optimal physic-chemical and biopharmaceutical properties and improved safety profiles
Mangifera indica L. extract tablets supplementation in patients with knee osteoarthritis pain. A controlled pilot study.
Context: Several experimental results and clinical reports using Mangifera indica L extract (MSBE) suggest its potential utility in osteoarthritis (OA) mixed pain. Aims: To examine the possible therapeutic effects and safety of supplementation on osteoarthritis (OA) pain. Methods: Fifty patients with painful knee OA who had undergone a year of conventional treatment that included paracetamol and non-pharmacological therapies were randomly allocated to the experimental group (n = 21), which received a daily dose of 900 mg of extract supplementation or preceding usual treatment and placebo in the same form (n = 17) for a period of 120 days. The primary measure outcome was the change in the average daily pain diary score (ADPS) using the Likert scale. Also, a multidimensional measure of pain, stiffness and functional disability on The Western Ontario and Mc Master Universities (WOMAC) index for knee OA and ultrasonographic chronic signs of synovitis such as effusion and synovial thickness were evaluated. Results: Change from baseline in ADPS of the MSBE supplemented group showed a significant reduction after two weeks that lasted for 120 days with respect to the placebo group. Significant improvements in pain and functional disability WOMAC sub-scores, number of joints with synovial thickness and effusion after MSBE supplementation vs. placebo were observed. Non-adverse effects were reported in the experimental group. Conclusions: These results suggest that MSBE supplementation has a beneficial effect on OA pain and disability
Identifying the core concepts of pharmacology education : a global initiative
Background and Purpose: In recent decades, a focus on the most critical and fundamental concepts has proven highly advantageous to students and educators in many science disciplines. Pharmacology, unlike microbiology, biochemistry or physiology, lacks a consensus list of such core concepts . Experimental approach: We sought to develop a research-based, globally relevant list of core concepts that all students completing a foundational pharmacology course should master. This two-part project consisted of exploratory and refinement phases. The exploratory phase involved empirical data mining of the introductory sections of five key textbooks, in parallel with an online survey of over 200 pharmacology educators from 17 countries across six continents. The refinement phase involved three Delphi rounds involving 24 experts from 15 countries across six continents. Key Results: The exploratory phase resulted in a consolidated list of 74 candidate core concepts. In the refinement phase, the expert group produced a consensus list of 25 core concepts of pharmacology. Conclusion and Implications: This list will allow pharmacology educators everywhere to focus their efforts on the conceptual knowledge perceived to matter most by experts within the discipline. Next steps for this project include defining and unpacking each core concept and developing resources to help pharmacology educators globally teach and assess these concepts within their educational contexts
Diagnóstico rápido para empresas del sector de la construcción que permite posteriormente implementar un sistema de gestión ambiental (SGA)
La actividad constructora en los últimos dos años se encuentra orientada en su mayor proporción a la vivienda (76%) de acuerdo al número de metros cuadrados licenciados. Esta actividad constructora impacta el medio ambiente y es de capital importancia en la calidad de vida de la sociedad.
Se evaluaron cinco empresas en tres ciudades, las cuales contaban en promedio con 18 empleados directos.Se encontró que ninguna de ellas tenía un Sistema de Gestión Ambiental y mucho menos programas orientados a controlar y prevenir el impacto medio ambiental de sus procesos. Igualmente presentaban desconocimiento de la legislación y de la importancia de diseñar con el objetivo de preservar el medio ambiente, con el fin que los proyectos de construcción de vivienda podrían ejecutarse extendiendo y cerrando los ciclos de vida de los materiales y del producto terminado, con lo cual se lograría minimizar y mitigar los impactos que genera esta actividad económica.
