    AI For Sheep Using Frozen-thawed Semen.

    End of Project ReportInternational experience has been that cervical insemination of sheep with frozen-thawed semen usually yields unacceptably low pregnancy rates (10 to 30%). An exceptional case has been Norway where non-return rates in on-farm usage are around 60%. The objective of the work described in this report was to develop an AI procedure for Irish conditions, based initially on Norwegian protocols, using semen from individual rams. Such a procedure would greatly facilitate and enhance genetic improvement programmes for sheep. The work undertaken had two separate aspects:- (i) studies on semen, including processing and freezing methods, laboratory evaluation of semen quality post thawing and the relationship of in vitro evaluation to fertilisation rate in vivo (ii) studies on pregnancy rate following AI in relation to issues such as ram breed effects, effects of synchronisation, operator differences and the role of ewe breed inducing the timing of ovulation and various physical and physiological assessments of the cervix at AI. The main results in relation to semen studies were that, while a range of differential staining procedures could be used to objectively evaluate semen with respect to proportion of live speramatozoa and the integrity of sperm cells after thawing, these results were not useful as indicators of fertilisation capacity in vivo. The in vitro fertilisation (IVF) of sheep oocytes recovered from abattoir material gave promising results as a method for evaluating the fertilisation capacity of frozen-thawed semen. The results differ significantly from previous values deduced from angular correlation measurements which are in serious conflict with all sophisticated theoretical approaches. Our results support the trend of theoretical predictions for P2 and confirm that its value is negative at electron scattering angles above 100±, as predicted by theory

    The H alpha Galaxy Survey VI. Star-forming companions of nearby field galaxies

    We present a search for star-forming satellite galaxies that are close enough to their parent galaxies to be considered analogues of the Magellanic Clouds. Our search technique relied on the detection of the satellites in continuum-subtracted narrow-band H alpha imaging of the central galaxies, which removes most of the background and foreground line-of-sight companions, thus giving a high probability that we are detecting true satellites. The search was performed for 119 central galaxies at distances between 20 and 40 Mpc, although spatial incompleteness means that we have effectively searched 53 full satellite-containing volumes. We find only 9 probable star-forming satellites, around 9 different central galaxies, and 2 possible satellites. After incompleteness correction, this is equivalent to 0.17/0.21 satellites per central galaxy. The Small Magellanic Cloud is just below the median values of both star formation rate and R-band luminosity of the 9 probable satellites. The Large Magellanic Cloud, however, has a higher R-band luminosity than any of the 9 and is only exceeded in star formation rate by the one satellite that appears to be undergoing a tidally-induced starburst. Thus the Milky Way appears to be quite unusual, both in having two star-forming satellite galaxies and in the high luminosity of the Large Magellanic Cloud.Comment: 13 pages, accepted for publication in A&

    Safety Recommendations for Evaluation and Surgery of the Head and Neck During the COVID-19 Pandemic

    Importance The rapidly expanding novel coronavirus disease 2019 (COVID-19) pandemic, caused by severe acute respiratory syndrome coronavirus 2, has challenged the medical community to an unprecedented degree. Physicians and health care workers are at added risk of exposure and infection during the course of patient care. Because of the rapid spread of this disease through respiratory droplets, health care workers who come in close contact with the upper aerodigestive tract during diagnostic and therapeutic procedures, such as otolaryngologists–head and neck surgeons, are particularly at risk. A set of safety recommendations was created based on a review of the literature and communications with physicians with firsthand knowledge of safety procedures during the COVID-19 pandemic. Observations A high number of health care workers were infected during the first phase of the pandemic in the city of Wuhan, China. Subsequently, by adopting strict safety precautions, other regions were able to achieve high levels of safety for health care workers without jeopardizing the care of patients. The most common procedures related to the examination and treatment of upper aerodigestive tract diseases were reviewed. Each category was reviewed based on the potential risk imposed to health care workers. Specific recommendations were made based on the literature, when available, or consensus best practices. Specific safety recommendations were made for performing tracheostomy in patients with COVID-19. Conclusions and Relevance Preserving a highly skilled health care workforce is a top priority for any community and health care system. Based on the experience of health care systems in Asia and Europe, by following strict safety guidelines, the risk of exposure and infection of health care workers could be greatly reduced while providing high levels of care. The provided recommendations, which may evolve over time, could be used as broad guidance for all health care workers who are involved in the care of patients with COVID-19

    The recent assertion of [1.] that Fe-EDTA does not catalyze superoxide dismutation is disputed. By directly observing superoxide generated during pulse radiolysis, we have confirmed the results of a previous study ( [2.]) which concluded that Fe-EDTA catalyzed superoxide dismutation.     SummaryThe iron containing superoxide dismutase from E. coli B is shown to catalyze superoxide dismutation by a mechanism which exhibits saturation kinetics. This behavior is quite different from that observed previously with bovine Zn/Cu- and iron-containing superoxide dismutase from P. leiognathi. Two parameters of catalysis were measured in the pH range 7.2 to 10.4: kcat was found to be independent of pH and Km varied with the function Km = Km(low pH) [1 + exp(pH -- 8.8)].     Image-based view-angle independent cardiorespiratory motion gating and coronary sinus catheter tracking for x-ray-guided cardiac electrophysiology procedures

    Determination of the cardiorespiratory phase of the heart has numerous applications during cardiac imaging. In this article we propose a novel view-angle independent near-real time cardiorespiratory motion gating and coronary sinus (CS) catheter tracking technique for x-ray fluoroscopy images that are used to guide cardiac electrophysiology procedures. The method is based on learning CS catheter motion using principal component analysis and then applying the derived motion model to unseen images taken at arbitrary projections, using the epipolar constraint. This method is also able to track the CS catheter throughout the x-ray images in any arbitrary subsequent view. We also demonstrate the clinical application of our model on rotational angiography sequences. We validated our technique in normal and very low dose phantom and clinical datasets. For the normal dose clinical images we established average systole, end-expiration and end-inspiration gating success rates of 100%, 85.7%, and 92.3%, respectively. For very low dose applications, the technique was able to track the CS catheter with median errors not exceeding 1 mm for all tracked electrodes. Average gating success rates of 80.3%, 71.4%, and 69.2% were established for the application of the technique on clinical datasets, even with a dose reduction of more than 10 times. In rotational sequences at normal dose, CS tracking median errors were within 1.2 mm for all electrodes, and the gating success rate was 100%, for view angles from RAO 90° to LAO 90°. This view-angle independent technique can extract clinically useful cardiorespiratory motion information using x-ray doses significantly lower than those currently used in clinical practice

    Metformin reduces airway glucose permeability and hyperglycaemia-induced Staphylococcus aureus load independently of effects on blood glucose

    Background Diabetes is a risk factor for respiratory infection, and hyperglycaemia is associated with increased glucose in airway surface liquid and risk of Staphylococcus aureus infection. Objectives To investigate whether elevation of basolateral/blood glucose concentration promotes airway Staphylococcus aureus growth and whether pretreatment with the antidiabetic drug metformin affects this relationship. Methods Human airway epithelial cells grown at air–liquid interface (±18 h pre-treatment, 30 μM–1 mM metformin) were inoculated with 5×105 colony-forming units (CFU)/cm2 S aureus 8325-4 or JE2 or Pseudomonas aeruginosa PA01 on the apical surface and incubated for 7 h. Wild-type C57BL/6 or db/db (leptin receptor-deficient) mice, 6–10 weeks old, were treated with intraperitoneal phosphate-buffered saline or 40 mg/kg metformin for 2 days before intranasal inoculation with 1×107 CFU S aureus. Mice were culled 24 h after infection and bronchoalveolar lavage fluid collected. Results Apical S aureus growth increased with basolateral glucose concentration in an in vitro airway epithelia–bacteria co-culture model. S aureus reduced transepithelial electrical resistance (RT) and increased paracellular glucose flux. Metformin inhibited the glucose-induced growth of S aureus, increased RT and decreased glucose flux. Diabetic (db/db) mice infected with S aureus exhibited a higher bacterial load in their airways than control mice after 2 days and metformin treatment reversed this effect. Metformin did not decrease blood glucose but reduced paracellular flux across ex vivo murine tracheas. Conclusions Hyperglycaemia promotes respiratory S aureus infection, and metformin modifies glucose flux across the airway epithelium to limit hyperglycaemia-induced bacterial growth. Metformin might, therefore, be of additional benefit in the prevention and treatment of respiratory infection
